Several laboratories have shown that a primary function of IQGAP proteins is to modulate cytoskeletal architecture. Yeast IQGAP homologues have a role in the recruitment of actin filaments, are components of the spindle pole body and are required for actomyosin ring assembly and cytokinesis (Eng et al., 1998; Epp & Chant, 1997; Lippincott & Li, 1998; reviewed in Machesky, 1998). Similarly, a homologue of IQGAP1 in Hydra has a role in tentacle formation (Venturelli et al., 2000). Mammalian IQGAP1 enhances actin polymerization in vitro (Bashour et al., 1997; Erickson et al., 1997; Fukata et al., 1997) and colocalizes with actin in lamellipodia (Hart et al., 1996). This conservation strongly suggests that cytoskeletal regulation was a primordial function of IQGAP1. Environmental cues such as heat shock, UV irradiation, hypoxia, and oxidative stress initiate many eukaryotic cellular responses, including protective responses to ensure their survival. One of the most rapid of these responses is to prevent mRNA translation to into protein to allow cells to adapt to stress. This stalled mRNA is sequestered to specific cytosolic compartments known as stress granule. Stress granules represent a complex assemblage of translational initiation factors, proteins involved in translational control, the microtubule array, and chaperone proteins such as G3BP-1 (Ras-GTPase-activating protein Src homology 3 domain-binding protein-1). During migration, exocytic vesicles fuse with the cell's leading edge to enable directional growth. This polarized secretion is thought to be promoted by the exocyst complex - which tethers vesicles to the plasma membrane - and by septin proteins that associate with the exocyst. However, little is known about how the process is regulated. Now, Mahasin Osman and colleagues (p. 391) propose that the actin-modulating protein IQGAP1 activates polarised secretion. Using co-immunoprecipitation, the authors show that the N-terminal region of IQGAP1 binds the exocyst-septin complex directly in pancreatic β-cells. This interaction is disrupted by the expression of the GTPase CDC42, which is known to bind the C-terminal region of IQGAP1. Moreover, CDC42 also inhibits IQGAP1-mediated insulin secretion. We for the first time report common genomic IQGAP1 copy gain, associated with overexpression of IQGAP1 protein, in thyroid cancer, with a preferential association with invasive types of thyroid cancer, such as, ATC, TCPTC, and FTC. The IQGAP1 gene is located at 15q26, a chromosome area that is frequently amplified in many human malignancies. IQGAP1 itself is also amplified in other cancers (4) and was associated with IQGAP1 protein overexpression in gastric cancer cells (11). The IQGAP1 copy gain was a specific event in thyroid cancer as IQGAP3 did not show copy gain. Moreover, we saw only rare copy gain of TMEM84, a gene that is located at 15q23, about 20 Mb from IQGAP1 in the same chromosome, in striking contrast with the common copy gain of the IQGAP1 gene.
