We evaluated the morphology of mixed species biofilms of S. epidermidis and C. albicans, in vitro. We observed enhancement of biofilms in a mixed species environment. In a mouse subcutaneous catheter model of biofilm infection, we noted increased catheter infection and systemic dissemination of S. epidermidis in a mixed species environment. To further explore the reasons for increased pathogenicity of S. epidermidis in mixed species biofilm infections with C. albicans, we evaluated the transcriptome of S. epidermidis in a mixed species environment and found that the repressors of autolysis, lrgA and lrgB were highly down regulated. Staphylococcus hominis is a coagulase-negative member of the bacterial genus Staphylococcus, consisting of Gram-positive, spherical cells in clusters. It occurs very commonly as a harmless commensal on human and animal skin and is known for producing thioalcohol compounds that contribute to body odour. Like many other coagulase-negative staphylococci, S. hominis may occasionally cause infection in patients whose immune systems are compromised, for example by chemotherapy or predisposing illness. Colonies of S. hominis are small, usually 1-2 mm in diameter after 24 hours' incubation at 35 °C, and white or tan in colour. In this study, the in vitro bacterial growth inhibition, antioxidant activity and the content in bioactive components of Plectranthus barbatus, P. hadiensis var. tomentosus, P. madagascarensis, P. neochilus and P. verticillatus aqueous extracts were investigated and compared by three extraction methods (infusion, decoction and microwave extractions). The microwave extract of P. madagascariensis showed the higher antimicrobial activity against the Staphylococcus epidermidis strain with a minimum inhibitory concentration of 40 µg/mL. This extract also showed no toxicity in a general toxicity assay and no considerable cytotoxicity against a human keratinocyte cell line. The antioxidant activity of the extracts was assessed using 2,2-diphenyl-1-picrylhydrazyl (DPPH method), and all showed antioxidant activity. The microbiome can promote or disrupt human health by influencing both adaptive and innate immune functions. We tested whether bacteria that normally reside on human skin participate in host defense by killing Staphylococcus aureus, a pathogen commonly found in patients with atopic dermatitis (AD) and an important factor that exacerbates this disease. High-throughput screening for antimicrobial activity against S. aureus was performed on isolates of coagulase-negative Staphylococcus (CoNS) collected from the skin of healthy and AD subjects. CoNS strains with antimicrobial activity were common on the normal population but rare on AD subjects. A low frequency of strains with antimicrobial activity correlated with colonization by S. aureus. In 42333 anti-VEGF intravitreal injections performed, an acute endophthalmitis occurred in ten patients (0.02%), one after Bevacizumab, one after Aflibercept and eight after Ranibizumab. The mean post operative onset was 3, 4 days. Vitreous tap revealed 5 cases of Staphylococcus Epidermidis, one case of Staphylococcus Warneri, one case of Staphylococcus Haemolyticus and in 3 cases was negative. One patient was lost to follow-up during the first month after injection. The remaining 9 patients had a mean follow-up of 38 months (min. 18 - max. 60). The mean VA was 0.49±0.22 before endophthalmitis, 0.28±0.22 after one month and 0.33±0.26 at the last follow-up. Seven patients recovered at least the pre-endophthalmitis VA score during follow-up (5 patients between 1 and 3 months, one patient in 16 months and one in 36 months). Usnic acid, a potent antimicrobial and anticancer agent, poorly soluble in water, was complexed to novel antimicrobial polyacrylamides by establishment of strong acidic-base interactions. Thermal and spectroscopic analysis evidenced a molecular dispersion of the drug in the polymers and a complete drug/polymer miscibility for all the tested compositions. The polymer/drug complexes promptly dissolved in water and possessed a greater antimicrobial activity against Staphylococcus epidermidis than both the free drug and the polymer alone. The best results were obtained with the complex based on the lowest molecular weight polymer and containing a low drug content. Such a complex showed a larger inhibition zone of bacterial growth and a lower minimum inhibitory concentration (MIC) with respect to usnic acid alone. This improved killing effect is presumably due to the reduced size of the complexes that allows an efficient cellular uptake of the antimicrobial complexes. The chemical structures of the isolated compounds were elucidated on the basis of one- and two-dimensional NMR spectroscopy supported by UV and HRESIMS data. The compounds exhibited inhibitory activities against Staphylococcus epidermidis, Xanthomonas campestris and Propionibacterium acnes. The patient considered follow-up unnecessary, and he was not concerned when the device began to show through the skin. Only when it fell out, 2 weeks before admission, did he notify his physician. No systemic reaction occurred, and the pacemaker functioned normally. The patient prevented the device from catching in his clothing by holding it against his chest wall with a gauze flat and adhesive tape. Explantation and replacement of a new unit under the pectoral major muscle on the opposite side was accomplished without incident. Cultures of the wound were positive for Staphylococcus epidermidis. The objective of this thesis was to investigate the feasibility of counting and sizing of bacteria with the Coulter Counter and to evaluate orifices which were prepared in this laboratory.. Size distributions of Pasteurella multocida and Proteus vulgaris were obtained with the electronic particle counter. These distributions appeared to follow the normal distribution when relative per cent was plotted against diameter, The two modes of the distributions appeared far enough apart to warrant future attempts to separate quantitatively P. vulgaris and P. multocida in mixed suspensions.
