*  Microbiology Online Focus On: Dark Field Microscopy (Part IV) | Microbiology Online
The condenser lens used by the dark field microscopy is called cardioid condenser and there are two types: dry and wet. The wet one requires a liquid medium between the objective and the slide, while the dry one does not. The wet one provides clearer images, in opposite of the dry one. For this reason, the most frequently used type of condenser is the wet one and the liquid used between the slide and the objective is the immersion oil ...

*  Three-dimensional animation with conventional light microscopy | (1997) | Tympel | Publications | Spie
A small depth of focus is very often a problem in the conventional high resolution light microscopy, because the objects are higher than the depth of focus. The connections of conventional light microscopes, video techniques and computers open new fields of applications for microscopy. It is possible to generate an image containing focused areas from a series of images of different focus- or z-positions. In this way it is possible to extend the depth of focus without a physical limitation of the numerical aperture (NA) of the objective lens. The connection of microscope, video and PC opens also the field of high resolution low light color applications, like fluorescence in situ hybridization (FISH). Image processing technology enables the enhancement of contrast for objects with a very low contrast. And such a system opens also the way to view objects with small motions, like processes of growing of biological objects. It is also the elimination of the limits in the ...

*  Viruses | Free Full-Text | Superresolution Imaging of Human Cytomegalovirus vMIA Localization in Sub-Mitochondrial Compartments
The human cytomegalovirus (HCMV) viral mitochondria-localized inhibitor of apoptosis (vMIA) protein, traffics to mitochondria-associated membranes (MAM), where the endoplasmic reticulum (ER) contacts the outer mitochondrial membrane (OMM). vMIA association with the MAM has not been visualized by imaging. Here, we have visualized this by using a combination of confocal and superresolution imaging. Deconvolution of confocal microscopy images shows vMIA localizes away from mitochondrial matrix at the Mitochondria-ER interface. By gated stimulated emission depletion (GSTED) imaging, we show that along this interface vMIA is distributed in clusters. Through multicolor, multifocal structured illumination microscopy (MSIM), we find vMIA clusters localize away from MitoTracker Red, indicating its OMM localization. GSTED and MSIM imaging show vMIA exists in clusters of ~100-150 nm, which is consistent with the cluster size determined by Photoactivated Localization ...

*  A New Tool for CSF-Based Parkinson Disease Diagnostics: Super-Resolution Microscopy Investigation of Alpha-Synuclein Aggregates...
The field of optical microscopy has been revolutionized in the last few years, starting with the invention of so-called super-resolution microscopes in the early 2000 s. We have long been involved in this process, and we created optimal ways to prepare and analyze samples for this microscopy technology. We will use here super-resolution microscopy to detect and image synuclein aggregates in the cerebro-spinal fluid from PD patients and healthy control persons. We will compare the amount, the size and the shape of the aggregates between PD patients and controls, and we will thus verify which of these parameters can be used reliably as a diagnostic.. Relevance to Diagnosis/Treatment of Parkinson s Disease ...

*  Biochips for Live Cell Microscopy
Overview. Microfluidic biochips are an attractive tool for live cell microscopy as their dimensions can mimic the dimensions of human capillaries thereby enabling researchers to simulate in vivo microenvironments. Biochips offer distinct advantages over traditional glass capillaries (known for optical aberrations) or perfusion chambers as cutting edge fabrication techniques result in in vitro models that allow high-resolution digital microscopy for brightfield, phase contrast, fluorescent imaging and confocal microscopy. The surface of the microcapillaries may be pretreated to support adhesion of proteins / ligands or culturing of different cells types; thereby resulting in a very flexible and adaptable research tool.. Assays:. ...

*  Quantification of DNA-associated proteins inside eukaryotic cells using single-molecule localization microscopy : Sussex...
Development of single-molecule localization microscopy techniques has allowed nanometre scale localization accuracy inside cells, permitting the resolution of ultra-fine cell structure and the elucidation of crucial molecular mechanisms. Application of these methodologies to understanding processes underlying DNA replication and repair has been limited to defined in vitro biochemical analysis and prokaryotic cells. In order to expand these techniques to eukaryotic systems, we have further developed a photo-activated localization microscopy-based method to directly visualize DNA-associated proteins in unfixed eukaryotic cells. We demonstrate that motion blurring of fluorescence due to protein diffusivity can be used to selectively image the DNA-bound population of proteins. We designed and tested a simple methodology and show that it can be used to detect changes in DNA binding of a replicative helicase subunit, Mcm4, and the replication sliding clamp, PCNA, between different ...

Optical microscopes are microscopes that typically use visible light and a system of lenses to magnify images of small samples. Industrial microscopes incorporate many complex designs that aim to improve resolution and sample contrast. Images from an optical microscope can be captured by normal light-sensitive cameras to generate a micrograph. Modern developments in CMOS and charge-coupled device (CCD) cameras allow the capture of digital images. Digital microscopes are available with a CCD camera to examine a sample, and the image is shown directly on a computer screen without the need for eye-pieces.
