*  Cytokeratin phosphorylation, cytokeratin filament severing and the solubilization of the maternal mRNA Vg1. | JCB
During meiotic maturation, the cortical cytokeratin filament system of the Xenopus oocyte disappears (Klymkowsky, M. W., and L. A. Maynell. 1989. Dev. Biol. 134:479). Here we demonstrate that this disappearance results from the severing of cytokeratin filaments into a heterogenous population of oligomers, with S- values ranging from 12S and greater. Cytokeratin filament severing correlates with the hyperphosphorylation of the type II cytokeratin of the oocyte. Both the severing of cytokeratin filaments and cytokeratin hyperphosphorylation are reversed by treatment with cycloheximide. These data suggest that fragmentation of cytokeratin filaments is controlled, at least in part, by the phosphorylation of the type II cytokeratin, and that the cytokeratin kinase activity responsible is biosynthetically labile. Cytokeratin filaments have been suggested to anchor the maternal mRNA Vg1 to the vegetal cortex of the oocyte (Pondel, M., and M. L. King. 1988. Proc. Natl. Acad. Sci. USA. 85:7216). By ...

*  Immunological and Immuno-Fluorescent Studies on Keratin of the Hair Follicle | Journal of Cell Science
Keratins from the differentiating cortical cells of the guinea-pig hair follicle have been studied by immunological and immunofluorescent procedures. Antisera were prepared to the unfractionated keratin proteins from the follicle and the specificity of these antisera examined by immunodiffusion and immunoelectrophoresis. The major follicle antigens were the 'low-sulphur' keratins, and the antibodies were specific for these and related αkeratins. Use of the precipitin reaction as a quantitative immunoassay for follicle keratins has been demonstrated. The fluorescent antibody technique has been applied to the hair follicle and the follicle antigens have been localized by this procedure in the cytoplasm of the developing cortical cells. By this technique keratin could not be detected in the proliferative cells of the bulb region of the follicle supporting the view that keratin synthesis begins only when the cells enter the upper bulb region of ...

*  Keratin 7 - Wikipedia
Keratin, type II cytoskeletal 7 also known as cytokeratin-7 (CK-7) or keratin-7 (K7) or sarcolectin (SCL) is a protein that in humans is encoded by the KRT7 gene. Keratin 7 is a type II keratin. It is specifically expressed in the simple epithelia lining the cavities of the internal organs and in the gland ducts and blood vessels. Keratin-7 is a member of the keratin gene family. The type II cytokeratins consist of basic or neutral proteins which are arranged in pairs of heterotypic keratin chains coexpressed during differentiation of simple and stratified epithelial tissues. This type II cytokeratin is specifically expressed in the simple epithelia lining the cavities of the internal organs and in the gland ducts and blood vessels. The genes encoding the type II cytokeratins are clustered in a region of chromosome 12q12-q13. Alternative splicing may result in several transcript variants; however, not all variants have been fully described. Keratin-7 is found in simple ...

*  Hitchhiker's Guide to the Galaxy on Keratin
Metabolism ceases, and the cells are almost completely filled by keratin. During the process of epithelial differentiation, cells become cornified as keratin protein is incorporated into longer keratin intermediate filaments. Eventually the nucleus and cytoplasmic organelles disappear, metabolism ceases and cells undergo a programmed death as they become fully keratinized. In many other cell types, such as cells of the dermis, keratin filaments and other intermediate filaments function as part of the cytoskeleton to mechanically stabilize the cell against physical stress. It does this through connections to desmosomes, cell-cell junctional plaques, and hemidesmosomes, cell-basement membrane adhesive structures.. Cells in the epidermis contain a structural matrix of keratin, which makes this outermost layer of the skin almost waterproof, and along with collagen and elastin, gives skin its strength. Rubbing and pressure cause thickening of the outer, cornified layer of the epidermis and form ...

*  Keratin 8 overexpression promotes mouse Mallory body formation | JCB
Intermediate filaments (IFs) are a large family of cytoplasmic and nuclear proteins that are involved in a broad range of diseases that reflect their cell and tissue-specific expression (Herrmann et al., 2003; Coulombe and Wong, 2004; Omary et al., 2004). The nuclear IFs include lamins, whereas examples of cytoplasmic IFs include desmin and neurofilaments that are preferentially expressed in muscle and neuronal cells, respectively. In epithelial cells, cytoplasmic IFs consist of types I and II keratins that are found as obligate noncovalent heteropolymers and are expressed in an epithelial cell-specific manner (Coulombe and Omary, 2002). Type I and II keratins stabilize each other via formation of a coil-coil α helix and are found in cells in a 1:1 type I/II stoichiometric ratio. In digestive organ epithelia, the major IFs are keratin polypeptides 8 and 18 (K8/18), with variable levels of K7, 19, and 20 depending on the tissue/cell type (Ku et al., 1999). Adult hepatocytes ...

*  "Genomic organization and molecular phylogenies of the beta (β) keratin" by Matthew J. Greenwold and Roger H. Sawyer
Background: The epidermal appendages of reptiles and birds are constructed of beta (β) keratins. The molecularphylogeny of these keratins is important to understanding the evolutionary origin of these appendages, especially feathers. Knowing that the crocodilian β-keratin genes are closely related to those of birds, the published genomes ofthe chicken and zebra finch provide an opportunity not only to compare the genomic organization of their β- keratins,but to study their molecular evolution in archosaurians. Results: The subfamilies (claw, feather, feather-like, and scale) of β-keratin genes are clustered in the same 5' to 3' orderon microchromosome 25 in chicken and zebra finch, although the number of claw and feather genes differs between the species. Molecular phylogenies show that the monophyletic scale genes are the basal group within birds and thatthe monophyletic avian claw genes form the basal group to all feather and feather-like genes. Both ...

Keratin is a type of protein that is found in our hair, skin, nails and teeth. Keratin is formed by keratinocytes or living cells that are found in our hair, skin and other parts of our body.. When the keratin in our hair gets damaged due to several chemical treatments like colouring and so on, our hair starts looking frizzy, dull and unmanageable. By applying keratin solution back on the hair, the hair shaft gets a protective layer and turns smooth once again.. Keratin helps in changing the structure of the hair from inside the shaft and locks the hair from outside thereby making your hair strong and healthy.. Keratin is difficult dissolve as it contains a content called as cysteine disulfide. This gives the ability of forming disulfide bridges to the keratin. The disulfide bridges create helix shape and this is extremely strong in nature as sulphur atoms bond with each other in the helix and create a fibrous matrix making the solution difficult to dissolve.. What does Keratin Treatment do ...

*  Increased expression of keratin 16 causes anomalies in cytoarchitecture and keratinization in transgenic mouse skin. | JCB
Injury to epidermis and other stratified epithelia triggers profound but transient changes in the pattern of keratin expression. In postmitotic cells located at the wound edge, a strong induction of K6, K16, and K17 synthesis occurs at the expense of the keratins produced under the normal situation. The functional significance of these alterations in keratin expression is not known. Here, we report that overexpression of a wild-type human K16 gene in a tissue-specific fashion in transgenic mice causes aberrant keratinization of the hair follicle outer root sheath and proximal epidermis, and it leads to hyperproliferation and increased thickness of the living layers (acanthosis), as well as cornified layers (hyperkeratosis). The pathogenesis of lesions in transgenic mouse skin begins with a reorganization of keratin filaments in postmitotic keratinocytes, and it progresses in a transgene level-dependent fashion to include disruption of keratinocyte cytoarchitecture and structural alterations ...

Thirteen meningiomas of varying light microscopic features were studied immunohistologically using a panel of monoclonal antibodies directed against epithelial, mesenchymal, and neural components. All 13 meningiomas showed expression of epithelial membrane antigen, vimentin, and S100 protein, as did normal meninges. Five of the 13 meningiomas also showed focal expression of cytokeratins, with double labelling showing expression of cytokeratins and vimentin by different cells. The cytokeratin expression was especially noticeable in cells surrounding the hyaline bodies of meningiomas. These results provide further evidence that meningiomas have features of both mesenchymal and epithelial tissues.. ...

*  Why I Choose not to do Keratin Treatments
As a stylist I try to refrain from jumping on the latest trends - especially if they involve a chemical process. I have learned that it is difficult to assess the pros and cons of a new process without factoring in the effects over time. I regret now that I may have jumped the gun on endorsing one particular process on this blog in the past - keratin treatments. I am rescinding that endorsement in light of new facts.. The problem with keratin treatments - no brands excluded - is that they only work to smooth the hair if they contain dimethicone, a non-water soluble silicone found in many hair care products. Keratin alone will not smooth the hair - I know this because I worked for a distributor of a keratin treatment which shall remain nameless… When complaints were made to OSHA regarding formaldehyde levels in keratin treatments the manufacturer tried playing with the formula and removing different elements like the dimethicone; the problem was, the keratin treatment no longer worked without ...
