*  Molecular basis for specific viral RNA recognition and 2'-O-ribose methylation by the dengue virus nonstructural protein 5...
Zhao Y., Soh T.S., Lim S.P., Chung K.Y., Swaminathan K., Vasudevan S.G., Shi P.Y., Lescar J., Luo D.. Dengue virus (DENV) causes several hundred million human infections and more than 20,000 deaths annually. Neither an efficacious vaccine conferring immunity against all four circulating serotypes nor specific drugs are currently available to treat this emerging global disease. Capping of the DENV RNA genome is an essential structural modification that protects the RNA from degradation by 5' exoribonucleases, ensures efficient expression of viral proteins, and allows escape from the host innate immune response. The large flavivirus nonstructural protein 5 (NS5) (105 kDa) has RNA methyltransferase activities at its N-terminal region, which is responsible for capping the virus RNA genome. The methyl transfer reactions are thought to occur sequentially using the strictly conserved flavivirus 5' RNA sequence as substrate (GpppAG-RNA), leading to ...

*  Cleavage of the IPS-1/Cardif/MAVS/VISA does not inhibit T cell-mediated elimination of hepatitis C virus non-structural 3/4A...
The underlying mechanisms that help HCV to establish and maintain a chronic infection in humans are still poorly understood.4 11 40 Several mechanisms that seem to promote viral persistence have been identified in vitro and in animal studies.7 41-44 Many HCV proteins seem to interfere with host cell signalling, where the NS3/4A complex is the best characterised, so far.3 19 20 The NS3/4A-mediated cleavage of IPS-1 has been documented in human and tamarin cells.34 35 However, there are significant difficulties in studying these events repeatedly in infected human livers. It is therefore of the utmost importance to determine if there are other simpler model systems that may in the same, or in a different way, represent the in vivo situation. Since a number of transgenic mouse lineages exist where NS3/4A should be present,16 17 45 we studied the effects of NS3/4A in murine cells.. In line with previous reports3 19 23 we found that over-expression of NS3/4A in human cells reduced ...

*  Hepatitis C virus non-structural 3/4A protein interferes with intrahepatic interferon-γ production | Gut
Results The activation of hepatic signal transducer and activator of transcription 1 and 2 was impaired in NS3/4A-Tg mice after treatment with LPS/d-galN or TNFα/d-galN. This was paralleled by a reduction in hepatic interferon-γ (IFNγ). Reconstitution of IFNγ reverted the resistance to LPS/TNFα in NS3/4A-Tg mice. Subsequently, blocking IFNγ in vivo rendered wild-type mice resistant against treatment with LPS/TNFα. A new Tg mouse expressing an inactivated NS3/4A protease had the same phenotype as wild-type mice with respect to hepatic IFNγ levels and sensitivity to LPS/d-galN. Finally, the chemokine profile was altered in the NS3/4A-Tg mice towards an anti-inflammatory state, which helps to explain the altered immune cell subsets and reduction in hepatic IFNγ production. ...

American Association of Avian Pathologists Annual Meeting, July 11-15, 2009, Seattle, Washington. CD-ROM, paper No. 7586. Interpretive Summary: Technical Abstract: The NS1 protein of influenza A virus plays an important role in blocking the induction of type I interferon and other regulatory functions in infected cells. However, differences in length of the NS1 protein has been observed in highly pathogenic H5N1, H5N2, and H7N1 subtype avian influenza viruses. To evaluate the influence of truncated NS1 gene on H5N1 virus pathogenicity in chickens, using reverse genetics, we generated polybasic H5 hemagglutinin (HA) containing A/turkey/Ireland/83 and A/chicken/Indonesia/7/03 viruses with full length and truncated NS1 genes. The viruses were compared by intravenous pathotyping and mucosal ...

*  Single-Base Pair Unwinding and Asynchronous RNA Release by the Hepatitis C Virus NS3 Helicase | Science
Nonhexameric helicases use adenosine triphosphate (ATP) to unzip base pairs in double-stranded nucleic acids (dsNAs). Studies have suggested that these helicases unzip dsNAs in single-base pair increments, consuming one ATP molecule per base pair, but direct evidence for this mechanism is lacking. We used optical tweezers to follow the unwinding of double-stranded RNA by the hepatitis C virus NS3 helicase. Single-base pair steps by NS3 were observed, along with nascent nucleotide release that was asynchronous with base pair opening. Asynchronous release of nascent nucleotides rationalizes various observations of its dsNA unwinding and may be used to coordinate the translocation speed of NS3 along the RNA during viral replication.. ...

DescriptionInfluenza A is responsible for tens of thousands of annual fatalities in the United States. The conventional medical strategy relies on prevention of infection using vaccines where are mildly effective at best and require yearly updates to reflect the expected prevalent strains, and anti-viral drugs for which there is significant clinical resistance. With this in mind, the non-structural protein 1 of influenza A (NS1A) is an appropriate target with high conservation and a multitude of anti-immune effects. It features an array of high surface area interactions which may be targetable using peptides. We are developing molecular design tools to reliably create short ...
