*  IJMS | Free Full-Text | Hypoxia Is a Critical Parameter for Chondrogenic Differentiation of Human Umbilical Cord Blood...
Umbilical cord blood (UCB) is an attractive alternative to bone marrow for isolation of mesenchymal stem cells (MSCs) to treat articular cartilage defects. Here, we set out to determine the growth factors (bone morphogenetic protein 2 (BMP-2) and transforming growth factor-β (TGF-β1)) and oxygen tension effects during chondrogenesis of human UCB-MSCs for cartilage engineering. Chondrogenic differentiation was induced using 3D cultures in type I/III collagen sponges with chondrogenic factors in normoxia (21% O2) or hypoxia (|5% O2) for 7, 14 and 21 days. Our results show that UCB-MSCs can be committed to chondrogenesis in the presence of BMP-2+TGF-β1. Normoxia induced the highest levels of chondrocyte-specific markers. However, hypoxia exerted more benefit by decreasing collagen X and matrix metalloproteinase-13 (MMP13) expression, two chondrocyte hypertrophy markers. However, a better chondrogenesis was obtained by switching ...
*  A Multi-Center, Phase III, Randomized Trial of Reduced Intensity (RIC) Conditioning and Transplantation of Double Unrelated...
Reduced intensity conditioning (RIC) blood or marrow transplantation (BMT) has allowed older and less clinically fit patients to receive potentially curative treatment with allogeneic HCT for high risk or advanced hematological malignancies. Patients lacking an HLA-matched sibling may receive a graft from a suitably HLA-matched unrelated donor. However, up to a third of patients will not have an HLA-matched sibling or a suitably matched adult unrelated donor (i.e., no more than a mismatch at a single locus). Even when a suitably matched unrelated donor is identified, data from the National Marrow Donor Program (NMDP) indicate that a median of four months is required to complete searches that result in transplantation; thus, some number of patients succumb to their disease while awaiting identification and evaluation of a suitably matched adult unrelated donor.. Single or dual center studies have shown that partially HLA-mismatched related bone marrow ...
*  Polymorphism in TGFB1 is associated with worse non-relapse mortality and overall survival after stem cell transplantation with...
Transforming growth factor β-1, encoded by the TGFB1 gene, is a cytokine that plays a central role in many physiologic and pathogenic processes. We have sequenced TGFB1 regulatory region and assigned allelic genotypes in a large cohort of hematopoietic stem cell transplantation patients and donors. In this study we analyzed 522 unrelated donor-patient pairs and examined the combined effect of all the common polymorphisms in this genomic region. In univariate analysis, we found that patients carrying a specific allele, p001, showed significantly reduced overall survival (5-year overall survival 30.7% for p001/p001 patients vs 41.6% others; p=0.032) and increased non-relapse mortality (1-year non-relapse mortality: 39.0% vs 25.4%; p=0.039) after transplantation. In multivariate analysis, the presence of a p001/p001 genotype in patients was confirmed as an independent factor for reduced overall survival (hazard ratio=1.53 [1.04-2.24]; p=0.031), ...

*  USF Health News Archives » USF and Saneron find additional benefits of cord blood cells in mice modeling ALS
We hypothesized that the effect of the multiple MNC hUCB cell administrations decreased neuroinflammation in the spinal cord, even when administered into symptomatic mice, resulting in neuroprotection that promoted motor neuron survival," said co-author Maria C. O. Rodrigues, MD, PhD, associated with both USF and the University of Sao Paulo.. Additionally, the researchers found that although the number of grafted cells identified in the spinal cord was low, the treatment was effective, suggesting that various factors secreted by the cells accounted for the therapeutic impact.. Functional improvement in the test mice was determined through a number of tests.. "Because functional improvement was detected in the mice shortly after MNC hUCB administration, a neuroprotective function of the factors secreted by the administered cells is likely, along with some ...

*  Market Design: Organ preservation could bring big changes to transplantation
Abstract: The ability to replace organs and tissues on demand could save or improve millions of lives each year globally and create public health benefits on par with curing cancer. Unmet needs for organ and tissue preservation place enormous logistical limitations on transplantation, regenerative medicine, drug discovery, and a variety of rapidly advancing areas spanning biomedicine. A growing coalition of researchers, clinicians, advocacy organizations, academic institutions, and other stakeholders has assembled to address the unmet need for preservation advances, outlining remaining challenges and identifying areas of underinvestment and untapped opportunities. Meanwhile, recent discoveries provide proofs of principle for breakthroughs in a family of research areas surrounding biopreservation. These developments indicate that a new paradigm, integrating multiple existing preservation approaches and new technologies that have flourished in the past 10 years, could transform preservation ...

Why not save umbilical cord blood and tissue upon birth? Stem cells are invaluable; they are smart, powerful, and specific genetically for the child. Stored cord blood can later regenerate healthy blood and contribute to immune systems. Stored cord tissue can create structure and connective tissue in the body. It's no secret the immense potential value this offers your child, should health care issues arise.. Areas of current use of blood stem cells include immune and metabolic disorders, cancers, and blood disorders. There are trials underway for use in autism and cerebral palsy treatment, hearing loss recovery, and stroke.. Areas of potential use of umbilical cord tissue stem cells ...

Now for something off-topic, but it's for a good cause. I hope that piques your interest. My dad, computer whiz and father figure extraordinaire, is the IT manager for a relatively new organization called The Cleveland Cord Blood Center. Why am I bringing this up? Well, first let me tell you a little bit about what the CCBC is all about (you can also click on the link provided to sleuth for yourself). It is Ohio's first and only public cord blood bank. How does it work? A full-term baby is born, the umbilical cord is cut, the blood (full of stem cells!) is extracted and sent to CCBC's facility where it can then be distributed world-wide for transplants or for research. Why are stem cells important? They have the ability to renew themselves and then differentiate into specialized cell types. ...
