Cooke and colleagues set out to determine if serum PSA is more useful in discriminating various forms of prostatic disease than serum ACP. Among 349 patients who subsequently had prostatic biopsy or surgery, PSA did slightly better than ACP. Improvement in accuracy, however, is accompanied by a higher cost (U.S. $45.35 vs U.S. $21.65 at our hospital). These findings raise a number of questions, the most important of which is whether either test is suitable for screening of the general population. Using reported sensitivity and specificity figures and the prevalence of prostate cancer in the general United States population, a PSA level of ≥ 23 ng/mL yields a positive predictive value of 1.7%. In other words, for every 1000 men with a PSA level ≥ 23 ng/mL, only 17 will subsequently be found to have prostate cancer. We investigated the formation of tartrate-resistant acid phosphatase positive (TRAP+) colonies and multinucleated cells (MNCs) in rat and mouse marrow cultures alone and cocultured with the chondrogenic cell line, RCJ3.1C5.18 on different substrata. In mouse marrow cultured in 35 mm dishes, few TRAP+ MNCs developed, while in rat marrow cultures, many TRAP+ MNCs, which possessed calcitonin receptors and resorbed bone, developed. In both rat and mouse cultures, TRAP+ MNC first appeared at Day 4 and grew in number up to Day 8. When both marrows were cocultured with RC3.1C5.18 cells, TRAP+ colony numbers increased relative to marrow alone. In marrow populations or C5.18 cocultures, TRAP+ colony and TRAP+ MNC formation differed in accordance to the substrates used, i.e., 35 mm Falcon tissue culture plastic dishes or insert baskets. We conducted a transcriptomic screen of osteosarcoma (OS) biopsies and found that expression of osteoclast-specific tartrate-resistant acid phosphatase 5 (ACP5/TRAP) is significantly downregulated in OS compared with nonmalignant bone (P , 0.0001). Moreover, lesions from OS patients with pulmonary metastases had 2-fold less ACP5/TRAP expression (P , 0.018) than lesions from patients without metastases. In addition, we found a direct correlation (P = 0.0166) between ACP5/TRAP expression and time to metastasis. Therefore, we examined whether metastasis-competent (MC) OS cells could induce loss of ACP5+ osteoclasts and contribute to metastasis. We found that MC OS cell lines can inhibit osteoclastogenesis in vitro and in vivo. In addition, osteoclasts can inhibit the migration of MC OS cells in vitro. Histochemical observations were carried out on various hydrolytic enzymes in the spontaneous mammary carcinoma of C3H-strain mice.. Alkaline phosphatase activity was observed in the luminal border of the tubular structure and in the periluminal surface, which showed a cystic dilatation in ducts and papillae. Capillaries in the stroma showed a high enzymatic activity. Acid phosphatase activity was moderate in the inner layer of large ductlike structure of neoplasms and rather slight in other parts. Esterase activity was variable in neoplastic epithelia. Activities for β-glucuronidase and aminopeptidase were low in tumor cells. There was a complete lack of alkaline phosphatase activity in a medullary type of mammary cancer.
