*  Classification of Myoviridae bacteriophages using protein sequence similarity.
BACKGROUND: We advocate unifying classical and genomic classification of bacteriophages by integration of proteomic data and physicochemical parameters. Our previous application of this approach to the entirely sequenced members of the Podoviridae fully supported the current phage classification of the International Committee on Taxonomy of Viruses (ICTV). It appears that horizontal gene transfer generally does not totally obliterate evolutionary relationships between phages. RESULTS: CoreGenes/CoreExtractor proteome comparison techniques applied to 102 Myoviridae suggest the establishment of three subfamilies (Peduovirinae, Teequatrovirinae, the Spounavirinae) and eight new independent genera (Bcep781, BcepMu, FelixO1, HAP1, Bzx1, PB1, phiCD119, and phiKZ-like viruses). The Peduovirinae subfamily, derived from the P2-related phages, is composed of two distinct genera: the "P2-like viruses", and the ...

*  Characterization of bacteriophages infecting clinical isolates of Pseudomonas aeruginosa stored in a culture collection
Some clinical isolates of Pseudomonas aeruginosa stored in our culture collection did not grow or grew poorly and showed lysis on the culture plates when removed from the collection and inoculated on MacConkey agar. One hypothesis was that bacteriophages had infected and killed those clinical isolates. To check the best storage conditions to maintain viable P. aeruginosa for a longer time, clinical isolates were stored at various temperatures and were grown monthly. We investigated the presence of phage in 10 clinical isolates of P. aeruginosa stored in our culture collection. Four strains of P. aeruginosa were infected by phages that were characterized by electron microscopy and isolated to assess their ability to infect. The best condition to maintain the viability of the strains during storage was in water at room temperature. Three Siphoviridae and two Myoviridae phages were visualized and characterized ...

*  Isolation of Phages for Phage Therapy : A Comparison of Spot Tests and Efficiency of Plating Analyses for Determination of Host...
The increase of multi-resistant bacteria highlights that the golden era of antibiotics is ending and that alternative treatmentsare urgently needed. Phages have been historically used to treat bacterial infections prior to the discovery of antibiotics and have gained renewed interest in the past decade. Despite the advantages of phage therapy over traditional antibiotic usage, a number of concerns persist over their clinical application centring on their efficacy and safety. This thesis presents four papers that focus on the isolation and characterization of phages that target reference strains and drug-resistant strains of E. coli as well as their infection dynamics and kinetics. In Paper I, six of thirty isolated phages were selected to be characterized for their growth parameters and host range using two commonly used methods. The study showed that the host range (an important selection criteria for ...

The first therapeutic usage of phage was in 1919, to treat a child with severe dysentery. In a classic case of old-school science, the lead researchers (which included d'Herelle) drank portions of the bacteriophage preparation the day before treating the child, to check the treatment's safety in humans. The child fully recovered within a few days, and further tests confirmed the result. In the 1930s and 1940s, bacteriophage preparations were developed and sold by L'Oréal and the Eli Lilly Company. The development and mass production of antibiotics in the late 1940s halted research into phage therapy in the West, due to the superior efficacy and simplicity of antibiotics. ...

Viruses and bacteria evolve over time and can develop a resistance to antibiotics. In theory, this resistance can also apply to phages, but it may be less difficult to overcome compared to antibiotics.. Because phages are target specific, meaning only a one or very few bacterial strains are targeted upon, it is easier to develop new phages than new antibiotics. A time period of only a few days or weeks is needed to acquire new phages for resistant strains of bacteria, whereas it can take years to obtain new antibiotics. When resisting bacteria evolve, the assigned phages also evolve, so when super bacterium appears, an equivalent super phage fights it as long as the phage is derived from the same environment.. Compared to antibiotics, phages go deeper into the infected area. Antibiotics, on the other hand, ...
