The hydrolysis of collagen (collagenolysis) is one of the committed steps in extracellular matrix turnover. Within the matrix metalloproteinase (MMP) family distinct preferences for collagen types are seen. The substrate determinants that may guide these specificities are unknown. In this study, we have utilized 12 triple-helical substrates in combination with 10 MMPs to better define the contributions of substrate sequence and thermal stability toward triple helicase activity and collagen specificity. In general, MMP-13 was found to be distinct from MMP-8 and MT1-MMP(Delta279-523), in that enhanced substrate thermal stability has only a modest effect on activity, regardless of sequence. Matrix metalloproteinases (MMPs) are a group of structurally related proteolytic enzymes containing a zinc ion in the active site. In the Materials and Methods section it is stated that plasma MMP-9 levels were measured by a commercially available ELISA assay. The Amersham Biosciences, from which the assay was purchased, provides different types of assay for determination of MMP-9. One of them is for human MMP-9 and measures both free pro-MMP-9 and pro-MMP-9 complexed to TIMP-1. The other method, called MMP-9 activity assay, is used for determination of the active form of the compound.. It is not clear which form of MMP-9 was determined in the study. The MMP-9 human assay's range of detection in plasma is 4 to 128 ng/mL, whereas the median values obtained in the study were within the range of 54 to 225 ng/mL. Membrane-type 1 matrix metalloproteinase (MT1-MMP) is a type I transmembrane protein which exhibits biological activities on the cell surface. One of the characteristic functions of this enzyme is activation of proMMP-2 on the cell surface (1). This process can be monitored using gelatin zymography to detect the pro- and active forms of MMP-2. Cellular MT1-MMP activity can also be directly detected by in situ degradation of layers of either gelatin or collagen. By combining inhibitors, western blotting, and immunohistochemistry, one can detect and conclude the presence of MT1-MMP activity in the cells.
