Circulating IL-6 levels correlate with the severity of blood-stage malaria in humans and mouse models, but the impact of IL-6 classic signaling through membrane IL-6Rα, as well as IL-6 trans-signaling through soluble IL-6Rα, on the outcome of malaria has remained unknown. In this study, we created IL-6Rα-deficient mice that exhibit a 50% survival of otherwise lethal blood-stage malaria of the genus Plasmodium chabaudi. Inducing IL-6 trans-signaling by injection of mouse recombinant soluble IL-6Rα in IL-6Rα-deficient mice restores the lethal outcome to malaria infection. In contrast, inhibition of IL-6 trans-signaling via injection of recombinant sGP130Fc protein in control mice results in a 40% survival rate. Our data demonstrate that IL-6 trans-signaling, rather than classic IL-6 signaling, contributes to malaria-induced lethality in mice, preceded by an increased inflammatory response. Most breeders do not even look at soluble solids [sugars, organic acids, flavanoids] that give the fruit their flavor. Processing tomatoes are also less messy because the jelly does not squirt all over the place. When I started my PhD, I was able to browse through the fields in Davis and pick out the best tasting tomatoes to start my work [UC82B, marketed by a major seed company now, forget the name]. Background and objectives: Cardiospermum halicacabum L. is a climber plant of Sapindaceae family with certain medicinal properties that have made it to be used traditionally in different systems of medicine such as Ayurveda, Homeopathy and Unani medicine. Literature survey has revealed the reports about some investigations but references were not available about the seeds of C. halicacabum; hence, the seeds were selected for the present investigation. Methods: Detailed macroscopic and microscopic studies of C. halicacabum seeds and powder were carried out, physico-chemical parameters, preliminary and quantitative phytochemical analyses of different extracts of the seeds performed. Besides, the amount of total phenols and flavonoids in different extracts were determined. Results: The phytochemical screening gave a positive test for presence of various phytoconstituents like alkaloids, flavanoids, and tannins. The amount of total phenols was found to be between 29.697±0.232 to 187.372±0.615 mg
