Coloured scanning electron micrograph (SEM) of Dust mite (Dermatophagoides pteronyssinus). Millions of dust mites inhabit the home, feeding on dead human skin that are common in house dust. The mite's body is in three parts: the gnathosoma (head region) adapted for feeding on dead skin, the propodosma (carrying the 1st and 2nd pair of walking legs) and the hysterosoma (locating the 3rd and 4th pairs of legs). Dust mites produce 10-20 waste particles per day. The dead bodies and faecal pellets can trigger allergic responses. The whole life cycle from egg to adult takes approximately one month to complete, mature female mites can lay from 1-2 eggs per day. Adult mites can live up to two months. The most important house dust mites worldwide are Dermatophagoides farinae and Dermatophagoides pteronyssinus. Magnification: x78 when shortest axis printed at 25 millimetres. - Stock Image F017/4240

From houses and flats in Copenhagen inhabited by people allergic or non-allergic to house-dust, 69 samples of dust from mattresses have been collected and investigated for their content of mites and other arthropods. Three species of house-dust mites (Dermatophagoides pteronyssinus (Trt.), D. farinae Hughes, Euroglyphus maynei (Cooreman) Fain) were thus for the first time registered from Denmark besides some other mites and insects. The house-dust mites showed no preference for any of the rooms of the allergic patients nor for those of the controls (Table I). Outside the mattresses house-dust mites were found numerously below cross-bars, on wooden fillets and on floor panels beside the bed. In dust containing no skin scales living mites were not found; with air currents empty skins of mites had been transported around the room (Table 2). The house-dust mites are regarded ecologically to be the best adapted group of indoor mites to stand the low humidities (30-40 % R. H.) of ...

*  Intracellular 3′,5′-Adenosine Cyclic Monophosphate Level Regulates House Dust Mite-Induced Interleukin-13 Production by T...
SummaryWe studied the relationship between cAMP and house dust mite-induced cytokine production in T cells from mite-sensitive patients with atopic dermatitis. T cells from atopic dermatitis patients secreted high level of interleukin-13 (mean 851.1 pg per ml) when cultured with autologous monocytes pulsed with Dermatophagoides pteronyssinus extract. Dermato- phagoides pteronyssinus-induced interleukin-13 secretion was not detected in normal subjects. Adenylate cyclase inhibitor MDL 12,330A and cyclic nucleotide phosphodiesterase type 4 inhibitor rolipram blocked Dermatophagoides pteronyssinus-induced interleukin-13 secretion in atopic dermatitis T cells. In atopic dermatitis T cells, cAMP level rose at 5 min after Dermatophagoides pteronyssinus stimulus then decreased to the basal level at 1 h. MDL 12,330A blocked the Dermatophagoides pteronyssinus-induced ...

The current allergy diagnostic reagent of house dust mites marketed in the USA is at 10,000 AU/ml. This is based on the IDEAL50 concept put forth by Turkeltaub et al, almost 3 decades ago. Such concentration has been shown to give large degree of false positives even in the US population.. In Thailand, it was anecdotally observed that at such concentration, sizes of positive tests among atopic subjects were quite large and could lead to discomfort and a possible anaphylaxis in certain allergic individuals. Moreover, up to 40% of normal individuals could give positive reaction to this concentration.. It is therefore desirable to reexamine the appropriate concentration of diagnostic reagent of house dust mites among the Thai population and will be the main objective of this research. ...

*  Crystal Structure of Der f 7, a Dust Mite Allergen from Dermatophagoides farinae | ScholarBank@NUS
Background: Der f 7 is the group 7 allergen from the dust mite Dermatophagoides farinae, homologous to the major allergen Der p 7 from D. pteronyssinus. Monoclonal antibody that bind to residues Leu48 and Phe50 was found to inhibit IgE binding to residue Asp159, which is important for the cross-reactivity between Der f 7 and Der p 7. Methodology/Principal Findings: Here, we report the crystal structure of Der f 7 that shows an elongated and curved molecule consisting of two anti-parallel β-sheets - one 4-stranded and the other 5-stranded - that wrap around a long C-terminal helix. The overall fold of Der f 7 is similar to Der p 7 but key difference was found in the β1-β2 loop region. In Der f 7, Leu48 and Phe50 are in close proximity to Asp159, explaining why monoclonal antibody binding to Leu48 and Phe50 can inhibit IgE binding to Asp159. Both Der f 7 and Der p 7 bind weakly to polymyxin B via a similar binding site that is formed by the N-terminal helix, the 4-stranded ...

*  Analysis of predicted B and T-cell epitopes in Der p 23, allergen from Dermatophagoides pteronyssinus
House dust mite (HDM) allergy is the leading cause of IgE-mediated hypersensitivity. Therefore identifying potential epitopes in the Dermatophagoide pteronyssinus 23 (Der p 23), a major house dust mite allergen will aid in the development of therapeutic vaccines and diagnostic kits for HDM allergy. Experimental methods of epitope discovery have been widely exploited for the mapping of potential allergens. This study sought to use immunoinformatic methods to analyze the structure of Der p 23 for potential immunoreactive B and T-cell epitopes that could be useful for AIT and allergy diagnosis. We retrieved a Der p 23 allergen sequence from Genbank database and then analyzed it using a combination of web-based sequence analysis tools including the Immune Epitope Database (IEDB), Protparam, BCPREDS, ABCpred, BepiPred, Bcepred among others to predict the physiochemical properties and epitope spectra of the Der p 23 allergen. We then built 3D models of the predicted B-cell epitopes, T cell ...

*  Intradermal Skin Test in Dogs with Allergic Dermatoses - WSAVA2004 - VIN
Intradermal skin test is a method for diagnosis of allergic dermatoses and revelation of the allergens involved in dogs, but standardization of the procedure remains incomplete. The aim of the present study was to interpret the results of allergens, evaluate their clinical relevance and selection of candidate allergens for immunotherapy.. Thirty-nine aqueous allergen extracts and Staphylococcus phage lysate (SPL) were used for intradermal skin testing. Intradermal skin test was applied to 42 atopic dogs.. Positive intradermal skin test has been found in 26 (62%) dogs with allergic dermatitis. The most common positive hypersensitivity was against Dermatophagoides farinae (n=11), Dermatophagoides pteronyssinus (n=9), Staphylococcus aureus (n=9) and flea bite hypersensitivity (n=7). Specific immunomodulatory solutions were applied to 4 dogs against D. farinae and D. pteronyssinus. SPL intradermal test positive 7 dogs with recurrent pyoderma ...

The main objective of this study was to investigate the prevalence of Tp sensitization in elderly subjects in a veterans' nursing home using mite allergen extracts and recombinant allergens. A total of 199 subjects were enrolled in this study: 112 elderly subjects from a nursing home and 87 healthy young adults from the hospital staff as controls. The prevalence of Tp hypersensitivity was determined by specific IgE measurements and basophil histamine release. Immunoblotting with or without inhibition with Dermatophagoides pteronyssinus (Dp) was performed to identify the major allergens and species-specific allergen to Tp. It was determined that 39.3% (44/112) of the elderly population were sensitized to Tp and 17.9% (20/112) to Tp alone. There was a significantly higher prevalence of Tp hypersensitivity in elderly subjects in comparison with the young adult population. In the age association study of Tp and Dp sensitization, the elderly subjects were more sensitized to Tp ...

Background: TH17 cells participate in the pathogenesis of autoimmune and inflammatory diseases, such as asthma. Recently, our group identified associations of asthma and total serum IgE levels with single nucleotide polymorphisms (SNPs) in TH17 pathway genes. We now investigated functional effects of these SNPs on gene expression within the TH17 pathway in vitro.. Methods: TH17 pathway SNPs were genotyped (23 by MALDI-TOF MS; 2 by RFLP) in a randomly selected cohort (n=57, German adults). Expression of ten signature TH17 pathway genes was measured by quantitative real-time PCR in peripheral blood mononuclear cells, stimulated with phytohemagglutinin, D. pteronyssinus, lipid A, or peptidoglycan. Induction of mRNA levels was compared between unstimulated and stimulated cells applying the delta-delta Ct method and correlated with genotypes. Measurements of TH17 pathway cytokines were performed.. Results: The RORyt SNP rs6693413, with minor allele frequency (MAF) 0.48, displayed allele-specific ...

I graduated in the summer of 2008 right when the bottom fell out, and not everyone can be an engineer or a doctor, so some of us who are more creative or struggled with math and science and wanted to go to into careers that suit our talents couldn't find a job in those fields and ended up unemployed because there wasn't much professional work available. It took me two and a half years to find my first "real job" after working two jobs part time to get me experience (many jobs after the bank crash weren't entry level and many of them wanted 3 years of experience) for the field I wanted to get into: marketing. Happy to say I'm now employed as a Marketing Associate and while I wish this would have happened a couple years earlier I just have to grudgingly admit that sometimes life gives you a challenge and that you just have to work hard and stick it out to do what you want to do and be where you want to be. ...
