*  Cells | Free Full-Text | Optimal Thawing of Cryopreserved Peripheral Blood Mononuclear Cells for Use in High-Throughput Human...
Cryopreserved peripheral blood mononuclear cells (PBMC) constitute an important component of immune monitoring studies as they allow for efficient batch- testing of samples as well as for the validation and extension of original studies in the future. In this study, we systematically test the permutations of PBMC thawing practices commonly employed in the field and identify conditions that are high and low risk for the viability of PBMC and their functionality in downstream ELISPOT assays. The study identifies the addition of ice-chilled washing media to thawed cells at the same temperature as being a high risk practice, as it yields significantly lower viability and functionality of recovered PBMC when compared to warming the cryovials to 37 °C and adding a warm washing medium. We found thawed PBMC in cryovials could be kept up to 30 minutes at 37 °C in the presence of DMSO before commencement of washing, which surprisingly identifies exposure to DMSO as a low risk step during the thawing process.

Embryo cryopreservation involves the generation of pre-implantation embryos using males provided by the investigator and the controlled rate cooling of collected embryos to -30°C followed by rapid freezing of embryos in liquid nitrogen to -196°C.. While embryo cryopreservation is the gold standard for preserving mouse germplasm, not all mouse strains are suitable for embryo cryopreservation. However, we have been successful at cryopreserving embryos from several different mouse strains.. Embryos for cryopreservation can be obtained in two basic ways: ...

*  Embryo Cryopreservation - Frisco Fertility Clinic - Fertility Specialists of Texas
There are many benefits of oocyte and embryo cryopreservation. When used as part of the IVF treatment process, cryopreservation can increase the likelihood of achieving pregnancy for many patients. Cryopreservation also allows couples to continue to see their families grow as IVF cycles can be repeated, years later, from the same group of eggs originally recovered for treatment.. During an IVF cycle, between 10 and 30 eggs can typically be recovered from the ovary. While it is likely that not all of these eggs will be deemed appropriate for cryopreservation, the process frequently allows couples to save a number of eggs for multiple conception attempts. In some cases, this can lead to successful treatment without the use of fertility drugs. It also reduces the likelihood that another egg retrieval procedure will be required. The number of embryos transferred depends on the number of eggs available, age, and other factors that are unique to ...

Populations of Redside Dace Clinostomus elongatus have declined in many areas across the species'North American range. Therefore, the development of sperm cryopreservation technology would provide an invaluable means of preserving genetic diversity in populations that are in imminent danger of extirpation. We developed cryopreservation protocols by testing the effects of diluent (buffered sperm motility-inhibiting saline solution [BSMIS]; BSMIS + glycine; sucrose; and Hanks' balanced salt solution [HBSS]), cryoprotectant (dimethyl sulfoxide [DMSO]; propylene glycol [PG]; N,N-dimethylacetamide [DMA]; and methanol), freezing rate (1, 5, and 10◦C/min), and male-to-male variation on sperm quality. Incubating sperm in extenders affected motility;BSMIS + glycine + methanol,BSMIS + glycine + PG, and HBSS + methanol were the only treatments for which motility was not significantly different from that of fresh sperm. Sperm frozen with sucrose had higher motility than sperm frozen with ...

*  Summary: Cryopreservation of Human Unfertilized and Fertilized Oocytes
Cryopreservation of human fertilized oocytes and embryos are nowadays well established in IVF practice with a wide range of clinical applications. However, freezing of unfertilized MII oocytes turned out to be one of the greatest challenges in the field of human reproductive cryobiology since the protocols remained ineffective for over 25 years. Only in the last 10 years the efficiency and safety of oocyte cryopreservation tremendously improved with the realization that zona hardening occurs in the process of cryopreservation and the introduction of vitrification. The possibility to cryopreserve unfertilized oocytes introduces new applications in the field of cryopreservation. We have now the opportunity to preserve fertility for cancer patients or to freeze oocytes for IVF patients when now sperm is available at the day of pick-up. This chapter will be dedicated to the vitrification of unfertilized MII oocytes and fertilized oocytes ...

*  Effective Cryopreservation of Neural Stem or Progenitor Cells Wit...: Ingenta Connect
Development of effective cryopreservation protocols will be essential to realizing the potential for clinical application of neural stem and progenitor cells. Current cryopreservation protocols have been largely employed in research, which does not require as stringent consideration of viability and sterility. Therefore, these protocols involve the use of serum and protein additives, which can potentially introduce contaminants, and slow cooling with DMSO/glycerol-based cryopreservation solutions, which impairs cell survival. We investigated whether serum- and protein-free vitrification is effective for functional cryopreservation of neurosphere cultures of neural stem or progenitor cells. To protect the samples from introduction of other contaminants during handling and cryostorage, an original "straw-in-straw" method (250 l sterile straw placed in 500 l straw) for direct immersion into liquid nitrogen and storing the samples was also ...

*  The effect of developmental status and excision injury on the success of cryopreservation of germplasm from non-orthodox seeds.
The zygotic germplasm of plant species producing desiccation-sensitive seeds can be conserved in the long-term only by cryopreservation. Usually the embryonic axis is excised from the cotyledons and is used as the explant for cryopreservation as it is small and provides a large surface area:volume ratio. However the shoot of the axis of most species studied does not develop after excision, with the result that survival after cryopreservation is often recorded as callus production or simply explant enlargement and/or greening. Thus, besides explant size, factors such as in vitro regeneration techniques, physical injury induced upon excision and developmental status of the seed could compromise the success of cryopreservation. This study investigated the effect of the factors mentioned above, with particular attention to the developmental status of the seeds on explant in vitro development (section 3.1), response to dehydration (section 3.2) ...

There are multiple reasons to consider egg cryopreservation. There are multiple reasons to consider egg cryopreservation. Women who are not ready to become pregnant but who want to have a family later in life may choose to freeze eggs for social reasons. It is also useful for women with certain genetic conditions known to reduce the ovaries' ability to produce eggs. Cryopreservation may also be used to save eggs after they have been retrieved for IVF if sperm are unable to be obtained the same day. Couples may also choose to preserve eggs as an alternative to embryo cryopreservation during an IVF cycle.. ...

Vitrification is an ultra‐rapid cryopreservation technique that provides excellent survivability by using optimal concentrations of cryoprotectants used in a step‐wise process supporting rapid dehydration of the oocyte. The dehydrated oocyte is loaded into a thin storage device that will facilitate ultra‐rapid cooling of the oocyte when plunged into liquid nitrogen - eliminating concerns of damaging ice‐crystal formation associated with traditional slow‐freezing procedures. Thaw solutions are formulated to support rehydration through the rapid warming process that follows vitrification to optimize cellular survival.. Irvine Scientific's vitrification system is being used in hundreds of clinics worldwide to support robust cryopreservation programs that are delivering babies every day. These solutions support implantation and pregnancy rates comparable to those of fresh cycles.. ...

*  Risk of transferring malignant cells with transplanted frozen-thawed ovarian tissue - Danish National Research Database-Den...
Ovarian tissue cryopreservation and transplantation is a real option to preserve and restore fertility in young cancer patients. However, there is a concern regarding the possible presence of malignant cells in the ovarian tissue, which could lead to recurrence of the primary disease after reimplantation. A review of the existing literature was done to evaluate the risk of transplanting malignant cells in case of the main malignant indications for ovarian tissue cryopreservation. For ovarian tissue from patients with hematologic malignancies, it is of paramount importance to identify minimal residual disease before ovarian tissue transplantation. Indeed, these pathologies, reviewed here in detail, are considered to be most at risk of ovarian metastasis ...

STUDY QUESTION: What is the intracellular concentration of cryoprotectant (ICCP) in mouse zygotes during vitrification (VIT) and slow-freezing (SLF) cryopreservation procedures? SUMMARY ANSWER: Contrary to common beliefs, it was observed that the ICCP in vitrified zygotes is lower than after SLF, although the solutions used in VIT contain higher concentrations of cryoprotectants (CPs). WHAT IS KNOWN ALREADY: To reduce the likelihood of intracellular ice crystal formation, which has detrimental effects on cell organelles and membranes, VIT was introduced as an alternative to SLF to cryopreserve embryos and gametes. Combined with high cooling and warming rates, the use of high concentrations of CPs favours an ...

*  Research in Cryobiology, Cryopreservation and Cryonics
At present, cryopreservation technology is only successful for cell lines and small tissues. More research is required before whole organs can successfully be cryopreserved while retaining their biological integrity. Given the importance of organ transplants and the growing field of tissue engineering, perfecting cryopreservation methods would have a profound impact on medicine.. There has been progress in cryopreservation and cryogenics. A team led by Hal Sternberg of Biotime claimed to bring back to life baboons that were previously cooled to one degree Celsius. They injected a substance called Hextend to replace the blood and protect the cells from deterioration. Incredibly, this may soon become routine for emergency care patients. Hasan Alam at Massachusetts General Hospital is working on a similar approach to drop the temperature of emergency patients and protect the bodies from damage while undergoing surgery. Replacing the blood of critical care ...

This advisory is written for those who would like to understand the processes of mouse strain cryopreservation, why it is important and how to take advantage of the services offered. The most efficient method to protect mouse strains is by cryopreservation of gametes or embryos. Cryopreservation, and especially re-animation from frozen material, is not a trivial exercise, nor is the establishment of a cryopreservation and IVF laboratory cheap. There are numerous laboratories around the world that offer these services.

... or embryo freezing is a method used to preserve embryos by cooling and storing them at low temperatures (-196°C). They can then be thawed at a future date and transferred to the uterus, providing additional opportunity for achieving conception.. As part of the usual process of in vitro fertilization, multiple eggs may be stimulated to grow, be recovered from the ovary and become fertilized. This may result in additional embryos in excess of the number that a couple would desire to have transferred back to the uterus at one time. If the additional embryos are of sufficiently good quality to undergo the process of cryopreservation, this can be performed in order to provide another opportunity for embryo transfer. Depending on the embryo stage at the time of freezing, between 60-90% survive freeze/thaw process resulting in a future pregnancy.. If the IVF fresh embryo transfer does not result in pregnancy, the frozen embryos can be subsequently thawed and transferred to the ...
