*  British Library EThOS: Total synthesis of the macroline-related alkaloid (±)-alstonerine
This thesis examines the total synthesis of the macroline-related indole alkaloid alstonerine and related compounds. It is divided into three sections: The first section provides a review of the total synthesis efforts reported by Cook, Martin, Kuethe, and Kwon, as well previous work within the Craig group. The second section discusses the results of our investigations. The optimisation of the synthesis of key intermediate α,β-unsaturated lactam alcohol via directed-aziridine ring-opening is presented in detail. Our progress towards the synthesis of macroline-related alkaloids macroline, alstolactone, anhydromacrosalhine-methine and alstonerinal, as well as their N4-tosyl derivatives, from the key intermediate is discussed. The findings from these studies are presented en route to the total synthesis of alstonerine. The third section contains experimental procedures and characterisation data for compounds synthesised ...

*  British Library EThOS: Dendron-based synthetic bacteriophages for the treatment of Proteus mirabilis infections
In the last two decades a surge in antibiotic resistance has limited antibiotic effectiveness increasing the risk of uncontrolled epidemics especially for biofilm-related infections. The National Institute of Health reports that 80 % of human infections are biofilm related. The Proteus mirabilis bacteria were focused on in this study as they are significant biofilm formers in chronic infections such as biofilm-related urinary tract infections for which there are currently no completely effective treatment strategies. As biofilms can increase antibiotic resistance by up to 1000-fold, there is an urgent need for the development of novel antimicrobials. Thus, bacteriophages which are viruses that target and kill bacteria have been proposed as suitable alternatives, but factors like storage stability and re-isolation pose limitations. Towards investigating the development of new antimicrobial strategies, the aims of this thesis were to assess: (i) the therapeutic potential of bacteriophages against ...

*  British Library EThOS: The role of p53 and ASPP2 in neurodegenerative disease
Two cellular processes of central importance to cancer and neurodegeneration are apoptosis and cellular senescence. Both are a means of cellular suicide that are utilized in time- and context-dependent manners and have important evolutionary purposes. However, they are also the drivers of many deleterious processes underlying cancer and neurodegeneration. Many of the cellular responses to aging and age-related diseases, including apoptosis and senescence, converge on the tumor suppressor pathway, p53. Here I examine the molecular basis for loss of cell polarity and accelerated cell death mediated by apoptosis stimulating protein of p53 2 (ASPP2) in neurodegeneration. In this study we find that ASPP2 mediates STAT1Linduced apoptosis. Lipopolysaccharide (LPS) induces ASPP2 mRNA expression in vitro. Also, LPS induces nuclear ASPP2 in vivo at the blood-cerebral spinal fluid-barrier (BCSFB), the brain's barrier to inflammation. Consistent with ASPP2's role as a gatekeeper to inflammation, ASPP2 ...

*  British Library EThOS: Bionics chemical synapse
This thesis presents the very first bionics chemical synapse which has the capability to sense the neurotransmitter (glutamate) and imitates the physiological behaviour of certain chemical synapse receptors (i.e. AMPA, NMDA, GABAA and GABAB). This bionics chemical synapse consists of two main parts: the glutamate ISFETs that act as neurotransmitter sensors and the current-mode CMOS circuits that have been designed to match the physiological behaviour of the chemical synapses. This bionics chemical synapse requires a sub-nano Siemens operational transconductance amplifier (OTA) to develop a low conductance gain for each chemical synapse receptor (0.1nS). A combination of two OTA designs was required to decrease the overall transconductance gain, which were: the bulk driven transistor and the drain current normalisation. To create the bionics chemical synapse, a neurotransmitter sensor is required as the chemical front-end for each receptor circuit. The sensor that was used is an enzyme-modified ...

*  British Library EThOS: Obesity and bone metabolism
Obese adults have a lower risk of hip and vertebral fracture, but a greater risk of lower limb and proximal humerus fracture, compared to adults with a normal body mass index (BMI). Differences in fracture risk by skeletal site in obesity might be attributed to differences in bone mineral density (BMD), bone microstructure and bone strength between obese and normal BMI individuals and/or differences in physical function, possibly related to fall frequency and/or direction. The role of vitamin D in bone metabolism and physical function in obesity is unclear. The effect of obesity on bone microstructure and strength in young and older, men and women, has not been investigated in a matched case control design. BMD, microstructure and strength were determined using novel imaging technologies. The roles of various adipose compartments and biochemical factors on BMD and microarchitecture were investigated. Physical function and vitamin D metabolism of obese and normal BMI individuals was compared. ...
