*  J Clin Psychiatry/Clozapine-Induced Agranulocytosis in Finland, 1982-2007: Long-Term Monitoring of Patients Is Still Warranted
Objective: Recent studies suggest that restrictions on the use of clozapine should be reassessed considering the risk-benefit ratio. We analyzed all cases of clozapine-induced agranulocytosis reported to the Finnish National Agency for Medicines between 1982 and 2007.. Method: In this retrospective longitudinal study, we defined agranulocytosis as a neutrophil count below 0.5 × 109/L and, accordingly, identified a total of 163 patients with clozapine-induced agranulocytosis. We collected all available information on patient demography, as well as on daily clozapine doses, treatment duration, concomitant medication prior to the onset of agranulocytosis, and infections during the adverse event. The amount of clozapine used annually in Finland was estimated on the basis of the defined daily dose, and the frequency of agranulocytosis was calculated from the absolute number of cases in relation ...

*  Pentazocine and Agranulocytosis | Annals of Internal Medicine | American College of Physicians
To the editor: Adverse reactions to drugs are an important cause of hospital-acquired illness, occurring in 10% to 18% of medical patients (1) and producing fatality rates of up to 13%. Drug-induced neutropenia or agranulocytosis has been associated with many types of drugs and often is associated with a fatal outcome. Recently Hoppin and colleagues (2) described three cases of pentazocine-related agranulocytosis. All of their cases occurred in patients with previously injured bone marrows but with normal peripheral granulocyte counts before this phenomenon.. We wish to present an additional case of pentazocine-induced agranulocytosis, but one that occurred without a previous ...

*  Agranulocytosis, Acquired - NORD (National Organization for Rare Disorders)
Symptoms of the following disorders can be similar to those of Acquired Agranulocytosis. Comparisons may be useful for a differential diagnosis:. Chronic Granulomatous Disease is a very rare blood disorder that affects specialized white blood cells (i.e., neutrophils) and is characterized by widespread granular lesions in many areas of the body. People with this disorder have repeated infectious diseases, including abscesses of the liver; respiratory infections and pneumonia; and inflammation of the lymph nodes (suppurative lymphadenitis) and the spinal cord (osteomyelitis). Chronic infections may be evident in the liver, gastrointestinal tract, eyes, and/or brain. (For more information on this disorder, choose "Granulomatous Disease" as your search term in the Rare Disease Database.). Wegener's Granulomatosis is a rare multisystem disease that is characterized by inflammation and degenerative changes in the blood vessels that serve the lungs and upper respiratory tract. *  Antithyroid agent - Wikipedia
An antithyroid agent is a hormone antagonist acting upon thyroid hormones. The main antithyroid drugs are carbimazole (in the UK), methimazole (in the US), and propylthiouracil/PTU. A less common antithyroid agent is potassium perchlorate. In Graves' disease, treatment with antithyroid medications must be given for six months to two years, in order to be effective. Even then, upon cessation of the drugs, the hyperthyroid state may recur. Side effects of the antithyroid medications include a potentially fatal reduction in the level of white blood cells. A randomized control trial testing single dose treatment for Graves' found methimazole achieved euthyroid state more effectively after 12 weeks than did propylthyouracil (77.1% on methimazole 15 mg vs 19.4% in the propylthiouracil 150 mg groups). But generally both drugs are considered equivalent. A study has shown no difference in outcome for adding thyroxine to antithyroid medication and continuing thyroxine ...
