*  The C3-V4 region is a major target of autologous neutralizing antibodies in human immunodeficiency virus type 1 subtype C...
The early autologous neutralizing antibody response in human immunodeficiency virus type 1 (HIV-1) subtype C infections is often characterized by high titers, but the response is type specific with little to no cross-neutralizing activity. The specificities of these early neutralizing antibodies are not known; however, the type specificity suggests that they may target the variable regions of the envelope. Here, we show that cross-reactive anti-V3 antibodies developed within 3 to 12 weeks in six individuals but did not mediate autologous neutralization. Using a series of chimeric viruses, we found that antibodies directed at the V1V2, V4, and V5 regions contributed to autologous neutralization in some individuals, with V1V2 playing a more substantial role. However, these antibodies did not account for the total neutralizing capacity of these sera against the early autologous virus. Antibodies directed ...

*  PLOS Pathogens: Broadly Neutralizing Human Anti-HIV Antibody 2G12 Is Effective in Protection against Mucosal SHIV Challenge...
Author Summary An effective HIV vaccine should elicit broadly neutralizing antibodies, i.e. antibodies that neutralize a wide spectrum of different HIVs in vitro. A number of human monoclonal antibodies have been isolated with broad neutralization and shown to protect macaques against vaginal HIV challenge. Protection is generally correlated with neutralization and requires relatively high antibody concentrations that may be difficult to achieve by vaccination. Here, we show that one monoclonal antibody (2G12) is unusually potent in protection relative to its neutralizing ability as hinted at by earlier data. Further studies eliminate an unusual ability of 2G12 to be transported to the vagina (site of infection) as a possible explanation for our observations. Although the precise mechanism is unclear, the studies have important implications for HIV vaccine design in general by suggesting ...

*  4. IMMUNOLOGY. - :: BIOTED
This experiment's aim is to provide students the molecular biology and pathogenesis basics of acquired immunodeficiency syndrome (AIDS).. An HIV test detects HIV infection indirectly using an ELISA test against HIV antibodies in the blood. The test works by taking antibodies from the patient's blood and adding them to a microtiter plate coated with HIV antigen. If HIV antibodies are present in the blood, they will bind to the antigens on the plate. This binding is detected with an enzyme-linked secondary antibody that causes a color change upon addition of substrate. In this experiment, students will perform an ELISA test by coating microtiter plate wells with simulated HIV antigen and then test simulated donor serum for anti-HIV antibodies.. , Kit includes: ...

*  Building Better HIV Antibodies - Redorbit
Caltech biologists create neutralizing antibody that shows increased potency. Using highly potent antibodies isolated from HIV-positive people, researchers have recently begun to identify ways to broadly neutralize the many possible subtypes of HIV. Now, a team led by biologists at the California Institute of Technology (Caltech) has built upon one of these naturally occurring antibodies to create a stronger version they believe is a better candidate for clinical applications.. Current advances in isolating antibodies from HIV-infected individuals have allowed for the discovery of a large number of new, broadly neutralizing anti-HIV antibodies directed against the host receptor (CD4) binding site-a functional site on the surface of the virus that allows for cell entry and infection. Using a technique known as structure-based rational design, the team modified one ...
