Chondroitin/dermatan sulfate glycosyltransferase genes are essential for craniofacial development.
Habicher J, Varshney GK, Waldmann L, Snitting D, Allalou A, Zhang H, Ghanem A, Öhman Mägi C, Dierker T, Kjellén L, Burgess SM, Ledin J PLoS Genet. 18 (2) e1010067 [2022-02-00; online 2022-02-22] Chondroitin/dermatan sulfate (CS/DS) proteoglycans are indispensable for animal development and homeostasis but the large number of enzymes involved in their biosynthesis have made CS/DS function a challenging problem to study genetically. In our study, we generated loss-of-function alleles in zebrafish genes encoding CS/DS biosynthetic enzymes and characterized the effect on development in single and double mutants. Homozygous mutants in chsy1, csgalnact1a, csgalnat2, chpfa, ust and chst7, respectively, develop to adults. However, csgalnact1a-/- fish develop distinct craniofacial defects while the chsy1-/- skeletal phenotype is milder and the remaining mutants display no gross morphological abnormalities. These results suggest a high redundancy for the CS/DS biosynthetic enzymes and to further reduce ...

Ceramide-Specific Glycosyltransferase
The synthesised haem is transported outside of mitochondria and utilised for the maturation of haem proteins. to stress insults. Since the stress-induced cell damage was exacerbated from the pharmacological blockade of haem rate of metabolism but was ameliorated by the addition of biliverdin and bilirubin, it is likely the de novo synthesis of haem and subsequent conversion to bilirubin play indispensable cytoprotective functions against cell damage. The biosynthesis of haem requires eight enzymes, whereas its catabolism requires three. The 1st and last three methods in haem biosynthesis take place in the mitochondria (Supplementary Fig. S1). In the first step, 5-aminolevulinic acid (ALA) synthase catalyses the condensation of glycine and succinyl-CoA to form ALA1,2. Ferrochelatase is the terminal enzyme in haem biosynthesis, catalysing the insertion of ferrous ions into protoporphyrin IX (PPIX) to form haem3,4. The synthesised haem is definitely transported outside of mitochondria and utilised ...
