Incubation of adriamycin resistant Chinese hamster lung cells with low levels of N-ethylmaleimide (NEM) results in a major increase in the cellular accumulation of drug. When resistant cells are prelabeled with [32Pi] and thereafter treated with NEM there also occurs a selective superphosphorylation of an 180K plasma membrane glycoprotein (P-180). This phosphorylation reaction occurs at both serine and threonine residues. In similar experiments with drug sensitive cells only minor levels of this protein can be detected. Detailed studies have established that in cells which have reverted to drug sensitivity there is a parallel loss in the presence of phosphorylated P-180. Treatment of patients with cyclosporin A (CsA) increases low-density lipoprotein (LDL) cholesterol levels. We investigated whether an elevated hepatic secretion of apolipoprotein (apo) B-100-containing lipoproteins is responsible for the increase of LDL by using the human hepatoma cell line HepG2. Addition of CsA to the culture medium of HepG2 cells resulted in a dose- and time-dependent decrease in the secretion of apoB-100. Maximal inhibition (-50%), which was obtained at 5 mumol/L CsA, was achieved within 8 hours. The secretion of apoA-I, albumin, and [35S]methionine-labeled proteins was not affected by CsA. We have previously described avian leukosis virus-based packaging cell lines that express gag, pol, and env proteins from two transcomplementing genomes and produce helper-free stocks of retroviral vectors with different host ranges. In this report, we demonstrated that (i) despite the deletion of the psi packaging sequence, the packaging-defective transcomplementing retroviral transcripts were packaged into virions at a level that could reach 2.3% of a wild-type virus packaging level and (ii) despite deletion of the 3' LTR, these genomes were transferred along with the vector to target cells. As these genomes were also bearing a selectable gene, titers of the resulting contaminant particles could be estimated, depending on the cell line to be between 0 and 6 infectious particles/ml of supernatant.
