Angiotensin II (AII) and K+ raise the cytosolic free Ca2+ concentration [(Ca2+]i) and stimulate aldosterone production in isolated bovine adrenal glomerulosa cells. The mechanisms leading to an elevation of [Ca2+]i were analysed with the fluorescent Ca2+ probe quin 2. (1) Whereas [Ca2+]i rose transiently and returned to basal values within 5 min in response to AII, the effect of K+ was sustained for at least 15 min. (2) AII released Ca2+ from intracellular stores, whereas the [Ca2+]i response to K+ depended solely on extracellular [Ca2+]. With increasing concentrations of angiotensin II logarithmic dose-response curves for aldosterone and cyclic AMP production were similar. Here, we report the novel findings that in human adrenal glomerulosa cells both basal and stimulated rates of aldosterone production depend on voltage-gated calcium currents through T-type as well as L-type calcium channels. These findings are surprising because patch-clamp studies consistently report the basal membrane potential of ZG cells to be very hyperpolarized (around −80 mV), a potential at which high threshold L-type calcium channels are not expected to be active. We therefore examined the effects of nuclear retinoid X receptor (RXR) pan-agonist PA024 on CYP11B2 expression, aldosterone secretion and blood pressure, to elucidate its potential as a novel anti-hypertensive drug. The corticosteroids are synthesized from cholesterol within the adrenal cortex. Most steroidogenic reactions are catalysed by enzymes of the cytochrome P450 family. They are located within the mitochondria and require adrenodoxin as a cofactor (except 21-hydroxylase and 17α-hydroxylase). Aldosterone and corticosterone share the first part of their biosynthetic pathway. The last part is either mediated by the aldosterone synthase (for aldosterone) or by the 11β-hydroxylase (for corticosterone). These enzymes are nearly identical (they share 11β-hydroxylation and 18-hydroxylation functions). But aldosterone synthase is also able to perform a 18-oxidation. Moreover, aldosterone synthase is found within the zona glomerulosa at the outer edge of the adrenal cortex; 11β-hydroxylase is found in the zona fasciculata and reticularis. The G protein-coupled receptor GPR30 has recently been identified as a nonnuclear estrogen receptor. The classic renin-angiotensin system is partly responsible for controlling aldosterone secretion from the adrenal cortex via the peptide angiotensin II (AngII). In addition, there is a local adrenocortical renin-angiotensin system that may be involved in the control of aldosterone synthesis in the zona glomerulosa (ZG). In order to characterize the long-term control of adrenal steroidogenesis, we utilized adrenal glands from renin knockout (KO) rats and compared steroidogenesis in vitro and steroidogenic enzyme expression to wild-type (WT) controls (Dahl S rat). Adrenal capsules (ZG; aldosterone production) and subcapsules (zona reticularis/fasciculata [ZFR]; corticosterone production) were separately dispersed and studied in vitro. Corticosterone is important in the regulation of blood glucose, the ability to handle stress, and maintaining normal immune function. Aldosterone results in the retention of sodium and excretion of potassium causing water retention, a greater blood volume, and an increase in blood pressure (hypertension). Aldosterones role in hypertension has previously been studied using receptor blocking agents or via complete adrenalectomy (removal of both adrenal glands). Complete adrenalectomy removes all hormones produced by the adrenal gland, and often results in death. Adrenal venous blood was collected from hypophysectomized and sham-hypophysectomized dogs 6 days postoperatively. 17-Hydroxycorticosterone, corticosterone, 11-desoxy-17-hydroxycorticosterone and aldosterone were isolated by paper chromatography. Histological examination of the sellar and suprasellar regions of the hypophysectomized dogs demonstrated the absence of pituitary tissue. The adrenal glands of the hypophysectomized dogs showed cortical atrophy which did not involve the zona glomerulosa. The rate of secretion of aldosterone by the hypophysectomized dogs was found to be approximately 66% of that of the control, sham hypophysectomized, dogs. The rates of secretion of 17-hydroxycorticosterone, corticosterone and 11-desoxy-17-hydroxycorticosterone were found to be approximately 10% of that of the controls. The diameter data were then processed by Bachs method which deals with the sphere size distribution. The structural parameters of the mitochondria were computed with the aid of an electronic computer. The total number of mitochondria in all the parenchymal cells of the zona fasciculata were calculated. The surface area of the inner mitochondrial membrane was then determined stereologically. Biochemical parameters were obtained for the protein, the phospholipid, and the cytochrome P-450 content, per averaged mitochondrion. The number of cytochrome P-450 molecules contained in the inner membrane was determined in terms of the unit surface area and of the unit amount of phospholipid.
