DNA double-strand breaks (DSBs) containing unligatable termini are potent cytotoxic lesions leading to growth arrest or cell death. The Artemis nuclease and tyrosyl-DNA phosphodiesterase (TDP1) are each capable of resolving protruding 3′-phosphoglycolate (PG) termini of DNA double-strand breaks (DSBs). Consequently, a knockout of Artemis and a knockout/knockdown of TDP1 rendered cells sensitive to the radiomimetic agent neocarzinostatin (NCS), which induces 3′-PG-terminated DSBs. Unexpectedly, however, a knockdown or knockout of TDP1 in Artemis-null cells did not confer any greater sensitivity than either deficiency alone, indicating a strict epistasis between TDP1 and Artemis. Moreover, a deficiency in Artemis, but not TDP1, resulted in a fraction of unrepaired DSBs, which were assessed as 53BP1 foci. To further understand the involvement of INT6 in DDR, we investigated whether the ATM kinase, essential for cellular signaling in response to DNA damage, was normally activated upon INT6 deficiency. First, the phosphorylation status of its substrate CHK2 was examined. As expected, irradiation of control siRNA-treated cells led to a rapid and optimal phosphorylation of CHK2 within 30 minutes after irradiation that significantly decreased by 2 hours (Fig. 2A). In contrast, INT6-depleted cells failed to show similar levels of increase at 30 minutes or at a later time point. This defect in CHK2 phosphorylation was not caused by a reduction in CHK2 protein levels (Fig. 2A). Phosphorylation of the checkpoint kinase CHK1 was also reduced in these cells (Fig. 2A). Nine-membered enediyne antitumor antibiotics C-1027, neocarzinostatin (NCS), and kedarcidin (KED) possess enediyne cores to which activity-modulating peripheral moieties are attached via (R)- or (S)-vicinal diols. We have previously shown that this stereochemical difference arises from hydrolysis of epoxide precursors by epoxide hydrolases (EHs) with different regioselectivities. The inverting EHs, such as SgcF, hydrolyze an (S)-epoxide substrate to yield an (R)-diol in C-1027 biosynthesis, whereas the retaining EHs, such as NcsF2 and KedF, hydrolyze an (S)-epoxide substrate to yield an (S)-diol in NCS and KED biosynthesis. The translocation of protein toxins into a cell relies on a myriad of protein-protein interactions. One such group of toxins are enzymatic E colicins, protein antibiotics produced by Escherichia coli in times of stress. These proteins subvert ordinary nutrient uptake mechanisms to enter the cell and unleash nuclease activity. We, and others, have previously shown that uptake of ColE9 (colicin E9) is dependent on engagement of the OM (outer membrane) receptors BtuB and OmpF as well as recruitment of the periplasmic protein TolB, forming a large supramolecular complex. Intriguingly, colicins bind TolB using a natively disordered region to mimic the interaction of TolB with Pal (peptidoglycan-associated lipoprotein). Scientific literatures have not documented any evidence of the antitumour potential of Bauhinia variegata against B16F10 melanoma tumor model in C57BL mice. The present investigation was undertaken to explore the antitumour activity of Leaf, stem bark and flower extract of Bauhinia variegata against B16F10 melanoma tumour model in C57BL mice. Hydro-methanolic extract prepared from the leaf, stem bark and flower of Bauhinia variegata were assessed for their antitumor activity. Inositol pyrophosphates, such as diphosphoinositol pentakisphosphate (IP(7)), are water-soluble inositol phosphates that contain high energy diphosphate moieties on the inositol ring. Inositol hexakisphosphate kinase 1 (IP6K1) participates in inositol pyrophosphate synthesis, converting inositol hexakisphosphate (IP(6)) to IP(7). In the present study, we show that mouse embryonic fibroblasts (MEFs) lacking IP6K1 exhibit impaired DNA damage repair via homologous recombination (HR). IP6K1 knock-out MEFs show decreased viability and reduced recovery after induction of DNA damage by the replication stress inducer, hydroxyurea, or the radiomimetic antibiotic, neocarzinostatin. Cells lacking IP6K1 arrest after genotoxic stress, and markers associated with DNA repair are recruited to DNA damage sites, indicating that HR repair is initiated in these cells. Our study provides new insights into mechanisms underlying the mitochondrial dysfunction in WFS1-deficient cells involving NCS1, a WFS1 interacting protein. We demonstrated that WFS1 interacted with the Ca2+ sensor NCS1 (39) and that NCS1 protein abundance was substantially decreased in WFS1-deficient cells. Addition of NCS1 to planar lipid bilayers or overexpression of NCS1 in PC12 cells results in increased IP3R channel activity (30-32), which should lead to increased Ca2+ release from the ER. In fibroblasts from our Wolfram syndrome patients, a decrease in Ca2+ flux from the ER was observed only under histamine application, whereas ER Ca2+ load was unchanged, suggesting a role of NCS1 in regulating IP3R activity. In addition, there was a concomitant decrease in the [Ca2+]m uptake. This year North Carolina Swimming will recognize its Top Five swimmers, All Star swimmers, and Swimmers of the Year for the past three seasons; the 2011 Long Course season, the 2011-12 Short Course season, and the 2012 Long Course season.
