We show that pH homeostasis varies among individual bacterial cells, both among planktonic cells adhered to a slide and among cells within a biofilm grown on a slide. This variability implies that the pH response and recovery measurements obtained for cell suspensions miss important aspects of pH homeostasis of individual cells. The variation we saw could be associated with substrate adherence and with variability among suspended cells. Properties of adherent cells are highly important for medical, environmental, and industrial applications.. To measure pH homeostasis, we used ratiometric GFP combined with fluorescence microscopy, both for E. coli (Fig. 4) and for B. subtilis (Fig. 8). Ratiometric pHluorin (18) combines the advantages of GFP fluorimetry (14, 15, 36) and fluorescence microscopy. The fact that four of the nine acid-inducible genes in the gadA region encode transcription factors suggests that the AR genes are subject to complex regulation. We previously identified a conserved 20-bp sequence element upstream of gadA, gadB, and hdeA (25). This 20-bp sequence was recently shown to function in pH control of transcription (8). We and others have suggested that this 20-bp sequence might be the target of binding by the transcription factor YhiX (8, 25). In fact, overexpression of YhiX causes elevated expression of gadA, gadB, and hdeD, suggesting that YhiX is an activator of these genes (20, 23). Future work planned in this laboratory will shed light on regulation by YhiX. In response to changes in ambient pH the opportunistic pathogen Candida albicans differentially expresses a number of genes. The response to pH affects morphological differentiation and virulence. The pathway controlling the pH response terminates in the zinc-finger containing transcription factor encoded by RIM101/PRR2. By analogy to the pH response pathway of Aspergillus nidulans, PRR1 of C. albicans encodes a protein that is presumably required to convert Rim101p from an inactive to an active form by proteolytic removal of a C-terminal peptide. A prr1Delta mutant is compromised in its ability to differentiate into the filamentous form. Spontaneous phenotypic revertants of a prr1Delta mutant were selected by their ability to form filamentous colonies. These mutants were also found to be defective in pH-dependent gene expression.
