Crude glycerol has been widely investigated as a renewable carbon source for biodiesel production. In the present study, this feedstock was supplemented by various inducers: surfactants and oils to enhance lipid and lipase production by the newly isolated yeast, Yarrowia lipolytica. Results have shown that a culture medium composed of olive oil could enhance lipase activity at 25 U/mL and lipid content up to 35% (w/w). The fortification of the medium with crustacean waste increased the lipase activity up to 38 U/mL. Different parameters such as incubation time, inoculum level, initial moisture content, carbon level and nitrogen level of the medium were optimized. Screening of various process variables has been accomplished with the help of Plackett-Burman design. The maximum lipase activity of 18.58 units per gram of dry fermented substrate (U/gds) was observed with the substrate of Palm Kernal cake in four days of fermentation.

Development of yeast strains that do not switch between yeast and hyphal growth is critical for the utilization of fungi in reproducible bioprocesses. In this work, we isolated five spontaneous Y. lipolytica mutants that grow only in the yeast phase and do not form hyphae when cultivated on solid agar or in liquid medium in flasks or during bioreactor cultivation (Fig. 1). Mitochondrial Complex I (CI; also known as NADH:ubiquinone oxidoreductase), the first protein complex of the respiratory chain, is involved in the oxidation of NADH and the initiation of electron transfer across the mitochondrial inner membrane into the mitochondrial electron transport chain. CI is a large L shaped complex consisting of a hydrophilic peripheral arm that protrudes into the matrix and a hydrophobic membrane arm embedded in the inner membrane. The matrix arm transfers electrons to ubiquinone across a chain of seven Fe-S clusters while the membrane arm pumps protons from the matrix to the intermembrane space providing up to 40% of the protons necessary for ATP production (Braun et al., 2014). Signal N1a is absent from the spectrum of the NADH-reduced V96P/N142M variant (Figure 4), indicating that cluster N1a cannot be reduced by NADH because its potential has shifted more negatively. For the cluster to be ,10% reduced at −0.37 V it must have a reduction potential below −0.43 V. Thus the two mutations have shifted the potential more in the intact E. coli enzyme than in the isolated E. coli subunit, but by an amount consistent with the double mutations in the isolated B. taurus and Y. lipolytica subunits. The difference is probably due to small changes to the cluster environment upon its incorporation into the complex. The alternative explanation, that the cluster is not present in the V96P/N142M variant, is highly unlikely to be correct.
