The sexual differentiation paradigm contends that the female pattern of the reproductive system is established by default because the male reproductive tracts (Wolffian ducts) in the female degenerate owing to a lack of androgen. Here, we discovered that female mouse embryos lacking Coup-tfII (chicken ovalbumin upstream promoter transcription factor II) in the Wolffian duct mesenchyme became intersex-possessing both female and male reproductive tracts. Retention of Wolffian ducts was not caused by ectopic androgen production or action. Instead, enhanced phosphorylated extracellular signal-regulated kinase signaling in Wolffian duct epithelium was responsible for the retention of male structures in an androgen-independent manner. We thus suggest that elimination of Wolffian ducts in female embryos is actively promoted by COUP-TFII, which suppresses a mesenchyme-epithelium cross-talk responsible for Wolffian duct maintenance. ...

Schematic presentation of the mice kidney development.. Top, Kidney is formed via reciprocal interactions between the Wolffian duct and the metanephric mesenchyme (MM), two precursor tissues derived from the intermediate mesoderm. MM-derived signals, mainly the glial-derived neurotrphic factor (GDNF), induce the budding of the ureteric bud (UB) from the Wolffian duct, into the MM. MM cells condense around the tips of the branching UB, forming the cap mesenchyme (CM).. Bottom, Sequential nephron development: The CM is a subset of cells that constitutes nephron progenitors. CM-derived cells form most of the nephron body (from glomerulus to distal tubule), while the UB-derived cells form the collecting ducts. The CM cells sequentially form the pretubular aggregate (PA), renal vesicle (RV), C-, and S-shaped bodies (CSB and SSB, respectively), and finally the mature nephron.. Proximal-distal patterning along the length of the nephron is evident as early as at the RV stage, at the gene expression ...
With the formation of the primary tubules and their glomeruli the growth of the Wolffian body is by no means complete. Two factors contribute to the further growth of this organ; first the lengthening out of the several portions of each tubule, the intensification of the primary curves and by the addition of new ones; second by the formation of secondary, tertiary and quaternary canals. I shall designate as secondary canals all tubules developing subsequent to the primary set. As the origin of the primary mesonephric tubules gave rise to several theories, we have likewise a number of different views regarding the origin of the secondary. (a) The first view-The secondary tubules and their glomeruli arise either by fission or buds from the primary set. Either of these processes may have as a starting point the wall of the Malpighian capsule or the tubule itself. Braun (77) found in reptiles and Spengel (76) in amphibia that the primary glomeruli are first divided by fissures which continue along ...

The urinary and reproductive organs are developed from the intermediate mesoderm. The permanent organs of the adult are preceded by a set of structures that are purely embryonic and that, with the exception of the ducts, disappear almost entirely before the end of fetal life. These embryonic structures are on either side: the pronephros, the mesonephros and the metanephros of the kidney, and the Wolffian and Müllerian ducts of the sex organ. The pronephros disappears very early; the structural elements of the mesonephros mostly degenerate, but the gonad is developed in their place, with which the Wolffian duct remains as the duct in males, and the Müllerian as that of the female. Some of the tubules of the mesonephros form part of the permanent kidney. ...
Three pairs of kidneys successively form within the intermediate mesoderm: pronephros, mesonephros and metanephros. The pronephros and mesonephros are transient structures, and only the metanephros remains as the functional kidney of the newborn. The metanephros is characterized by its branched collecting duct system and large number of nephrons (∼11,000 in the mouse and ∼1 million in humans).. The metanephros develops through a complex set of reciprocal interactions between the metanephric mesenchyme (MM) and the Wolffian duct (nephric duct), which is derived from the intermediate mesoderm. The MM secretes GDNF, which upon binding to the c-RET receptor, induces outgrowth of ureteric buds (UB) from the nephric duct. The UBs then invades the MM. Mesenchymal cells condense to form condensed (cap) mesenchyme (CM) around each UB, a subset of cells which give rise to nephron progenitors. The ureteric bud is thereupon induced to branch. The CM proximal to the UB tips condenses to form pretubular ...

May 27, · The chromosomal sex of the embryo is established at fertilization. However, 6 weeks elapse in humans before the first signs of sex differentiation are noticed. Sex differentiation involves a series of events whereby the sexually indifferent gonads and genitalia progressively acquire male or female characteristics. Differences in development are dependent on a protein product of the Y chromosome SRY gene. The paired mesonephric ducts(Wolffian ducts) and paramesonephric ducts(Müllerian ducts) contribute the majority of male and female internal genital tract respectively.. How is sex differentiation in human embryos stem in Gladstone-Tannum Sands
In women, during embryologic development, the paired Müllerian (paramesonephric) ducts fuse distally and develop into the uterus, cervix, and upper vagina. If the Wolffian ducts persist in vestigial form, they can lead to Gartners cysts, mainly located in the right wall of the vagina. This is one of the few studies of Gartners cysts with a series of consecutive cases over a long period of time who were exclusively subject to clinical observation. Although Gartners cysts are found in approximately 0.1 to 0.2 % of women, controversy exists regarding the course of action to be taken. We describe the cases of four women who were 38-years old, 53-years old, 37-years old, and 49-years old at their first appointment and who were of mixed ethnicity, mixed ethnicity, black, and mixed ethnicity respectively. The follow-up of these patients ranged from 2 to 17 years. In these four cases the location of the cysts was the right wall of the vagina. Transvaginal ultrasound was the test of choice for diagnostic
organogenesis of the kidney 1. ORGANOGENESIS OF THE KIDNEY Wolffian duct In the outer part of the intermediate mesoderm, immediately under the ectoderm, in the region from the fifth cervical segment to the third thoracic segment, a series of short evaginations from each segment grows dorsally and extends caudally, fusing successively from before backward to form the pronephric duct.
Today, all 186 of the organs or structures present on Wiedersheim s list are known to have one or more specific uses or functions. The first group are those organs which have been incorrectly identified as useless but are now known to have a specific function such as the pineal gland, the pituitary gland and the lachrymal glands. The second group are those organs which are small and have only limited roles such as the wisdom teeth, the small toes and certain veins. The third group are those organs or structures which function only during certain stages in life such as the notochord, the posterior cardinal veins and the ducts of Cuvier. The fourth group are those organs which are developmental remnants of the reproductive structures of the opposite sex such as male nipples, male Mullerian ducts and female Wolffian ducts. These structures are not evolutionary remnants; rather they form prior to the sexual differentiation that occurs in the development of the human embryo ...
The acquisition of renogenic competence (the ability to give rise to kidney) during embryonic development is not yet fully understood. Clarifying the temporal and molecular aspects of this process is equally essential for understanding excretory system development and for devising methods for successful differentiation of embryonic stem cells (ESCs) to renal cells for disease modeling, toxicology screening and potential cell replacement therapies. In embryo development, the metanephric (permanent) kidney arises as a result of inductive interactions between two embryonic structures that arise in the intermediate mesoderm - the ureteric bud (UB, a diverticulum of the Wolffian duct) and the metanephric mesenchyme (MM). The UB develops into the collecting duct system and the MM undergoes an epithelial-to-mesenchymal transition to form the secretory units of the kidney - the nephrons. In this thesis, I used a tissue disaggregation-reaggregation method that allows the reconstruction of mouse ...
The major things I remembered about sexual differentiation in the fetus, from when I took development biology in grad school, are there are two pairs of tubes, the Müllerian and Wolffian duct, in all developing babies. For girls, one develops into the internal sex organs and the other recedes and for boys the opposite. Turns out that is a pretty good estimate of what happens. But lets look a little closer. The two duct pairs are part of urogenital development in the fetus and both extend passed the mesonephros (the primitive kidney) towards the caudal (tail) end of the organism, at the cloaca (define). The fetus remains gender neutral until about the 7 to 8 week, but one major thing has been in effect since way back when fertilization occurred. In terms of sex chromosomes, there are two an X and Y chromosome, each parent contributes one to the zygote to get a set. The mother can only contribute an X chromosome, since women are XX, but daddy can contribute either the X (this gives a daughter) ...

Cell migration is essential for many morphogenetic events in multicellular organisms. Yet much remains unknown about the mechanisms that allow stationary cells to become motile, to follow the migratory pathways appropriate to them and ultimately to stop migration at the specified destination. We have investigated migration of the axolotl pronephric duct (PND), a model system for elucidating the molecular mechanisms that control directed cell migration during embryogenesis. The PND (also termed the Wolffian, mesonephric or nephric duct) is the central component in the development of the vertebrate kidney. In addition to forming the waste conduit of the pronephros and mesonephros, the amniote PND generates the ureteric bud of the metanephros, which will undergo further differentiation to form the ureter - the structure that transports urine to the bladder. The PND also gives rise to the ductus deferens - the major sperm conduit of the male reproductive system - and participates in the induction of ...

The separation of the ureter from the Wolffian duct has commenced. In Fig. 388, C a still later stage is shown. Tubules now begin to form in the nephrogenic cap and collecting tubules to bud out from the pelvic bud. Collecting tubules arise by the division and redivision of the growing end of the pelvic outgrowths. In the third month the process of outgrowth from the ureteric bud continues ; the growing end of each bud divides and redivides, and in this manner the collecting tubules of the pyramids are formed (Fig. 389, C). In Fig. 389, A, the growing extremity of such a collecting duct is represented. Near one of its terminal buds is represented one of the numerous tubule-rudiments, formed from the nephrogenic tissue surrounding the ureteric outgroA^ths. Like a Wolffian tubule, it appears in a vesicular form. At one extremity it establishes a communication with the collecting tubule ; at the other a glomerulus develops (Fig. 389, B). The tubule elongates, becomes convoluted, and quickly assumes ...
The sex of a mammal is determined by the combination of sex chromosomes it has. If the mammal has two X chromosomes, it will likely become female, and if it has an X chromosome and a Y chromosome, it will likely become male. The story physiologists have been telling for decades is that there is a single gene located on the Y chromosome, called the SRY gene, that single-handedly makes an embryo become a male. When expressed, the SRY gene produces a protein, called testes-determining factor, which interacts with the cells of the undifferentiated gonads to turn them into testicular cells. These newly formed testicular cells produce two key hormones: testosterone, which causes the Wolffian ducts to become the epididymis, vas deferens, and seminal vesicles, and anti-Müllerian hormone (AMH), which causes the Müllerian ducts to degenerate. In other words, if an animal has a Y chromosome, it will typically have an SRY gene that will trigger the sequence of events that causes the animal to develop into ...

Hoxa 11 is a murine Abdominal-B-type homeobox gene. The structure of this gene is presented, including genomic and cDNA sequence. The cDNA includes the complete open reading frame and based on primer extension results is near full length. Surprisingly, the antisense strand of Hoxa 11 was found to be transcribed. Moreover, these antisense transcripts were processed and polyadenylated. The developmental expression patterns for both sense and antisense transcripts were examined using serial section and whole-mount in situ hybridizations. Hoxa 11 transcription patterns were defined in the limbs, kidney and stromal cells surrounding the Mullerian and Wolffian ducts. Of particular interest, in the developing limbs, the sense and antisense transcripts showed complementary expression patterns, with antisense RNAs increasing in abundance in regions where sense RNAs were diminishing in abundance. Furthermore, targeted mutation of Hoxa 11 is shown to result in both male and female sterility. The female ...

By contrast to the pronephros, the histological features of the mesonephros, with its primitive glomeruli, suggest that it probably functions as a primitive kidney, and is involved in the production of much of the amniotic fluid. Within the two mesonephroi, one located on either side of the dorsal mesentery of the hindgut, a substantial number (in the region of about 40 or more) of cranio-caudally segmented mesonephric tubules are formed. It has, however, been suggested that only the most rostrally located 4-6 pairs of mesonephric tubules drain into the mesonephric portion of the nephric duct. This is now seen to extend along the length of the mesonephroi, being located towards their lateral sides. The mesonephros is also retained over a considerably longer period than the pronephros, but gradually undergoes regression in a cranio-caudal direction. While the rostral part displays clear evidence of regression its more caudal part appears to display evidence of functional activity. Within the ...

1). Origin of the Antithesis. Rationalism connotes in philosophy the tendency of thought that lays special stress, not on the matter of experience, but on the products of the human reason, whether these consist of innate ideas or a priori concepts. The opposite principle is empiricism, which makes knowledge simply the reproduction of observed facts in their unity. In theology the term rationalism was first applied to criticism of church doctrine as practised by the Socinians and later by the deists. The real point of its application, however, is the stricter, scholastic form of the theological enlightenment which was assumed in Germany in dependence upon the Wolffian and Kantian philosophies. Rationalism unites itself organically with a universal movement of emancipation from ecclesiastical authority, partly in progress beforehand, and partly contemporaneous, in France and England, but assuming its characteristic type from certain philosophical schools and the German formative environment as a ...

Lethality: Ras signaling promotes the excretory duct cell fate, and mosaic analysis suggested that loss of this one specific cell can account for the zygotic lethality of let-60 null mutants (Yochem et al., 1997). The excretory duct cell is required for osmoregulation (Nelson and Riddle, 1984). let-60 loss-of-function mutants, which lack the excretory duct cell, die as rod-like larvae with a fluid-filled appearance (Figure 2). let-60 gain-of-function mutants sometimes have two excretory duct cells (Yochem et al., 1997). Ras signaling may have other (maternally-rescued) essential roles in addition to excretory duct formation, since mutations in let-23/EGFR, egl-15/FGFR and ptp-2/SHP-2 cause distinct scrawny and lethal defects that are rescued by constitutive forms of LET-60 Ras (DeVore et al., 1995; Gutch et al., 1998; Koga and Ohshima, 1995). Hyperactivation of egl-15/FGFR or let-60/Ras can also lead to lethality with a Clear, fluid-filled appearance (Kokel et al., 1998; Schutzman et al., ...

The kidney plays an essential role during excretion of metabolic waste products, maintenance of key homeostasis components such as ion concentrations and hormone levels. It influences the blood pressure, composition and volume. The kidney tubule system is composed of two distinct cell populations: the nephrons forming the filtering units and the collecting duct system derived from the ureteric bud. Nephrons are composed of glomeruli that filter the blood to the Bowmans capsule and tubular structures that reabsorb and concentrate primary urine. The collecting duct is a Wolffian duct-derived epithelial tube that concentrates and collects urine and transfers it via the renal pelvis into the bladder. The mammalian kidney function depends on the coordinated development of specific cell types within a precise architectural framework. Due to the availability of modern analysis techniques, the kidney has become a model organ defining the paradigm to study organogenesis. As kidney diseases are a problem

My research program focuses on urogenital system development (e.g. kidneys, ureters, prostate) and on the different cancers affecting this system (kidney and prostate). In the urinary system, we are studying how the first renal cell appear in the embryo, induced from progenitor cells (or stem cells). We are trying to understand how these cells organize themselves to generate the different tissues forming the urogenital system. The terms are used interchangeably. PTCs originate from the mesothelium and are presumed to be remnants of the Müllerian duct and Wolffian duct. PTCs have been reported in all female age groups and seem to be most common in the third to fifth decades of life. A study in Italy estimated their incidence to be about 3%, while an autopsy study of postmenopausal women detected them in about 4% of cases. These cysts constitute about 10% of adnexal masses. Most cysts are small and asymptomatic. Typical sizes reported are 1 to 8 cm in diameter. PTCs may be found at surgery or during an imaging examination that is performed for another reason. Larger lesions may reach 20 or more cm in diameter and become symptomatic exerting pressure and pain symptoms in the lower abdomen. Large cysts can lead to torsion of the adnexa inflicting ...
The epoophoron or epoöphoron (also called organ of Rosenmüller or the parovarium) is a remnant of the mesonephric tubules that can be found next to the ovary and fallopian tube. It may contain 10-15 transverse small ducts or tubules that lead to the Gartners duct (also longitudinal duct of epoophoron) that represents the caudal remnant of the mesonephric duct and passes through the broad ligament and the lateral wall of the cervix and vagina. The epoophoron is a homologue to the epididymis in the male. While the epoophoron is located in the lateral portion of the mesosalpinx and mesovarium, the paroophoron (residual remnant of that part of the mesonephric duct that forms the paradidymis in the male) lies more medially in the mesosalpinx. It has a unique histological profile. Clinically the organ may give rise to a local paraovarian cyst or adenoma.
