We report eight new mutants in Arabidopsis thaliana possessing altered leaf morphology and epicuticular wax. These were isolated from a T-DNA-mutagenized population using a visual screen for altered leaf reflectance, i.e. increased glaucousness or glossiness. The mutants were placed into three distinct classes based on alterations in overall plant morphology: knobhead (knb), bicentifolia (bcf), and wax. The four knb mutants formed callus-like growths in the axillary region of the rosette leaves and apical meristem, the two bcf mutants produced hundreds of narrow leaves, and the two wax mutants had leaves and stems that were more glossy than wild type and organs that fused during early development. Twenty-two plant protection products and veterinary substances of which residues have already been detected in beeswax in Europe were selected. The potential chronic exposure was assessed applying a worst-case scenario based on the addition of a maximum daily intake through the consumption of honey and beeswax to the theoretical maximum daily intake through other foodstuffs. For each residue, the total exposure was finally compared to the acceptable daily intake. To test the pH I did use pH paper but I stirred the finished product into distilled water and then tested that. I was told that is the proper way to test the pH of something. Since I posed my question to you yesterday, I re-tested the pH using this method and got a pH of 7ish-8ish. I read too, that using borax and beeswax results in a saponification process and I attributed that process to the slightly higher pH that I got. So, that is why I decided to use citric acid to lower the pH and since then tried lactic acid 88% as well. For instance; when I used 5 gtts of lactic acid in 15 mls of my test emulsion i saw no change in the lotion but when I added 7 gtts to an equal amount of the same test-emulsion, it resulted in immediate separation. Expression of alcohol-forming fatty acyl-CoA reductases in E. coli can result in the biosynthesis of fatty alcohols from endogenous E. coli fatty acids, but the levels were quite low [19]. To improve the production of fatty alcohols, Steen et al. carried out a further genetic modification of E. coli, and achieved an increased titer (~60 mg/L) of the medium chain fatty alcohols (C12 or C14 alcohols) [3]. In their strategy, they employed thioesterases with different substrate specificities to tailor the composition of the FFAs, and used the aldehyde-forming fatty acyl-CoA reductase Acr1 for the conversion of fatty acyl-CoAs to fatty aldehydes. The synthesized fatty aldehydes can be further converted to fatty alcohols by an unknown alcohol dehydrogenase/aldehyde reductase of E. coli [6]..
