von Willebrand disease type 2B (vWD-type 2B) is characterized by gain-of-function mutations in von Willebrand factor (vWF) that enhance its binding to the glycoprotein Ib-IX-V complex on platelets. Patients with vWD-type 2B have a bleeding tendency that is linked to loss of vWF multimers and/or thrombocytopenia. In this study, we uncovered evidence that platelet dysfunction is a third possible mechanism for bleeding tendency. We found that platelet aggregation, secretion, and spreading were diminished due to inhibition of integrin αIIbβ3 in platelets from mice expressing a vWD-type 2B-associated vWF (vWF/p.V1316M), platelets from a patient with the same mutation, and control platelets pretreated with recombinant vWF/p.V1316M. Impaired platelet function coincided with reduced thrombus growth. Further, αIIbβ3 activation and activation of the small GTPase Rap1 were impaired by vWF/p.V1316M following exposure to platelet agonists (thrombin, ADP, or convulxin). Conversely, thrombin- or ...

In Gaucher disease type I bleeding is a common presenting symptom, that may manifest itself as frequent nose bleeds, easy bruising but can also cause substantial bleeding after surgical or dental procedures and may occur in association with pregnancy or delivery . The bleeding tendency is usually considered to be secondary to thrombocytopenia However 50,000 platelets are enough in healthy people to give a normal bleeding time but are associated with significant bleeding tendencies in Gaucher patients. Bleeding tendency might be attributed by genetic inherited or Gaucher related coagulation factors abnormalities which in some cases stabilize with ERT. However, In other cases the etiology is an abnormality of platelet function. This thrombocytopathy has not been delineated and apart from a few aggregation studies, no systematic analysis has been published that convincingly shows the cause of the disturbed function. While, experience shows that enzyme replacement (ERT, i.e: imiglucerase, ...

BALTIMORE -- Ryan Zimmerman endured another poor outing at the plate on Saturday night, raising the possibility that his ailing right shoulder might need another cortisone shot -- or, perhaps, a second stint on the disabled list.. Following a 3-1 win over the Orioles, manager Davey Johnson admitted that while Zimmerman isnt the type of player to speak up when something is physically wrong, he is concerned about his veteran third baseman. That quickly prompted questions as to whether Zimmerman, who wound up on the disabled list on April 27 with shoulder inflammation, could end up back on the DL.. Theres no question about it, Johnson said. I think possibly even getting another injection in the shoulder. Im that concerned.. Saturday night, Zimmerman went 0-for-4, dropping his batting average to .218. Though its still early -- and he did hit .289 last year while missing 61 games -- Zimmerman hasnt had a multihit game since June 8, and for the month hes batting .167 with one home run and ...
The 911 dispatcher who spoke to Zimmerman on the fatal night didnt tell him to stay in his car. Zimmerman said he was following a suspicious person, and the dispatcher told him, We dont need you to do that. Chief prosecutor Bernie de la Rionda conceded in his closing argument that these words were ambiguous. De la Rionda also acknowledged, based on witness and forensic evidence, that both men were scraping and rolling and fighting out there. He pointed out that the wounds, blood evidence, and DNA didnt match Zimmermans story of being thoroughly restrained and pummeled throughout the fight. But the evidence didnt fit the portrait of Martin as a sweet-tempered child, either. And the notion that Zimmerman hunted down Martin to accost him made no sense. Zimmerman knew the police were on the way. They arrived only a minute or so after the gunshot. The fight happened in a public area surrounded by townhouses at close range. It was hardly the place or time to start shooting ...
The Zimmerman case began with the media making up a racist story: a black kid carrying candy is shot in cold blood by a white man in a gated community. It was a story that sent fear and anger through black people across the country. President Obama was in a low point in his re-election campaign, trying to gin up the black vote. Zimmerman, as a scapegoat, was used as a chance for Obama to bond with fellow blacks. So, making history, the president of the United States intervened in a criminal justice matter to foment race-hatred and fear. President Obama himself made it an issue of skin color: If I had a son, he would look like Trayvon Martin.. Once the president of the United States weighed in, Zimmerman had a target on his back. An ounce of election advantage to our privileged, Ivy League president versus the ruination of a Hispanic mans life -- it was an easy choice for Obama. Obamas great appeal to voters in 2008 was his self-presentation as a black man without animus or grievance, eager ...

Also, the more I think about it, Zimmerman has no claim to Stand your Ground, its Trayvon that has the right to it, so even IF Zimmerman was hit by Trayvon (which we now KNOW Zimmermans first and second set of versions of his story are totally false with the police station videos being released)…Trayvon would have had the RIGHT to Stand HIS Ground. Trayvon was doing nothing illegal and was less than a block away from where he was going back to where he was a guest.. Personally, I cant wait to see the neighborhood closed caption video tapes (as the front of the gated community signs state they have), for sure it had to have caught the majority of this, especially since it was not yet DARK AS NIGHT even IF raining at that time, NOR was Trayvon walking on, or where he died anything but the UBLIC WALKWAYS thru the town homes, and again, was murdered just down the block from where he was going, which again, since the reports came out of exactly where his body was found, Zimmermans supporters ...
With the announcement of new details about events leading up to the shooting death of teenager Trayvon Martin by neighborhood-watch volunteer George Zimmerman, at least three things are clear: we likely will never know exactly what happened that evening of Feb. 26 in Sanford, Florida; the situation couldve been avoided had Zimmerman followed the 911 operators instructions and stayed in his vehicle; Martin and Zimmerman were both flawed human beings like the rest of us.. Yesterday, as prosecutors released more than 200 pages of photos and eyewitness accounts showing Zimmerman had wounds to his face and the back of his head, the national debate about the case (which consistently appears to be divided sharply along racial lines), was reignited. While supporters of Zimmerman and his claim of self-defense see the new evidence as proof of his innocence, others view it as a mixed bag that doesnt necessarily bolster his defense. The lead detective in the case against Zimmerman said he believes ...

were, I did not know if he (Trayvon Martin) was armed or not. IMO this statement is a direct admission by Zimmerman that he should not eligible for release under the self-defense provisions of the stand your ground law. How could he be in fear for his life if he did not know if Trayvon was armed or not? I submit that Zimmerman had NO FEAR of Trayvon Martin and went after Trayvon and when he caught up with Trayvon attempted to physically attempted to secure him. This immediately initiated a scuffle as from Trayvons persective he was being attacked by some stranger in the dark of night. At this time Trayvon was the one who was actually in fear for his life and most likely in desperation struck out at Zimmerman in an attempt to free himself, hitting Zimmerman in the nose, knocking him to the ground. When Zimmerman hit the ground he pulled out his pistol and shot Trayvon. I submit that at no time was Zimmerman in fear for his life and he simply shot Trayvon in retaliation for being knocked to ...

Judge Debra Nelson ruled that the methods used by the experts arent reliable. But her ruling doesnt prevent the 911 calls from being played at trial.. She reached the decision after hearing arguments that stretched over several days this month on whether to allow testimony from two prosecution experts. One expert ruled out Zimmerman as the screamer and another said it was Martin. Defense experts argued there was not enough audio to determine who the screams are coming from. Zimmermans attorneys also argued that the state experts analysis is flawed.. Opening statements are set for Monday in the second-degree murder trial for the former neighborhood watch volunteer who says he fired on the black teenager in self-defense last year. Zimmerman is pleading not guilty. The elimination of the audio experts will likely shorten the trial by a week. Before the ruling, attorneys had predicted the trial could last two to four weeks after opening statements.. A spokeswoman for prosecutors didnt ...

Pre-mRNA splicing and cell cycle control have independent functions for eukaryotic cells. However, these processes might be linked, because yeast proteins have been identified through genetic criteria as splicing factors and independently as essential for cell cycle progression (10). The conserved Myb-related CDC5/Cef1p proteins carry essential functions in both processes, because their genetic depletion in the fission (30) and budding (11) yeasts causes accumulation of pre-mRNAs and cell cycle arrest at G2/M. Through characterization of a novel temperature-sensitive allele of S. cerevisiae CEF1, cef1-13, we have demonstrated that G2/M cell cycle arrest results from inefficient splicing of the TUB1 pre-mRNA that encodes the major yeast α-tubulin. TUB1 intron deletion also partially suppresses the G2/M arrest observed in mutants of the splicing proteins Prp17p and Prp22p.. cef1-13 cells arrest homogeneously in G2/M prior to the first encountered anaphase when released to the restrictive ...
Trayvon Martin was a seventeen-year-old black teenager. On the evening of February 26, 2012, he was visiting with his father and his fathers fiancée in the Sanford, Florida multi-ethnic gated community where his fathers fiancée lived. Trayvon left her home on foot to buy a snack from a nearby convenience store. As he was returning, George Zimmerman, a white Hispanic male and the communitys neighborhood watch program coordinator, noticed him. In light of a recent rash of break-ins, Zimmerman called the police to report a person acting suspiciously, which he had done on many other occasions. The 911 operator told Zimmerman not to follow the teen, but soon after Zimmerman and Martin had a physical confrontation. According to Zimmerman, Martin attacked him, and in the ensuing scuffle Martin was shot and killed (CNN Library 2014).. A public outcry followed Martins death. There were allegations of racial profiling-the use by law enforcement of race alone to determine whether to stop and detain ...

Shellie Zimmerman 911 Call: He is in his car and he continually has his hand on his gun and he keeps saying step closer and he is just threatening all of us. And he is going to shoot us. He punched my dad on the nose. My dad has a mark on his face. I saw his glasses were on the floor. He accosted my father and then took my iPad out of my hand and smashed it and cut it with a pocket knife. There is a Lake Mary city worker across the street that I believe saw almost all of it. He is sitting in his truck right now. He just showed up here but my phone died so I had to call from my fathers phone. Im really, really afraid. I dont know what he is capable of. Im just really scared. Dad do you need medical? . He says no, but I think he does need medical. He is shaken. He says he feels like he is going to have a heart attack. His nose. Yes you do because your nose looks like it could be broken. I think he should have a medical. If we could have an ambulance come ...

George was going to the store, and he saw someone in his community that he did not recognize as living there. Because there had been a lot of break-ins in the area, he thought that was suspicious that someone would not be walking on the street or the sidewalk, but theyd be walking right behind the townhomes. ¶Robert Zimmerman said that his son, George Zimmerman, 28, then called a non-emergency number to report this stranger. He lost sight of the teen as he looked for an address to report to police. ¶Its my understanding that at that point Trayvon Martin walked up to him, asked him, Do you have a - beep - problem? George said No, I dont have a problem. ¶The elder Zimmerman said that as his son started to reach for his cell phone, Martin, 17, punched him in the nose, knocking the larger man to the pavement. ¶Trayvon Martin got on top of him and just started beating him, in the face, in his nose, hitting his head on the concrete, Robert Zimmerman said. ¶Thats when Martin spotted ...

Where have I said hang him? Where have I said Mr Zimmerman has no right to a fair trial? Fact is, I have NOT said that. It is my belief that self defense is not an option in this case based on the fact that Mr Zimmerman followed Martin. When you are following someone, you are not on the defensive. If you run from someone, as Mr Martin did, you are on the defensive. Zimmerman should be charged and stand trial. Let a jury hear all the evidence and then decide. How that opinion is a threat to freedom and liberty is very amusing to me ...
The initiation and evolution of myelodysplastic syndrome (MDS) and acute myeloid leukemia (AML) are driven by genomic events that disrupt multiple genes controlling hematopoiesis. Human genetic studies have discovered germline mutations in single genes that instigate familial MDS/AML. The best understood of these genes encode transcription factors, such as GATA-2, RUNX1, ETV6, and C/EBPα, which establish and maintain genetic networks governing the genesis and function of blood stem and progenitor cells. Many questions remain unanswered regarding how genes and circuits within these networks function in physiology and disease and whether network integrity is exquisitely sensitive to or efficiently buffered from perturbations. In familial MDS/AML, mutations change the coding sequence of a gene to generate a mutant protein with altered activity or introduce frameshifts or stop codons or disrupt regulatory elements to alter protein expression. Each mutation has the potential to exert quantitatively ...
My two primary research areas of interest are (i) understanding the structure-function of splicing factor U2AF-RNA interactions during translation and how its regulation controls gene expression and, (ii) how the cancer associated splicing factor mutations alter RNA fate leading to control of gene expression and their role in cancer biology.

The way I explain this to people is to imagine a petite woman about to be raped by a larger, stronger man. Clearly, if she is armed, you would not deny her the right to defend herself. You would not demand that she show the gun and attempt to get the man to stop raping her. No, as soon as she is threatened, she should be allowed to defend herself. As soon as Martin was a clear threat to Zimmermans life, Zimmerman is allowed to defend himself. The legal question is if Zimmerman had a legitimate reason to think he was in danger. I think it is very, very easy to prove he was. No one needs to submit to a few more punches or a few more head slams, just to see if they can resolve this without having to get violent. ...

Let me preface my remarks with a heartfelt congratulations that your wife made it through and best wishes for her continued good health.. Now…. 1. What are the breast cancer survival rates in other countries? What were they here a decade or two ago? If theyve improved (pretty sure that they have), to what do we owe that?. 2. It think that the key word in your statement is survived. I presume that, like most cancer victims, your wife endured a long and expensive course of treatment. Who paid for that? For most people, its an insurance company. In other words, people in the United States tend to pay a fraction of the actual cost of their health care, even in cases where the prognosis is bad and / or the treatment is extremely expensive.. My father has had three heart attacks plus various operations for such things as pacemaker installation and replacement. This did not bankrupt my parents; they did not lose their house, have to cash in their retirement, or anything else as they had (and ...
