In anesthetized dogs the intravenous infusion of arginine vasotocin, 25 mU/min, or vasopressin, 10 mU/min, inhibited renin secretion when control secretion rates were normal or when they were elevated by complete ureteral occlusion. Vasotocin, 5 mU/min, or vasopressin, 1 mU/min, was ineffective. Direct intraarterial infusion of vasotocin, 5 mU/min, into the right kidney inhibited renin secretion by the right kidney only. Glomerular filtration rate and renal plasma flow were not changed, and mean arterial pressure was either not changed or was decreased. One of the most established roles of oxytocin (OT) is in inducing uterine contractions and labour. Apart from contractions, our recent study have shown that OT can also activate pro-inflammatory pathways in both human myometrial and amnion cells which suggests the pro-inflammatory role of OT should be taken into account when developing tocolytics targeting the OT/OT receptor (OT/OTR) system. The OTR antagonist, Atosiban, is currently used therapeutically for the treatment of PTL. We have previously shown that Atosiban fails to inhibit the pro-inflammatory effects of OT in human amnion, and Atosiban alone activates NF-κB and MAPKs thus upregulating downstream pro-labour genes. We tested whether the sex pheromones that stimulate courtship clasping in male roughskin newts do so, at least in part, by amplifying the somatosensory signals that directly trigger the motor pattern associated with clasping and, if so, whether that amplification is dependent on endogenous vasotocin (VT). Female olfactory stimuli increased the number of action potentials recorded in the medulla of males in response to tactile stimulation of the cloaca, which triggers the clasp motor reflex, as well as to tactile stimulation of the snout and hindlimb. That enhancement was blocked by exposing the medulla to a V1a receptor antagonist before pheromone exposure. However, the antagonist did not affect medullary responses to tactile stimuli in the absence of pheromone exposure, suggesting that pheromones amplify somatosensory signals by inducing endogenous VT release.
