The aim was to identify the shortest NPY/PYY fragments that are capable of suppressing the contractions. 2. NPY (1-36) and C-terminal fragments of NPY (from 11-36 to 22-36) suppressed the electrically-evoked twitches in a concentration-dependent manner. Multiple redundant steps are routinely taken in order to ensure discontinuity of the vas deferens and prevent recanalization. Figure 1 illustrates how the vasal occlusion methods for vasectomy are performed. Careful examination of the ligation and fascial interposition method in this figure shows that the suture placed to do the fascial interposition forms a pursestring around the vas deferens itself. In practice, this will likely be tied too loosely and allow the vasal end to slip back inside the sheath, or tied too tightly and strangulate the vasal end, with both resulting in lack of fascial interposition. I suspect this accounts for the extraordinarily high rate of failure with this method. No vasectomist should accept a 16.7 percent failure rate. Smooth muscle from newborn guinea-pig vas deferens was enzymically dispersed into single cells or small clumps and grown in culture in the presence or absence of sympathetic ganglion explants.. Most single smooth muscle cells gradually lost their typical ultrastructural features and contractile properties during the first few days in culture. At 7 days of culture these dedifferentiated smooth muscle cells underwent extensive proliferation. If sufficient cells were present in the culture inoculate, a continuous monolayer formed at about 9 days of culture and redifferentiation of smooth muscle began. At 11-12 days of culture the cells reaggregated into clumps, began to contract spontaneously, and formed into well-organized muscle bundles in two layers at right angles, resembling the muscle layer organization of the in vivo vas deferens. A disposable vasectomy instrument is provided having a body portion and an insertable module. The body portion includes a jaw portion having a lower fixed jaw and an upper movable jaw into which the module is inserted. The module includes a cutting block having two spaced apart blades and a pair of ligature clips disposed on either side of the blades. The movable jaw is controlled by the operator to simultaneously move the blades forward and close the ligature clips. The physician isolates the vas deferense, engages it within the ligature clips and adjacent the blades. The movable jaw is operated, cutting out a segment of the vas deferens and closing the ligature clips on each severed end of the vas deferens. A separate module is provided for each vas deferens of the patient.

To investigate the biology of the male genital duct epithelium, we have established cell cultures from the ovine vas deferens and epididymis epithelium. These cells develop tight junctions, high transepithelial electrical resistance, and a lumen-negative transepithelial potential difference as a sign of active transepithelial ion transport. In epididymis cultures the equivalent short-circuit current (I(sc)) averaged 20.8 ± 0.7 μA/cm 2 (n = 150) and was partially inhibited by apical application of amiloride with an inhibitor concentration of 0.64 μM. In vas deferens cultures, I(sc) averaged 14.4 ± 1.1 μA/cm 2 (n = 18) and was also inhibited by apical application of amiloride with a half-maximal inhibitor concentration (K(i)) of 0.68 μM. The remaining amiloride-insensitive I(sc) component in epididymis and vas deferens cells was partially inhibited by apical application of the Cl - channel blocker diphenylamine-2-carboxylic acid (1 mM).
