In this report, we show that Cox-2 is essential for optimal humoral immune responses against HPV type 16. These new findings reveal that Cox-2 deficiency impairs neutralizing Ab production following HPV 16 VLP vaccination. These data have important implications regarding the use of NSAIDs and Cox-2-selective inhibitors during vaccination. Drugs that inhibit Cox-2 are widely used for relief of symptoms of pain and inflammation associated with vaccination and other inflammatory conditions. Our findings reveal that the combination of reduced class switch recombination, B cell differentiation to ASCs and Ig secretion are responsible for impaired Ab production by HPV 16 VLP-vaccinated Cox-2−/− mice. Genetic material (either RNA or DNA) is contained within the core of these virus particles. Papillomaviruses (PVs) infect epithelial cells and are the main causative agent of cervical carcinoma. There are today more than a hundred different PV types and these can be divided into high risk and low risk types. They infect differentiating epithelial cells which make it cumbersome to propagate and produce human papillomavirus (HPV) virions. A common method to study HPV infection is to use HPV virus like particles (VLPs) produced in recombinant eukaryotic expression systems. Glycosaminoglycans (GAGs) have been described as an initial attachment receptor for several viruses. Our aim was to study the interactions between HPV VLPs and different GAGs to determine how these could affect binding and internalization. We found that soluble heparin was the best GAG inhibitor of HPV-16 VLP binding followed by heparan sulfate of mucosal origin. The focus of this research was to compare the binding profiles of human papillomavirus (HPV) 11, 16, 18 and 45 virus-like particles (VLPs) to HaCaT cells and to the extracellular matrix (ECM) secreted by these cells. All four HPV types tested bind to a component(s) of the ECM. HPV11 VLP binding is blocked when the ECM is pretreated with an anti-laminin 5 (LN5) polyclonal antibody. A series of treatments utilizing heparins and heparinase revealed that HPV18 VLPs are dependent on heparan sulfates (HS) for binding to cells and ECM. HPV16 and HPV45 VLPs are dependent on HS for binding to HaCaT cells and dependent on both HS and LN5 for binding to ECM. These studies emphasize the need to study the binding characteristics of different HPV types before applying universal binding principles to all papillomaviruses.
