Novel tick-borne phleboviruses in the Phenuiviridae family, which are highly pathogenic in humans and all closely related to Uukuniemi virus (UUKV), have recently emerged on different continents. How phleboviruses assemble, bud, and exit cells remains largely elusive. Here, we performed high-resolution, label-free mass spectrometry analysis of UUKV immuno-precipitated from cell lysates and identified 39 cellular partners interacting with the viral envelope glycoproteins. The importance of these host factors for UUKV infection was validated by silencing each host factor by RNA interference. This revealed Golgi-specific brefeldin A-resistance guanine nucleotide exchange factor 1 (GBF1), a guanine nucleotide exchange factor resident in the Golgi, as a critical host factor required for the UUKV life cycle. Bunyaviridae is a large family of viruses that have gained attention as emerging viruses because many members cause serious disease in humans, with an increasing number of outbreaks. These negative-strand RNA viruses possess a membrane envelope covered by glycoproteins. The virions are pleiomorphic and thus have not been amenable to structural characterization using common techniques that involve averaging of electron microscopic images. Here, we determined the three-dimensional structure of a member of the Bunyaviridae family by using electron cryotomography. The genome, incorporated as a complex with the nucleoprotein inside the virions, was seen as a thread-like structure partially interacting with the viral membrane. Fibrillin microfibrils are extracellular matrix assemblies that provide the connective tissues of metazoan species with many of their biomechanical properties. They are also involved in regulating the production of extracellular matrix through their interactions with growth factors such as transforming growth factor-β. The process of microfibril assembly and its regulation are poorly understood. Jensen et al have investigated the role of the conserved C-terminal propeptide of fibrillin-1 using an in vitro microfibril assay in which HEK293T cells, transiently expressing a GFP-tagged variant of fibrillin-1, are co-cultured with fibroblasts to produce a recombinant microfibril network.
