The Nanion Vesicle Prep Pro is a stand-alone workstation for the preparation of giant unilamellar vesicles (GUVs). To form the GUVs, the principle of electro-swelling is used. Lipid is spread on the glass substrate of the vesicle chamber, so that a thin layer of lipid is formed on the substrate. With the application of an alternating electrical field, the lipid layer releases from the electrode and the vesicles are formed. As transparent ITO Slides are used as electrodes the whole process can be observed with a microscope. The electrodes, as well as the liquid reservoir, are held in the Nanion GUV Chamber, while the electrical field is generated by the Vesicle Prep Pro© station. In particular, the present invention relates to a medical device having a tubular membrane structure over a radially expandable structural frame, and to a method of forming the tubular membrane on the radially expandable structural frame. In one aspect, a structural frame is placed over a spinning mandrel and a fiber is electro-statically spun over at least a portion of the structural frame forming a membrane. A transfer sheath may be used between the mandrel and structural frame to prevent the electro-statically spun fiber from adhering to the mandrel. Stability of small unilamellar liposomes in serum and clearance from the circulation: the effect of the phospholipid and cholesterol components
We use fluorescence microscopy to directly observe liquid phases in giant unilamellar vesicles. We find that a long list of ternary mixtures of high melting temperature (saturated) lipids, low melting temperature (usually unsaturated) lipids, and cholesterol produce liquid domains. For one model mixture in particular, DPPC/DOPC/Chol, we have mapped phase boundaries for the full ternary system. For this mixture we observe two coexisting liquid phases over a wide range of lipid composition and temperature, with one phase rich in the unsaturated lipid and the other rich in the saturated lipid and cholesterol. We find a simple relationship between chain melting temperature and miscibility transition temperature that holds for both phosphatidylcholine and sphingomyelin lipids. We experimentally cross miscibility boundaries both by changing temperature and by the depletion of cholesterol with beta-cyclodextrin. We present optical observations of phase separation in mixed model membranes in the form of giant unilamellar vesicles. These observations are compared to the phase behavior of lipid mixtures, which we determined by X-ray scattering and differential scanning calorimetry or extracted from the existing literature. A microwave technique is demonstrated to measure floating giant unilamellar vesicle (GUV) membranes in a 25 μm wide and 18.8 μm high microfluidic channel. The measurement is conducted at 2.7 and 7.9 GHz, at which a split-ring resonator (SRR) operates at odd modes. A 500 nm wide and 100 μm long SRR split gap is used to scan GUVs that are slightly larger than 25 μm in diameter. The smaller fluidic channel induces flattened GUV membrane sections, which make close contact with the SRR gap surface. Small unilamellar vesicles (SUVs) of phospholipids are often used as a membrane model system for studying the interaction of molecules. When using NMR under the standard liquid-state conditions, SUV phospholipid proton spectra can be recorded, exhibiting sharp signals. This is not only because of the fast vesicular tumbling but also because of the combination of this tumbling with the individual motion of the lipids inside the bilayer. This appears evident because addition of cholesterol is responsible of broader resonances because of the slowing down of the lipid motion. On the other hand, no (1)H signal is detected for cholesterol in the bilayer. This lack of detection of the inserted molecules explains why generally SUVs are not considered as a good model for NMR studies under the standard liquid-state conditions. For the 15 years or so, we have developed a series of reconstitution platforms to investigate the molecular mechanism of SNARE-mediated membrane fusion, and the regulation of this process by regulatory factors. In 1998, we established the central function of the SNARE proteins as fusogens when we reconstituted these proteins into small unilamellar vesicle (SUV) and measured lipid mixing between liposomes containing cognate SNARE proteins.. We have functionalised microscope slides with BODIPY-based molecular rotor capable of viscosity sensing via its fluorescence lifetime.
