Cutting edge research leads to commercialisation of ultrafiltration membrane technology. The launch of the South Africas first locally produced ultrafiltration membrane technology will take place on the 29th October 2009 in Somerset West. Most membranes used in South Africa have been imported at great cost. With this in mind, a joint research project, funded by the Water Research Commission (WRC), was initiated in 1996 and succeeded to develop suitable and cost-effective, locally produced, ultrafiltration membranes and filtration systems for use in water treatment and industrial water management. This study aimed to develop improved cleaning strategies for controlling biofilms on the surfaces of membranes used in dairy ultrafiltration (UF) and reverse osmosis (RO) plants. Eleven UF / RO membrane modules from 7 different New Zealand dairy membrane processing plants were received after typical cleaning-in-place (CIP) procedures. Microorganisms were isolated from both the retentate and permeate sides of these membrane surfaces and from the liquids collected from a UF membrane plant. Also some foulants scraped from a RO membrane were tested. The routine CIP currently used in the dairy plants was not adequate to completely remove organic material, including microbial cells, proteins and carbohydrates from the membrane surfaces. ENGLISH ABSTRACT: The aim of the thesis was to investigate the use of flow destabilization methods, combined with permeate backflushing (BIF) or on their own, on flux recovery and maintenance in capillary UF membrane systems under cross-flow (XF) and dead-end (DE) operating conditions. Various hydraulic and mechanical methods have been used to remove the accumulated cake layer and improve steady state process flux. Permeate backflushing (B/F) is the most widely used but the drawbacks are loss of product and extensive down-time. In a pilot plant study for ultrafiltration of surface waters containing high NOM, turbidity and cation loads, the use of flow destabilization, or feed flow reversal (FFR) combined with cross-flow B/F was able to improve the normalised flux by 10.7 ± 3.4 %, compared with 3.2 ± 1.6 % improvement for BIF without FFR. Broth-culture filtrates of Campylobacter pylori induced non-lethal cytopathic effects in vitro in 7 of 9 mammalian cell lines tested. Transmission electronmicroscopy revealed that the response consisted of intracellular vacuolisation. Intestine 407 cells were among the most responsive and were used for routine assay. About 55% of isolates of C. pylori tested, originating from four geographic regions worldwide, produced cytotoxic activity. The activity was neutralisable by specific antisera to broth-culture filtrates or to sonicated bacteria but not by antisera to other bacterial preparations. Cytotoxic activity was heat-labile (70°C for 30 min), was protease-sensitive and ammonium-sulphate precipitable. It did not pass through an ultrafiltration membrane with a nominal mol.-wt limit of 100 x 103. It was concluded that C. pylori can produce a factor that alters cultured cells in vitro. Antioxidant activity, as determined by the oxygen radical absorbance capacity (ORAC) assay, was low for EYP and PPP hydrolysates with values of 613.1 and 489.2 µM TE×g−1 protein, respectively. UF-fractionation of EYP hydrolysate increased slightly the antioxidant activity in permeate fractions (720.5-867.8 µM TE×g−1 protein). Lignin, which is the second most abundant biomass component and has carbon-rich phenolic content, is a promising renewable raw material for multiple applications, such as carbon fibers, adhesives, and emulsifiers. To use lignin efficiently, it is important to ensure its purity and homogeneity. As a result, the separation of lignin into fractions with high purity and narrow molecular-weight distributions is likely a prerequisite for several applications.
