I choosed the following schema: 1. dig labeld mRNA probe of gene1---------mouse anti-dig(Roche)---------Goat anti mouse with HRP(Molecular Biology)------Tyramide Signal Amplification (Molecular Biology) 2. biotin labeld mRNA probe of gene2------------strepavidin-HRP (Jacson Immuno)------Tyramide Signal Amplification (Molecular Biology) The first one (dig) gave me nice staining, but the second one (strepavidin) always gave high background. I know the biotin labeled probe is good, since when I use this probe tested with mouse-anti-bio antibody, then followed by Goat-anti-mouse-HRP second antibody, it works beautifully. To clarify the biologic and clinical role of p53 alterations in HNSCC, we have examined the prevalence and the nature of p53 alterations in a large cohort of tumors from the different sites. For immunohistochemical analysis of p53 protein expression, we introduced tyramide signal amplification immunohistochemistry (TSA-IHC) on a tissue microarray. This allowed the discrimination between normal low-level expression and reduced or lost expression. Two hundred fifty-three tumors were subjected to mutational analysis by genomic DNA sequencing, employing also the p53 GeneChip from Affymetrix. The prevalence of all p53 alterations, i.e., mutations, overexpression and loss of expression, was significantly higher in hypopharyngeal tumors than in the other sites (p = 0.001). Biogenic amines (BAs) are organic amines present in meat, fish, dairy produce and wine due to the breakdown of amino acids, catalysed by microbial decarboxylases. BAs determination in food is important not only because of possible toxicological effects such as nausea, sweating and headache but also due to their possible role as indicators of food spoilage. Chromatographic methods are traditionally applied for determination of BAs in food [1], which usually require preliminary operations for sample pre-treatment that are laborious and difficult to automate. Absorption isnt so easy. I can think of some interesting interactions. For example if taking an MAO inhibitor (e.g., clorgyline) then the ingestion of sympathomimetic amines (e.g. amphetamine or even tyramine from cheese) will increase their [amines] apparent absorption as there is no longer functional MAO in the gut epithelium to degrade it. This can lead to the cheese effect which results in severe hypertension, flushing and can be fatal ...
