Trichuris trichiura is a form of nematode roundworm / whipworm which present in the intestine. Trichuris trichiura can be detected by stool examination which may reveal the present of barrel shaped eggs with plug at each end.
Whipworms (Trichuris sp) are successful gastrointestinal dwelling nematode parasites of many species including man. The human parasite Trichuris trichiura is currently thought to infect up to 1 billion people worldwide resulting in considerable morbidity. Trichuris muris is very closely related to the human parasite T. trichiura sharing cross reactive antigens. Moreover, it is a remarkably tractable model system for dissecting immune responses and host parasite relationships and is actively being investigated in a number of laboratories worldwide. T. muris is a naturally occurring nematode parasite of mice which resides in the caecum and colon and has a direct oral faecal life cycle. Embryonated eggs are ingested and hatch in the crypts of the large intestine. After undergoing a series of moults, they become adult parasites which mate and release eggs that are shed in the faeces and can develop externally. The adult worms have an unusual niche in that the anterior end burrows through epithelial ...
The whipworm of the dog is substantially smaller than the other worms and is rarely seen as it lives in the cecum. If whipworms are present embedding themselves in the large intestine tissue, tremendous inflammation can result leading to a bloody, gooey diarrhea. A second syndrome of infection has emerged but is not well understood, this being symptoms mimicking those of Addison?s disease. Species of whipworms that can infect cats: Trichuris serrata in North America and Trichuris campanula in Europe.
Methods, devices, kits and compositions for detecting the presence or absence of whipworm in a fecal sample are disclosed herein. The methods, devices, kits and compositions of the present invention may be used to confirm the presence or absence of whipworm in a fecal sample from a mammal that may also be infected with one or more of hookworm, roundworm, and heartworm. Confirmation of the presence or absence of whipworm in the mammal may be made, for example, for the purpose of selecting an optimal course of treating the mammal and/or for the purpose of determining whether the mammal has been rid of the infection after treatment has been initiated.

The gut microbiota (GM) displays a profound ability to adapt to extrinsic factors, such as gastrointestinal pathogens and/or dietary alterations. Parasitic worms (helminths) and host-associated GM share a long co-evolutionary relationship, exerting mutually modulatory effects which may impact the health of the host. Moreover, dietary components such as prebiotic fibers (e.g. inulin) are capable of modulating microbiota toward a composition often associated with a healthier gut function. The effect of helminth infection on the host microbiota is still equivocal, and it is also unclear how parasites and prebiotic dietary components interact to influence the microbiota and host health status. Some helminths, such as Trichuris suis (porcine whipworm), also exhibit strong immunomodulatory and anti-inflammatory effects. We therefore explored the effects of T. suis, alone and in interaction with inulin, both in fecal microbiota during the infection period and luminal microbiota across four intestinal segments
Whipworms tend to affect dogs, but it is not unnatural for cats to be infected by whipworms. Looking like small pieces of thread, whipworms tend to occupy your pets intestine. Whipworms do not lay many eggs, so several stool samples need to be examined to determine if whipworms are the potential culprit. Stool that is encased in mucus or extreme weight loss is usually attributed to whipworm infestation. Fortunately, whipworms hardly cause deaths.. Roundworms tend to affect puppies, especially puppies living in close quarters. Puppies that come from puppy mills and pet shops tend to be inundated with this parasite. Roundworms tend to be very common in puppies and kittens due to the way it is spread. The roundworm larva can migrate between the mother and the developing fetus. It can also be spread during nursing time. Once the larva migrates, they grow up to 5 inches in the dogs or cats intestine. There they start to absorb nutrients from the intestinal tract, then it will start laying eggs. ...
Effect of combinations of marketed human anthelmintic drugs against Trichuris muris in vitro and in vivo : Soil-transmitted helminth (STH) infections are responsible for a huge public health burden, however treatment options are limited. The discovery and development of novel efficacious drugs or drug combinations for the treatment of STH infections therefore has a high research priority. Human trichuriasis, caused by the gut-dwelling soil-transmitted helminth species Trichuris trichiura, represents a major public health problem in the developing world since an estimated 604-795 million people are infected worldwide[1]. T. trichiura infections are responsible for the loss of 349,000-1,061,000 disability-adjusted life years (DALYs) per year[2]. Particularly affected, school- and preschool-aged children are confronted with the largest morbidity burden[3]. Mild infections are often asymptomatic, however, in severe and chronic infections patients can suffer from dysentery, anemia and impairment of physical and mental development[3]. The drugs recommended by the World Health Organization[4], albendazole and mebendazole, have been in use for decades and display only moderate cure rates when administered as single doses[5-7]. Although this has not arisen as a problem yet, potential emergence of drug resistance is of concern[5, 6]. Thus, there is a need to find novel anthelmintic drugs ...

Human helminthic parasites were considered as a therapeutic option. Many could not be used because there are no available sources other than a human carrier. Eggs from such a source would risk inadvertent transmission of pathogenic microbial agents. Also, some human helminths have disease potential or raise public health concerns.. Trichuris species are helminths with favourable characteristics for therapeutic use. Their life cycle minimises the risk of inadvertent colonisation. Trichuris ova mature in the soil and are ingested by the host. Ova hatch in the duodenum, releasing larvae that ultimately grow in 6-8 weeks into adult worms. They migrate to the terminal ileum and colon but do not invade the host. Worms can remain viable for 1-2 years in the natural host. Adult worms release ova that are shed into the stool. These ova are immature and are not capable of colonising another host until they incubate in the soil for several weeks to allow embryonation.. We chose T suis as the helminth to ...

In fact, whipworm infection is actually the leading global cause of inflammatory bowel disease, and allergies have been linked to toxicara infestation in American cities, Hotez pointed out.. But Elliott argues that real-world examples indicate differently. South Korea recently launched a major helminth eradication program, and shortly thereafter, he said, Crohns disease appeared on the scene. Hotez counters, however, that an increase in inflammatory bowel diseases would also reflect massive changes in a variety of factors such as diet, lifestyle, and gut flora that also accompany development.. For villages in the middle of nowhere suffering as a result of hookworm infection, Elliott supports vaccine development. He points out, though, that individuals in need of vaccination also tend to have low intakes of protein and iron- two dietary deficiencies that lead to problems from otherwise relatively benign parasites.. Hotez, however, believes helminthic therapy is an unsafe practice whose ...
Interleukin (IL) 33, a member of the IL-1 family, is the ligand of ST2 that is expressed mainly on activated Th2 cells and mast cells. IL-33 can skew a predominantly Th1 cell population to a mainly Th2 cells phenotype in vivo. IL-33 messenger RNA is expressed early during infection of the intestinal-dwelling nematode Trichuris muris in mice. IL-33 treatment enhances resistance to Trichuris infection. IL-33 also effectively attenuates sepsis by mobilising the innate cells, neutrophils, to the site of infection, helping to clear the pathogens. Thus, IL-33 may be evolutionally preserved for the host defence against infections. IL-33 can reduce an ongoing atherosclerosis in ApoE−/− mice and attenuate adipocytes mainly by inducing the production of type II cytokines. In contrast, IL-33 can also exacerbate allergy and the inflammation in collagen-induced or serum-induced arthritis. Hence, IL-33 is a double-edged sword, and targeting IL-33 should be approached with caution.. ...
The authors collected fecal samples from hundreds of primates representing nine species that are sympatric in Kibale. With these samples in hand - and while wearing gloves I hope - the authors began their molecular investigation. First, they used microscopy to identify samples that were positive for Whipworm eggs. Next, using these Whipworm-positive samples, the authors designed a nested set of PCR primers to amplify short sequences of rRNA that were then Sanger sequenced. Interestingly, the sequencing revealed results that were hidden to the ...

Background: Intestinal epithelial cell (IEC) turnover has been implicated in the expulsion of and resistance to intestinal pathogens, including the murine whipworm Trichuris muris. Failure to initiate a transient increase in IEC turnover can lead to chronic infection. Over-expression of SOCS3 in vitro leads to decreased proliferation and reciprocally, lack of epithelial SOCS3 promotes proliferation and tumour growth in intestinal injury and cancer models. SOCS3 may have an important role in regulating the magnitude and/or duration of IEC turnover and consequently resistance to pathogens. This study aims to examine the ability of SOCS3 to regulate IEC turnover in an in vivo murine model of T. muris infection. Methods: SOCS3 expression was examined in the intestine of T. muris resistant or susceptible mice by immunohistochemistry and real-time PCR. Mice with and without IEC SOCS3 conditional knockdown were infected with T. muris and intestinal tissue examined at 3, 14, 21 and 35 days after ...

Identifying the cellular and molecular requirements for initiating and regulating type 2 immunity and inflammation is essential for the development of new vaccines against helminth parasites and treatments for atopic diseases. CD11c+ dendritic cells (DCs) are critical antigen-presenting cells (APCs) capable of priming and promoting the differentiation of naïve CD4+ T cells. However, the role of DCs in the initiation of Th2 cell differentiation following exposure to helminth parasites and allergens remains unclear. In Chapter 2, we examine the cellular requisites for initiating Th2 cytokine-dependent immunity and inflammation in the gastrointestinal tract utilizing infection with the helminth Trichuris muris. By genetic restriction of MHC class II expression to CD11c+ DCs we demonstrate that, in contrast to Th1-cytokine-mediated immunity, antigen presentation by CD11c+ cells is insufficient to generate protective type 2 immune responses in vivo, suggesting additional non-DC APC interactions may be

Background: In the murine model of whip worm infection Trichuris muris invades cecum and colonic epithelial cells and elicits either a Th1 (susceptible) or Th2 (resistant) immune response. An increase in intestinal epithelial cell (IEC) turnover has been implicated to aid in the physical expulsion of the worm in the resistant phenotype [1]. Failure to initiate a transient increase in IEC turnover can lead to chronic infection. SOCS3 limits IEC proliferation and its loss is associated with increased tumour growth and hyperproliferation in cancer and intestinal injury models [2]. SOCS3 may have an important role in regulating IEC in intestinal homeostasis and may mediate resistance to pathogens. This study aims to examine the role of SOCS3 in regulating IEC turnover in a murine model of T. muris infection. Methods: SOCS3 expression was examined in the intestine of T. muris resistant or susceptible mice by immunohistochemistry and real-time PCR. SOCS3 IEC (villin cre) knockdown (SOCS3fl/fl-VC) and ...

Author Summary Mixed infections with roundworm, whipworm and hookworm are common, but survey reports often give only the separate prevalence of each type. However, the combined prevalence is important to estimate accurately the number of individuals who would benefit from control programmes and to make decisions about the frequency of treatment. Previous work suggests that mixed infections involving hookworm occur randomly, but that roundworm and whipworm infections are found together more frequently than would be expected by chance. We used 63 data sets from community surveys that reported both the proportions infected with individual types of worms and the combined proportion infected with any worm. We then calculated the proportion that would be infected with any type of worm if infections had occurred randomly and compared it with the observed combined proportion infected. We found a strong correlation between the observed and predicted combined proportions infected. A small downward correction of

The idea is this: autoimmune disorders, in which the immune system, for varying reasons known and unknown, attacks a non-threatening substance or even body part, have risen in developed countries pretty much co-incident with the rise of widespread hygiene and the associated de-worming of our guts. According to Mental Floss, it took a dual-vocation gastrointologist and parasitologist and a long plane ride to notice this. As a gastrointologist, Dr. Joel Weinstock studied Chrohns Disease, in which the immune system attacks the intestines, leading to constant surgery and a generally very painful life outlook. He and his team decided to experiment with parasitic worms, to an astounding success rate. One example: in 2005 he infected 29 Crohns patients with Trichuris suis, a worm found in pigs and consequently, pig farmers, who tolerate them with little side effects. 23 patients showed improvements, 21 went into complete remission, according to a NY Times Article. Similar studies, by Dr. Weinstock ...

Most worms (nematodes) settle and grow in the small intestine, though some species are found in the cecum, heart, lung, and other tissues in various stages of development. The intestinal nematodes produce eggs, which are carried with the digestive products to exit in the feces. But since the egg-lay-ing does not always coincide with the dogs bowel movements, stool samples may not show the presence of worms. A 5?day sampling will probably reveal some eggs if hookworms or roundworms are present, but tapeworms or whipworms may still escape detection. For this reason, many breeders rely instead on outward signs of poor coat, flatu-lence and/or diarrhea, loss of weight, and an abnor-mal look or smell to the stool.. In many breeds, the topcoat should lie flat, straight, and smooth, giving a water?resistant thatch over the softer undercoat and skin. If these coarse, straight guard hairs stand up and out, or if the ends curl out away from the body, it may be a temporarily open condition due to worms. ...

The unembryonated eggs are passed with the stool. In the soil, the eggs develop into a 2-cell stage, an advanced cleavage stage, and then they embryonate; eggs become infective in 15 to 30 days. After ingestion (soil-contaminated hands or food), the eggs hatch in the small intestine, and release larvae that mature and establish themselves as adults in the colon. The adult worms (approximately 4 cm in length) live in the caecum and ascending colon. The adult worms are fixed in that location, with the anterior portions threaded into the mucosa. The females begin to oviposit 60 to 70 days after infection. Female worms in the caecum shed between 3,000 and 20,000 eggs per day. The life span of the adults is about 1 year.. ...
We demonstrate that both pathogens are capable of negatively affecting local and systemic immune responses towards each other by modifying cytokine phenotypes and by inducing general immune suppression. T. muris infection influenced non-specific and pathogen-specific immunity to M. bovis BCG by down-regulating pulmonary TH1 and Treg responses and inducing systemic TH2 responses. However, co-infection did not alter mycobacterial multiplication or dissemination and host pulmonary histopathology remained unaffected compared to BCG-only infected mice. Interestingly, prior M. bovis BCG infection significantly delayed helminth clearance and increased intestinal crypt cell proliferation in BALB/c mice. This was accompanied by a significant reduction in systemic helminth-specific TH1 and TH2 cytokine responses and significantly reduced local TH1 and TH2 responses in comparison to T. muris-only infected mice ...
Diagnosis of soil-transmitted helminths (STHs) in developing countries is commonly based on microscopic detection of eggs in stool samples, using the Kato-Katz (KK) method, which has a poor sensitivity for detecting light intensity infections. We compared the performance of the KK method and real-time PCR in the framework of a randomized trial, which evaluated four novel treatments against Trichuris trichiura and concomitant STH infections. Two stool samples obtained from 320 participants were examined at baseline and follow-up with quadruplicate KK and PCR analyses of one of the two samples using

Gastrointestinal helminth infections affect over 1 billion people worldwide and are a significant cause of human and livestock morbidity. With resistance to current treatments becoming increasingly problematic and with the lack of any vaccine, novel anthelmintics are urgently required [3]. In this study, it was demonstrated for the first time that RXR antagonists show anthelmintic activity and may therefore serve as important lead compounds in the search for new treatments for nematode infections. At L3, L4 and adult stages of the parasite, the human RXR antagonist HX531 was able to reduce both the motility and viability of T. muris larvae in vitro, as determined by a range of viability assays. The RXR agonist HX630 also reduced worm motility in the L3/L4 and mature adult only stages, however this was only observed in one viability assay. Earlier work has demonstrated that synthetic retinoids that are weak RAR agonists are able to interfere with the development of filarial nematodes in vitro[25, ...

There are five key parasites that pose a threat to your dogs health: fleas, heartworms, roundworms, hookworms (A. caninum) and whipworms. What people commonly refer to as worms are really internal parasites that live in the heart or intestinal tract of the host animal. Many internal parasites can be life-threatening to your dog, and most people dont realize that some of these parasites can be transferred from pets to humans, which is known as zoonosis. Children are especially susceptible to zoonotic infection because they are always grabbing, touching and sticking their hands in their mouths without regard to whether they are clean. People also dont realize just how prevalent internal parasites are. A national survey of veterinarians showed that more than one out of three dogs (36 percent) was infected by one or more of the three most common gastrointestinal nematodes - whipworms, roundworms and hookworms. Heartworms are also a serious year-round threat. More than 27 million dogs in the ...

Cats and dogs are the favorite nesting grounds of four principal groups of worms and a few species of microscopic protozoa. The four worms are roundworms, tapeworms, hookworms, and whipworms. Among the protozoa are coccidia, toxoplasma, giardia, and ameba.. Proper identification is vital. Unfortunately, in the case of parasites, identification isnt always easy because adult worms release their eggs sporadically. Knowing exactly what the problem is, is the first step in finding a solution.. Its very important to bring your pets fecal sample (bowel movement) to your veterinarian as often as requested up to one year of age. Collect fresh fecal sample within 12 hours of an examination. It is also very important to keep the samples cool or refrigerated. A microscopic examination of the fecal sample will be performed ...

Lead researcher, Dr Aaron Jex, Faculty of Veterinary Science, said, We know that humans infected with the harmless, pig whipworm can have significantly reduced symptoms linked to autoimmune diseases. And now we have the genetic sequence of the worm, it opens the door to future human drug designs and treatment.. Although the pig whipworm causes disease and losses in livestock, it does not cause disease in humans.. In contrast, the human whipworm infects around 1 billion people, mainly children in developing nations, and causes dysentery, malnourishment and impairment of physical and mental development.. Coauthor, Prof Robin Gasser, Faculty of Veterinary Science, said, The genes tells us about the proteins that this worm uses to interact with our immune systems. Knowing the worms molecular landscape could be very useful in starting to understand autoimmune diseases in humans.. The pig whipworm genome also provides a model to study the human parasite.. The study involved 11 institutions ...

To further back up our assumptions based on our own findings, there was one study of pathogen-related diarrhea 1 that showed 68% of tested shelter dogs with diarrhea were positive for giardia and 15% for coccidia. Of shelter cats with diarrhea, 11% tested positive for coccidia, but the majority tested negative for all viral and protozoal pathogens.. This study did not test for parasites such as roundworms, hookworms or whipworms which are very common in young animals, and even with negative fecal examinations and deworming, they should be considered highly possible to be present.. Statistical analysis of the causes of diarrhea is important because it shapes how a shelter can treat the group in the most efficient and cost effective way possible. If a large majority of the animals who have diarrhea are likely to be suffering from the same cause, then that treatment should be the first line of defense against diarrhea in all dogs and cats for the sake of efficiency and lifesaving. For most ...

A study of how the body expels parasitic worms has led to a surprising new discovery about the immune system that could help in the treatment of bowel cancer. Scientists investigating whipworms, parasites that infect one-fifth of the worlds population as well as livestock and domestic animals, have discovered a new way that the body effectively eliminates the parasites. The University of Manchester research, published in the US journal Science, found the reason why some hosts were able to expel the worms naturally without the need for treatment. This is a completely new way in which the immune system controls disease and may lead scientists to look at new ideas in the treatment of inflammatory bowel diseases and even bowel cancer, said Dr Laura Cliffe in the Faculty of Life Sciences, who carried out the research. During our investigations we discovered that the immune system does more than what it currently says in the text books it controls other physiological systems. The body ...
