We demonstrate trapping and characterization of multiple gold nanospheres with a setup composed of dark field imaging and optical tweezers. The number of trapped nanospheres is quantified by the overall dark-field scattering intensity. The spectra of the scattering intensity show that there is no interparticle coupling among trapped nanospheres when the density of nanospheres in the trap is low enough (less than 10 particles), while the density of nanosphere increases the interparticle coupling of nanospheres becomes obvious. Aim: To investigate the preparation, in vitro release, in vivo pharmacokinetics and tissue distribution of a novel polymeric micellar formulation of paclitaxel (PTX) with Pluronic P123. Methods: The polymeric micelles of paclitaxel with Pluronic P123 were prepared by a solid dispersion method. The characteristics of micelles including particle size distribution, morphology and in vitro release of PTX from micelles were carried out. PTX-loaded micellar solutions were administered through the tail vein to healthy Sprague-Dawley rats and Kunming strain mice to assess the pharmacokinetics and tissue distribution of PTX, respectively. Studies on a model ink suspension have shown that the effect of non-ionic dispersion agents on ink agglomeration using fatty acid chemistry as collector was highly temperature-dependent. The non-ionic surfactant reduced the agglomeration efficiency at low temperatures (below the cloud point of the surfactant) but this negative effect was reduced at increasing temperatures. At temperatures above the cloud point the non-ionic surfactant improved the agglomeration. The non-ionic surfactant adsorbs to both the ink and precipitated soap particles, which at low temperatures resulted in an increased colloidal stability of the particles but at higher temperatures led to a destabilisation of the particles due to the increased hydrophobicity of the surfactant. The surfactants clearly affected the precipitation of fatty acid anions to calcium soaps and at very high surfactant concentrations the formation of calcium soap particles was drastically reduced. Objective: In the current investigation, we aimed to study the combined cytotoxicity of curcumin, as a nanomicellar formulation, and galbanic acid (Gal), dissolved in DMSO against the murine C26 and human Caco-2 colon carcinoma cells. Further, curcumin potential for cisplatin and doxorubicin (Dox) co-therapy was studied. Materials and Methods: The combined cytotoxic effect of these phytochemicals at varying dose ratios were examined using the MTT colorimetric assay. Moreover, the time-dependent toxicity of curcumin, cisplatin, Dox, and pegylated liposomal Dox (Doxil) was determined. The interactive anti-proliferative behavior of these compounds was examined using the CompuSyn software. Results: Nanomicellar curcumin showed considerable cytotoxicity in C26 cells 24 hr post-treatment. The cloud points (CP) of 1 g/dl solutions of polyethylene oxide-polypropylene oxide (PEO-PPO) based triblock copolymers (Pluronics® P84, L64, L44 and Reverse Pluronics® 10R5, 25R4, 17R4) were measured as a function of their molecular weight and added ionic surfactant. For identical PEO/PPO ratios, copolymers with lower molecular weight show a larger increase in the cloud point in the presence of surfactants than polymers with higher molecular weight. The opposite trend has been observed for reverse Pluronics. The cloud points of polymers with different PEO/PPO ratios have also been reported. Polystyrene (PS)/multi-wall carbon nanotube (MWCNT) nanocomposites were prepared by melt mixing and by coagulation of PS/MWCNT sonicated dispersion. Before mixing and coagulation particles of MWCNT were covered with PS nanospheres by microemulsion polymerization to increase their dispersibility and compatibility with the matrix. Microemulsion polymerization was carried out under sonication in the presence of surfactants and a cross-linking agent. The structure of modified MWCNT filler was revealed by SEM and TEM microscopy analyses. They show that PS is well grafted onto surface of MWCNT in form of ca 30 nm nanospheres or as ca 3-8 nm PS film layer. Significant changes in properties were recorded mainly for the coagulated sample. Although no shift of T-g was recorded for the melt mixed composite, the coagulated sample shows an increase by 9 degrees C. We report our theoretical investigation onto the shape dependence of two-photon absorption (TPA) in semiconductor nanocrystals (NCs). Based on a four-band model under effective mass approximation, we have developed a simple analytical theory capable of providing a quantitative explanation of the recent TPA measurement on CdS nanorods [Appl. Phys. Lett. 94, 103117 (2009)]. With this theory, we have systematically revealed the characteristics of TPA in CdSe and ZnO NCs with four different shapes: sphere, cube, cylinder and cuboid. Due to the splitting of degenerate energy levels caused by the decreased degree of symmetry, nanocuboids and nanocubes exhibit greater TPA cross-sections than nanocylinders and nanospheres of similar sizes, respectively. Similarly, nanocuboids and nanocylinders possess larger TPA cross-sections than nanocubes and nanospheres of similar lateral dimension, respectively. This study was aimed at preparing, characterising and evaluating in situ gel formulations based on a blend of two hydrophilic polymers i.e. poloxamer 407 (P407) and poloxamer 188 (P188) for a sustained ocular delivery of ketorolac tromethamine (KT). Drug-polymer interaction studies were performed using {DSC} and FT-IR. The gelation temperature (Tsol-gel), gelation time, rheological behaviour, mucoadhesive characteristics of these gels, transcorneal permeation and ocular irritation as well as toxicity was investigated. {DSC} and FT-IR studies revealed that there may be electrostatic interactions between the drug and the polymers used. {P188} modified the Tsol/gel of {P407} bringing it close to eye temperature (35°C) compared with the formulation containing {P407} alone. Nitric oxide (NO) is involved in physiological processes, including vasodilatation, wound healing and antibacterial activities. As NO is a free radical, designing drugs to generate therapeutic amounts of NO in controlled spatial and time manners is still a challenge. In this study, the NO donor S-nitrosoglutathione (GSNO) was incorporated into the thermoresponsive Pluronic F-127 (PL) - chitosan (CS) hydrogel, with an easy and economically feasible methodology. CS is a polysaccharide with known antimicrobial properties. Scanning electron microscopy, rheology and differential scanning calorimetry techniques were used for hydrogel characterization. The results demonstrated that the hydrogel has a smooth surface, thermoresponsive behavior and good mechanical stability. The kinetics of NO release and GSNO diffusion from GSNO-containing PL/CS hydrogel demonstrated a sustained NO/GSNO release, in concentrations suitable for biomedical applications. The research presented in this dissertation involves the synthesis, modification, characterization, and the application of polymeric surfactants in micellar electrokinetic chromatography (MEKC). Sodium undecenoyl-L-Leucinate (L-SUL) was synthesized and modified by use of alkali metal counterions, alcohols, and Triton X-102. Characterization of the modified poly-L-SUL was performed in order to elucidate the structural changes on the surfactants by use of four techniques. While surface tensiometry was used for the determination of the critical micelle concentration (cmc), fluorescence measurements were used for the determination of the polarity of the modified polymeric surfactants. The information about the size of the polymeric surfactants was obtained by use of pulsed field gradient-NMR (PFG-NMR).
