Obesity is a worldwide pandemic that also contributes to the increased incidence of other diseases such as type 2 diabetes. Increased obesity is generally ascribed to positive energy balance. However, recent findings suggest that exposure to endocrine-disrupting chemicals such as obesogens during critical windows of development, may play an important role in the current obesity trends. Several experimental approaches, from in vitro cell cultures to transgenerational in vivo studies, are used to better understand the mechanisms of action of obesogens, each of which contributes to answer different questions. OBJECTIVES--To study the effect of bis (tributyl tin) oxide (TBTO) on permeability of the blood-brain barrier. METHODS--Electron microscopy and an x ray microanalyser with lanthanum chloride as a tracer were used, and blood tin concentrations were determined with an atomic absorption spectrophotometer. Adult male wistar rats received 0.05 ml/kg body weight of TBTO orally. RESULTS--A transient increase in paracellular permeability at the blood-brain barrier was found 2 h after the dose of TBTO. Electron dense lanthanum deposits penetrated tight junctions of the endothelia and permeated the subendothelial space. The x ray microprobe data showed an accumulation of TBTO at the tight junctions at 2 h. Leakage of tracer did not occur at 4 h, but oedematous changes in the surrounding glial cells were prominent between 4 and 8 h and had almost returned to normal by 24 h. Therefore, screening for tributyltin (TBT) resistant and degrading bacteria is relevant for selection of isolates with decontamination ability of TBT polluted areas. With this rationale, 105 strains were isolated from sediment and surface waters of Vishakapatnam shipping harbor and their tolerance to TBT was evaluated. Further screening was done based on the ability of bacteria to grow in Minerals Medium (MM) containing 3 mM TBT as sole source of carbon. Ten selective TBT resistant isolates showed cross tolerance to six heavy metals and ten antibiotics. Among them, one interesting isolate VBAK101 showing highest TBT resistance (4 mM) and maximum growth yield (A600 nm of 1.6 after 36 h) was selected for further study.
