Several FY 2015 toxicogenomic studies used NextGen sequencing technologies, which provided improvements to gene expression analysis, including base-pair level resolution of accuracy and increased sensitivity compared to microarray platforms. While micorarrays are a stable and well understood technology for assaying gene expression, NextGen sequencing methods like RNA-Seq should become more common as sequencing costs drop, and bioinformatic pipelines become standardized and integrated with genomic sequencing. A promising area of research is the use of exome sequencing (Exome-Seq) that can be applied to either frozen or formalin fixed, paraffin embedded (FFPE) tissues. The validation of in vitro skeletal muscle models may play a pivotal role in capturing safety endpoints early within the research and development process. Therefore, the primary aim of this project was to investigate the extent of translation from an in vivo rat model to an in vitro skeletal muscle model, using a toxicogenomics approach. To this end, the mechanisms of toxicity of three novel sulfonyl isoxazoline (SI) herbicides (two triazoles and one phenyl) developed by Syngenta were investigated in vivo and in vitro. In vivo histopathology studies identified striated muscle as the target organ of SI triazole toxicity, and the stomach and liver as the target organs of SI phenyl toxicity. Tell me a bit about your research.. So its about these filled systems, I would mix various fillers with some different chemicals to form a composite. A composite is a hybrid material whose properties differ from the individual materials from which it is formed. Concrete is an example of a composite since its different materials mixed. I was seeing how they would affect a process called frontal polymerization. The idea is that when we add these fillers were studying how they can affect the chemical aspects of the reaction and also the thermal aspects of it. The system is driven by heat so in theory if you add a system that is thermos-conductive, it should increase the speed of the process. However, we found out that some of them didnt have any effect and it decreased it, so, I studied it more and found out that there were some chemical effects and that these fillers can interact chemically with the reaction and can actually slow the reaction down. When reading about endocrine disruptors, I keep coming across the work of Dr. Tyrone Hayes, a professor in the Department of Integrative Biology, at the University of California-Berkeley. He has studied the effects on frogs of an agricultural pesticide called Atrazine. Hayes found that these chemicals, even at very low levels, were causing male frogs in the wild to develop eggs in their testes (!).. I came across a lecture Dr. Hayes gave called From Silent Spring to Silent Night. The lecture is about an hour long, but its worth watching (see below). In the aftermath of the large spill of crude 4-methylcyclohexylmethanol (MCHM) into the Elk River in 2014, there were reports of a strong licorice-smelling odor and reports of respiratory illness/cough suggestive of chemical-induced adverse airway effects. Levels of MCHM in the air were predicted to be as high as 0.39 ppm after 10 minutes of showering, suggesting that inhalation of MCHM vapors was a potential route of chemical exposure. My laboratory has studied the toxicological effects of tobacco products and tobacco-related disease for approximately 20 years. My laboratory has focused recently on new tobacco products including harm reduction cigarettes, Snus, dissolvables, and electronic cigarettes, and we are one of the few labs in the United States to have studied electronic cigarettes. Diesel fuels derived from different types of crude oil can exhibit different chemistry while still meeting market requirements and specifications. Oil sands derived fuels typically contain a larger proportion of cycloparaffinic compounds, which result from the cracking and hydrotreating of bitumens in the crude. In the current study, 17 refinery streams consisting of finished fuels and process streams were obtained from a refinery using 100% oil sands derived crude oil. All samples except one met the ULSD standard of 15 ppm sulfur. The samples were characterized for properties and chemistry and run in a simple premixed HCCI engine using intake heating for combustion phasing control. Results indicate that the streams could be equally well characterized by chemistry or properties, and some simple correlations are presented.
