N2 - The adipose tissue includes various stromal cells, such as preadipocytes, endothelial cells, fibroblasts, and immune cells, which are involved in adipose tissue functions. We previously reported that, in obese mice, the sodium-glucose cotransporter 2 inhibitor ipragliflozin (Ipra) promoted the expansion of the epididymal adipose tissue (Epi) with increase of serum ketone body concentration. The Ipra-induced adipose tissue expansion did not deteriorate adipose inflammation, or systemic glucose/lipid metabolism, referred to as healthy adipose tissue expansion. Here we found that Ipra promoted healthy adipose tissue expansion with a reduced ratio of pro-inflammatory M1-like adipose ...

TY - JOUR. T1 - Macrophage elastase suppresses white adipose tissue expansion with cigarette smoking. AU - Tsuji, Takao. AU - Kelly, Neil J.. AU - Takahashi, Saeko. AU - Leme, Adriana S.. AU - Houghton, A. Mc Garry. AU - Shapiro, Steven D.. PY - 2014/12/1. Y1 - 2014/12/1. N2 - Macrophage elastase (MMP12) is a key mediator of cigarette smoke (CS)-induced emphysema, yet its role in other smoking related pathologies remains unclear. The weight suppressing effects of smoking are a major hindrance to cessation efforts, and MMP12 is known to suppress the vascularization on which adipose tissue growth depends by catalyzing the formation of antiangiogenic peptides endostatin and angiostatin. The goal of this study was to determine the role of MMP12 in adipose tissue growth and smoking-related suppression of weight gain. Whole body weights and white adipose depots from wild-type and Mmp12-deficient mice were collected during early postnatal development and after chronic CS exposure. Adipose tissue ...
Liver X receptors (LXR) are deemed as potential drug targets for atherosclerosis, whereas a role in adipose tissue expansion and its relation to insulin sensitivity remains unclear. To assess the metabolic effects of LXR activation, C57BL/6 mice on a high-fat diet (HFD) were treated with the dual LXRα/β agonist T0901317 (30 mg/kg per day) for 3 weeks. Differentiated 3T3-L1 was used for analysing the effect of T0901317 on glucose uptake.T0901317 reduced fat mass, accompanied by a massive fatty liver and lower adipokine levels in circulation of HFD mice. Increased adipocyte apoptosis and macrophage infiltration were found in epididymal fat of T0901317-treated HFD mice. In addition, T0901317 treatment promoted basal lipolysis, but blunted the anti-lipolytic action of insulin. Furthermore, LXR activation antagonized PPARγ target genes in epididymal fat and PPARγ-PPRE binding activity in 3T3-L1 adipocytes. Although the glucose tolerance was comparable to that in vehicle-treated HFD
Women seeking breast reconstruction after a mastectomy often have to undergo a time-consuming, uncomfortable process to prepare their skin.
Accumulation of visceral adipose tissue correlates with elevated inflammation and increased risk of metabolic diseases. However, little is known about the molecular mechanisms that control its pathological expansion. Transcription factor interferon regulatory factor 5 (IRF5) has been implicated in polarizing macrophages towards an inflammatory phenotype. Here we demonstrate that mice lacking Irf5, when placed on a high-fat diet, show no difference in the growth of their epididymal white adipose tissue (epiWAT) but they show expansion of their subcutaneous white adipose tissue, as compared to wild-type (WT) mice on the same diet. EpiWAT from Irf5-deficient mice is marked by accumulation of alternatively activated macrophages, higher collagen deposition that restricts adipocyte size, and enhanced insulin sensitivity compared to epiWAT from WT mice. In obese individuals, IRF5 expression is negatively associated with insulin sensitivity and collagen deposition in visceral adipose tissue. Genome-wide
Expandable devices and methods for treating and enlarging a tissue, an organ or a cavity. The device is composed of a hollow expanding pouch made of a resorbable material or a perforated material that can be attached to a filling element. The pouch can be filled with a biocompatible materials, one or more times in few days interval, after the insertion of the device. While filling the pouch every few days the tissue expands and the filling material if it is bioactive start to function. Overfeeding experiments, in which we impose short-term positive energy balance, help unravel the cellular, physiological and behavioural adaptations to nutrient excess. These studies mimic longer-term mismatched energy expenditure and intake. There is considerable inter-individual heterogeneity in the magnitude of weight gain when exposed to similar relative caloric excess reflecting variable activation of compensatory adaptive mechanisms. Significantly, given similar relative weight gain, individuals may be protected from/predisposed to metabolic complications (insulin resistance, dyslipidaemia, hypertension), non-alcoholic fatty liver disease and cardiovascular disease. Similar mechanistic considerations underpinning the heterogeneity of overfeeding responses are pertinent in understanding emerging metabolic phenotypes, for example, metabolically unhealthy normal weight and metabolically healthy obesity. Intrinsic and extrinsic factors modulate individuals overfeeding response: intrinsic factors

The almost universal finding of a dilated RV with reduced ejection fraction has been taken as a manifestation of the inadequacy of the RV to perform as the systemic ventricle after the Mustard operation. Furthermore, this dysfunction has been implicated in the similarly universal finding of reduced exercise capacity in these often asymptomatic patients. There can be no doubt that RV function may be abnormal, but the impact of the RV dilatation and reduced myocardial shortening on functional performance is far from clear. These features may merely reflect a normal response of the systemic RV to its afterload, and it is no surprise that there is little7 or no18 direct relationship between these indexes and exercise performance. In this study, we confirmed a failure to maintain an increased stroke volume as the primary hemodynamic abnormality at peak exercise at a time when it would be expected to be ≈40% higher than resting values.19 Although cardiac output rose, the HR response was ...
Purpose: : High-hydrophilic osmotic self-inflating hydrogel expanders (osmed GmbH Ilmenau, Germany) are well-accepted for implantation to achieve tissue expansion in defined parts of the body like skin, breast and orbital soft tissue. The suitability of this hydrogel consisting of a co-polymer of N-vinyl-pyrolidone and methyl-methacrylate as a drug delivery system for antibiotics was investigated in a laboratory setting. Methods: : The dry expanders were incubated in a 0.3% solution of Ofloxacin or Tobramycin for 24 hours. The complete swollen expander had increased in volume from 0.3ml to almost 3ml (adsorbing 2.7 ml of the 0.3% solution, i.e. 8.1 mg Ofloxacin or Tobramycin, respectively). Addressing the elimination of both antibiotics, the concentrations in 15 ml elution medium (simulating the volume of the orbit in a newborn) were measured after 0.25, 1, 2, 6, 24, 48 and 72 hours of eluation. 0.9% sodium chloride (B.Braun Melsungen, Germany) was used as elution medium. To imitate fluid ...
When it comes to burn patients, particularly children, recovery is ongoing because skin cant grow and change as quickly as the body develops. Skin is a vital organ and is the first line of sensation and defense against external factors. But when skin is severely burned, it loses much of that function and requires reconstructive surgeries, rehabilitation, tissue expansion, pressure garments and ongoing check ups. Its because of this extensive and time consuming treatment that Shriners has been one of the first hospital networks in the country to employ telemedicine practices. ...

The manner in which white adipose tissue (WAT) expands and remodels directly impacts the risk of developing metabolic syndrome in obesity. Preferential accumulation of visceral WAT is associated with increased risk for insulin resistance, whereas subcutaneous WAT expansion is protective. Moreover, pathologic WAT remodeling, typically characterized by adipocyte hypertrophy, chronic inflammation, and fibrosis, is associated with insulin resistance. Healthy WAT expansion, observed in the metabolically healthy obese, is generally associated with the presence of smaller and more numerous adipocytes, along with lower degrees of inflammation and fibrosis. Here, we highlight recent human and rodent studies that support the notion that the ability to recruit new fat cells through adipogenesis is a critical determinant of healthy adipose tissue distribution and remodeling in obesity. Furthermore, we discuss recent advances in our understanding of the identity of tissue-resident progenitor populations in ...

Within reproductive genetics, the genetic causes of male infertility is a first topic of interest. To elucidate genetic causes of male infertility, genes with a testis-specific expression pattern will be examined and the whole genome will be investigated for the presence of deletions/duplications. Treatment of male infertility is a logical complement of the former topic. A specific subpopulation of male infertile patients are survivors of childhood cancers who underwent aggressive treatment including chemo- and radiotherapy and bone marrow transplantation. Studies to investigate the genetic and epigenetic aspects of spermatogonial stem cell (SSC) transplantation as well as cryopreservation and tissue expansion are mandatory before a clinical application becomes feasible ...
During development, multicellular organisms undergo stereotypical patterns of tissue growth to yield organs of highly reproducible sizes and shapes. How this process is orchestrated remains unclear. Analysis of the temporal dynamics of tissue growth in the Drosophila abdomen reveals that cell cycle times are spatially correlated and that growth termination occurs through the rapid emergence of a population of arrested cells rather than a gradual slowing down of cell cycle time. Reduction in apical tension associated with tissue crowding has been proposed as a developmental growth termination mechanism. Surprisingly, we find that growth arrest in the abdomen occurs while apical tension increases, showing that in this tissue a reduction in tension does not underlie the mechanism of growth arrest. However, remodeling of the extracellular matrix is necessary for tissue expansion. Thus, changes in the tissue microenvironment, and a rapid exit from proliferation, control the formation of the adult ...
Obesity originates from an imbalance between caloric intake and energy expenditure that promotes adipose tissue expansion, which is necessary to buffer nutrient excess. Patients with higher visceral fat mass are at a higher risk of developing severe complications such as type 2 diabetes and cardiovascular and liver diseases. However, increased fat mass does not fully explain obesitys propensity to promote metabolic diseases. With chronic obesity, adipose tissue undergoes major remodeling, which can ultimately result in unresolved chronic inflammation leading to fibrosis accumulation. These features drive local tissue damage and initiate and/or maintain multiorgan dysfunction. Here, we review the current understanding of adipose tissue remodeling with a focus on obesity-induced adipose tissue fibrosis and its relevance to clinical manifestations.. ...
Adipose tissue expansion is known to occur through proliferation and differentiation of preadipocytes; however, the mechanisms especially responsible for preadipocyte proliferation are incompletely understood (4). Numerous data suggest that only cleaved DLK1 inhibits adipogenesis by interfering with the differentiation machinery (8), but our results invoke an additional explanation suggesting that DLK1M slows preadipocyte proliferation by regulating numerous components in the G1-S phase of the cell cycle.. As such, we have examined the effect on preadipocyte proliferation of cleavable and noncleavable DLK1 isoforms by specifically reducing the endogenous level of specific DLK1 variants in cells that naturally express the protein. Although one study previously used antisense RNA molecules to study DLK1s function in adipocyte differentiation (7), the role of DLK1 has only been assessed by over-expressing the protein in cell types lacking endogenous Dlk1 (13,41) or expressing different endogenous ...
Apparatus for the treatment of volume deficiency disorders of body structures and related syndromes and, more particularly, apparatus that deliver fluid via a catheter at a controlled pressure into a balloon situated within or adjacent a body cavity to effect tissue expansion. A drainage mechanism, especially useful in treating bladder deficiencies, is disclosed which permits drainage of body fluids while the balloon is in its expanded state. The balloon can be adapted to fit within a recess of the catheter, for example, in a folded or rolled state, and the invention also provides means for delivering fluid at a controlled pressure into an inflatable balloon.
An important part of the RNA components of the transcriptional regulatory network is the so-called, microRNAs (miRNAs), 20-22nt single stranded non-coding RNAs that act mainly as posttranscriptional repressors. They have been shown to control the transcriptional networks by acting on multiple targets and regulating the protein dosage of entire signaling pathways and have been implicated in many diseases. Recently, long non-coding RNAs (lncRNA) and circular RNAs (circRNAs) have also been described to regulate gene expression in different ways.. In our laboratory we study the contribution of non-coding RNAs, including microRNAs, to the regulation of endothelial cell and macrophage functions. Both cell types play major role in controlling both angiogenic and inflammatory responses and the interplay between these two cell types has been shown to be critical for several pathophysiological conditions like atherosclerosis, cancer (tumor growth), adipose tissue expansion and wound healing, among others. ...

Each year, the number of breast cancer survivors who choose post-mastectomy breast reconstruction keeps rising. Among women who elect to pursue breast reconstruction, approximately 75% will choose prosthetic breast reconstruction. Implant-based breast reconstruction is frequently achieved in two-stages. The first stage consists of the placement of a tissue expander after mastectomy. This is followed by a period of biweekly tissue expansions that can last several months. In the second stage, the tissue expander is removed in a surgical procedure and replaced with a permanent breast implant. Tissue expansion is a well-established breast reconstruction technique characterized by high success rates and high patient satisfaction. Despite the well-recognized advantages of this successful breast reconstruction technique, the subpectoral placement of a tissue expander is associated with significant pain and discomfort in the immediate post-operative period and during the phase of tissue expansion. ...

The success of bone augmentation to a major degree depends on the biomechanics and biological conditions of the surrounding tissues. Therefore, an animal model is needed providing anatomical sites with similar mechanical pressures for comparing its influence on different biomaterials for bone regeneration. The present report describes the new bone formation associated to biomaterial in a bursa created in the epidural space, between dura mater and cranial calvaria, under the constant pressure of cerebrospinal fluid. Five adult California rabbits were used for the trial. In each animal, two bursae were created in the epidural spaces, in the anterior part of the skull, below both sides of the interfrontal suture. The spaces between dura mater and cranial calvaria were filled with in-situ hardening biphasic calcium phosphate containing hydroxyapatite and beta tricalcium-phosphate (BCP), in-situ hardening phase-pure beta-tricalcium phosphate (β-TCP) or without any biomaterials (sham). TED is a progressive, debilitating autoimmune disease with a limited window of active disease during which it can be treated without surgical intervention.1,2 While TED often occurs in people living with hyperthyroidism or Graves disease, it is a distinct disease that is caused by autoantigens activating an IGF-1R-mediated signaling complex on cells within the orbit.3,4 This leads to a cascade of negative effects, which cause long-term, irreversible damage. Active TED lasts for up to three years and is characterized by inflammation and tissue expansion behind the eye.5,1 As TED progresses, it causes serious damage - including proptosis (eye bulging), strabismus (misalignment of the eyes), and diplopia (double vision) - and in some cases can lead to blindness.2,6 Currently, patients must suffer through active TED until the disease becomes inactive - often left with permanent and sight-impairing consequences - before they can have complex and costly surgical procedures that may never fully ...

View more ,Abstract: Insulin resistance ensues when normal physiological concentrations of insulin are unable to induce effective cellular insulin signalling and glucose uptake by insulin sensitive tissues. It is caused by several abnormalities that include; 1) an overabundance of circulating free fatty acids (and dyslipidaemia), 2) systemic inflammation caused by increased tissue and circulating pro-inflammatory cytokines, and, 3) over activation of the systemic and organ specific renin-angiotensin systems. Although usually associated with obesity, insulin resistance is not a condition that only afflicts obese individuals. Dyslipidaemia which is implicated in the aetiology of insulin resistance can be caused by adipose tissue expansion (obesity) or the increased consumption of lipogenic fructose which has profound effects on liver metabolism and serum lipid profiles. The primary reason fructose is implicated in insulin resistance is because it induces hepatic lipogenesis which would directly ...
