Each time a specimen is exposed to air there will be some loss of some antigens and nucleic acids due to oxidation from atmospheric ozone, acids, etc circulating in the lab. Not all antigens are affected, not every antibody has a problem with it, but it does happen and is well recognized in tissue array labs. Many labs have documented this phenomenon. Most notable is David Rimms lab at Yale that has made an art out of preserving antigens on tissue arrays by storing them in a special cabinet filled with nitrogen gas. They see dramatic losses in staining intensity in sections left in air for just a few days. The problem with reheating tissue array cores is that this oxidation effect is grossly accelerated by the higher temperatures and air is allowed to penetrate farther into the core once the paraffin has softened from the heat. Background Pancreatic ductal adenocarcinoma (PDAC) is a deadly cancer worldwide, as a result of a late diagnosis and limited therapeutic options. Tumour microenvironment (or stroma) plays a key role in cancer onset and progression and constitutes an intrinsic histological hallmark of PDAC. Thus we hypothesised that relevant prognostic biomarkers and therapeutic targets can be identified in the stroma. Methods Laser microdissection of the stroma from freshly frozen PDAC was combined to gene expression profiling. Protein expression of candidate biomarkers was evaluated by immunohistochemistry on tissue microarrays (n = 80 tumours) and by ELISA in plasma samples (n = 51 patients). Results A signature made of 1256 genes that significantly discriminate the stroma from the non-tumour fibrous tissue was identified. Phospholipase Cγ1 (PLCγ1) is highly expressed in human tumours. Our previous studies reported that both stable and inducible PLCγ1 down-regulation can inhibit formation of breast-cancer-derived experimental lung metastasis. Further, high expression of PLCγ1 and its constitutively activated forms (i.e., PLCγ1-pY1253, PLCγ1-pY783) is associated with worse clinical outcome in terms of incidence of distant metastases, but not of local relapse in T1-T2, N0 breast cancer patients. In the present retrospective study, we analysed the prognostic role of PLCγ1 in early breast cancer patients stratified according to the St. Gallen criteria and to their menopausal status. PLCγ1-pY1253 and PLCγ1-pY783 protein expression levels were determined by immunohistochemistry on tissue microarrays, and were correlated with patients clinical data, using univariate and multivariate statistical analyses. Micro-Array can be done on Day 1, 3, or 5 embryos. Obviously, Day 5 is preferable, however were leaving it up to the embryologists to make the call on Day 3. If they look good, well continue to grow them out to Day 5, but if they have doubts, then weve also consented to a Day 3 biopsy.
