Thiostrepton is a natural cyclic oligopeptide antibiotic of the thiopeptide class, derived from several strains of streptomycetes, such as Streptomyces azureus and Streptomyces laurentii. Thiostrepton is a natural product of the ribosomally synthesized and post-translationally modified peptide (RiPP) class. Thiostrepton was discovered by Donovick et al. who described its antibacterial properties in 1955. Dorothy Crowfoot Hodgkin solved the structure of thiostrepton in 1970. Early in 1978, Bycroft and Gowland proposed the biosynthesis of thiostrepton, which was still unclear until 2009. This study shows for the first time that the novel thiazole antibiotic thiostrepton selectively induces cell death in breast cancer cells through the down-regulation of FOXM1 expression. Furthermore, thiostrepton treatment, accompanied by a loss of FOXM1 expression, results in a reduction in the proliferation, invasiveness, and transformation ability of breast cells. The conformational dynamics of the protein between the bound and unbound state play an important role in the binding process. We have studied those dynamical changes in detail for the highly conserved complex between the ribosomal protein L11 and the GTPase region of 23S rRNA. The RNA domain is compactly folded into a well defined tertiary structure, which is further stabilized by the association with the C-terminal domain of the L11 protein (L11ctd). Methods and results: Levels of FOXM1 and its targets were determined by immunoprecipitation and real-time PCR analyses in rat and human samples. Liver regeneration studies with transgenic mice demonstrated that FoxM1B regulates the onset of hepatocyte DNA replication and mitosis by stimulating expression of cell cycle genes (10, 22, 23, 26). In this study, we used Alb-Cre recombinase to generate a hepatocyte-specific deletion of the Foxm1b gene and demonstrated that Foxm1b is required for normal levels of hepatocyte DNA replication and is essential for mitosis in regenerating liver. We found no significant increase in hepatocyte apoptosis in regenerating Alb-Cre Foxm1b−/− liver (data not shown), suggesting that Foxm1b is required for hepatocyte proliferation but not survival. The transcription factor Forkhead box M1 (FOXM1) is a key regulator of cell proliferation and is overexpressed in many forms of primary cancers, leading to uncontrolled cell division and genomic instability. To address the role of FOXM1 in chemoresistance, we generated a cisplatin-resistant breast cancer cell line (MCF-7-CISR), which had an elevated level of FOXM1 protein and mRNA expression relative to the parental MCF-7 cells. A close correlation was observed between FOXM1 and the expression of its proposed downstream targets that are involved in DNA repair; breast cancer-associated gene 2 (BRCA2) and X-ray cross-complementing group 1 (XRCC1) were expressed at higher levels in the resistant cell lines compared with the sensitive MCF-7 cells.
