The three-dimensional structure of the proteasome from the archaebacterium Thermoplasma acidophilum has been elucidated by x-ray crystallographic analysis by means of isomorphous replacement and cyclic averaging. The atomic model was built and refined to a crystallographic R factor of 22.1 percent. The 673-kilodalton protease complex consists of 14 copies of two different subunits, alpha and beta, forming a barrel-shaped structure of four stacked rings. The two inner rings consist of seven beta subunits each, and the two outer rings consist of seven alpha subunits each. A narrow channel controls access to the three inner compartments. The alpha 7 beta 7 beta 7 alpha 7 subunit assembly has 72-point group symmetry. The structures of the alpha and beta subunits are similar, consisting of a core of two antiparallel beta sheets that is flanked by alpha helices on both sides. The binding of a peptide aldehyde inhibitor marks the active site in the central cavity at the amino termini of the beta subunits and

Imagine a discarded coal pile in a rural Indiana setting, burning slowly and steadily over time, like an unattended barbecue. This seems an unlikely place to look for life. Yet this is just the habitat from which the acid-loving archaeon T. acidophilum was first isolated by Darland and colleagues [2], in the late 1960s. T. acidophilum is an unusual microorganism from a variety of perspectives. It grows at very low pH values, ranging from 0.8 to 4.0, and at temperatures ranging from 45 to 63°C. Unlike most microorganisms, T. acidophilum has no cell wall - its cytoplasm is separated from the outside world by a thin membrane composed of phytanyl and biphytanyl ether lipids bound to a glycerol backbone. This membrane lipid composition, along with the evolutionary affinities of its ribosomal RNAs and some conserved proteins, identify T. acidophilum as an archaeon. Although T. acidophilum appears to fall within the archaeal subgroup Euryarchaeota, it is only distantly related to the extremely ...

The current state of GMOD presentations by Scott Cain, Dave Clements were very useful, as were the later updates on the release status of what you might call the core GMOD components (e.g., chado, GBrowse and also Mitchells talk on JBrowse.) I liked Michael Alauxs presentation on the Chado Controller because it was an example of coming up with a general solution to a problem that other people might be able to use too. I thought Brian OConnors Adventures in NOSQL was great because in 15 minutes he presented a bunch of useful information that it would take me hours to compile on my own. Dr. Martones invited talk was also excellent and very impressive and I thought the open mic session was also a great idea ...

If you want to migrate from one org to another, then you first need to get a Weekly Export Service download. Make sure you check the Include attachments box. You will get an attachments.csv and the actual attachments.. The key to importing attachments is a little known feature inside Data Loader. The part that is not documented is that the import wants the actual filename in the BODY field. So for example, the attachments themselves have a ID name, lets say its 123456789 and it is on your hard drive in the C:\Export folder. So in the BODY field you need to put C:\Export\123456789.. The only fields you need in the import file are - ParentId, Name, IsPrivate, OwnerId and Body. ...

In order to elucidate the functional and structural features of MTHFDC from archaeons at a molecular level, it was overexpressed in Escherichia coli and crystallized in the presence of its cofactor, NADP, at 295 K using polyethylene glycol (PEG) 4000 as a precipitant. The crystal is a member of the monoclinic space group P211, with the following unit cell parameters: ...

Broadly three major groups of hypotheses have been put forward for the possible origin of mitochondriate eukaryotes The first group postulates the endosymbiosis of an Archaea such as a Thermoplasma, methanogens, or a crenarchaeote (eocyte) with a Bacteria being a spirochaete, an H2-producing 8-Proteobacteria, or a Gram-negative bacterium, respectively. This is followed by an O2-consuming a-Proteobacteria such as a Rickettsia . The second group proposes that a Gram-positive bacterium gave rise to neomuran, the ancestor of both Archaea and current Bacteria The bacterial arm then engulfed again an O2-consum-ing a-Proteobacteria such as a Rickettsia . Both groups assume an amitochondriate eukary-ote as an intermediate . The fact that no ancestral amitochondriate eukaryote has ever been detected and that all extant amitochondriate eukaryotes have secondarily lost their mitochondria or still possess equivalents to mitochondria, led to the third group of hypotheses without an amitochondriate eukaryote ...

The immune system recognizes foreign pathogens and eliminates them. This occurs in several phases. In the early inflammation phase, the pathogens are recognized by antibodies that are already present (innate or acquired through prior infection; see also cross-reactivity). Immune-system components (e.g. complement) are bound to the antibodies and kept near, in reserve to disable them via phagocytosis by scavenger cells (e.g. macrophages). Dendritic cells are likewise capable of phagocytizing but do not do it for the purpose of direct pathogen elimination. Rather, they infiltrate the spleen and lymph nodes, and each presents components of an antigen there, as the result of which specific antibodies are formed that recognize precisely that antigen. These newly formed antibodies would arrive too late in an acute infection, however, so what we think of as immunology constitutes only the second half of the process. Because this phase would always start too late to play an essential role in the ...
It can be seen that Archeopteryx possesses many more characters which are present in dinosaurs and not in birds, than it does characters which are present in birds but not in dinosaurs. This is why Archeopteryx is a true transitional species, because it shares some characters which are diagnostic of one group whilst still retaining characters diagnostic of its ancestral group. Anyone who claims that Archae is 100% bird is wrong. Anyone who claims that Archaes skeleton is even predominantly bird- like is wrong. Anyone who claims Archae has a totally birdlike skull is wrong ...

Chimeric models claim that two prokaryotic cells existed initially - an archaeon and a bacterium. These cells underwent a merging process, either by a physical fusion or by endosymbiosis, thereby leading to the formation of a eukaryotic cell. Within these chimeric models, some studies further claim that mitochondria originated from a bacterial ancestor while others emphasize the role of endosymbiotic processes behind the origin of mitochondria. Based on the process of mutualistic symbiosis, the hypotheses can be categorized as - the serial endosymbiotic theory (SET),[122][123][124] the hydrogen hypothesis (mostly a process of symbiosis where hydrogen transfer takes place among different species),[117] and the syntrophy hypothesis.[125][126]. According to serial endosymbiotic theory (championed by Lynn Margulis), a union between a motile anaerobic bacterium (like Spirochaeta) and a thermoacidophilic crenarchaeon (like Thermoplasma which is sulfidogenic in nature) gave rise to the present day ...

I am trying to detect when there is a current in a given circuit using only a digital input and basic components (e.g. diodes, resisters, transistors); current is intermittent and power is small, 40-100mA @ 12V. I do not need to measure the current, simply know when it is present or not present.. I am trying to avoid additional chips e.g. INA219, optocouplers, etc. Is it possible to detect (again, not measure, only detect) current this way? Detection does not need to be rapid, within 100ms would be fine.. The load can have resistors, diodes, etc. in series or parallel. A voltage drop of up to 2 volts is fine. The power supply is stable around 12 volts and near the load. The digital input/output logic is 3.3V (Raspberry Pi); the presence of current would ideally be read as 3.3V (or at least 2) while the lack of current would ideally be read as 0V (or less than 2).. The important detail I forgot to mention is that the voltage source switches polarity. It is stable at 12 volts, but current can flow ...

This article describes a teaching and learning active methodology based on the use of physical models as tools in medical physiology. The fact that, at the end of the process, students were capable of successfully creating their own PPMs with great representativeness of physiological phenomena presents itself as the main finding of the reported experience.Concerning the effectiveness of the methodology described when it comes to the models used as learning tools, the integrative character of the proposal allowed to transpose physiological concepts from theory to an immediate and practical application. As shown in Figure 3, the level of difficulty for the construction of each model was directly related to the conceptual design and the materials used for concretizing the idea. Students were stimulated to use, whenever possible, materials easily available, such as recyclable ones. At specific circumstances, however, special components (e.g. a car battery or electronic systems) were required for ...
Computer Aided Design (CAD) and Computer Aided Geometric Design (CAGD) are used heavily in the representation of three-dimensional objects. Other than the areas of civil and mechanical engineering the methods from these fields are increasingly being used in the field of Medical Technology for the planning and production of medical tools and implants. It is important that the medical components (e.g., the surgical cutting blocks) be described with respect to the geometry of the anatomical structures (e.g, teeth, bones, organs, etc) so as to determine the the appropriate shape, size and position of the component. ...

Hello everybody, here it is my first question. I am using Ni-NTA agarose beads to purify a protein. According to the manufacturer the limit pressure is around 2 psi (nothing), and I am running the column to 50 psi. Can anyone please tell me about why does the pressure limit is important when dealing with affinity chomatography? How can this (pressure limit too high) affect my purification? Am I destroying the beads? I am running the column (home-prepacked tricorn) on an akta-prime. I would really appreciate any information since I couldnt find any info on the web. I have beeen unable to determine whan this was proposed. Everyone says recently so I would say in the last five to six years. I was not able to find it anywhere that this kingdom had been created. Everyone still seems to be saying that it falls under the kingdom of Fungi. These things take years to bring about. The last time a new kingdom of life was proposed, the Archae, it took 15 years to get the new kindom accepted. It took ten years for the majority other researchers in the field to stop calling the researcher that originally proposed a new kingdom a lunatic. I wish I were kidding. Found in all living organisms, terpenoids make up the largest group of natural products and are essential compounds for many major processes, including photosynthesis, respiration, hormone production, and electron transport. Additionally, they have commercial and medical value in products including fragrances, cosmetics, and medicines. Terpenoids originate from the five-carbon building blocks isopentenyl diphosphate (IPP) and dimethylallyl diphosphate (DMAPP), which are synthesized by the mevalonic acid (MVA) and methylerithritol phosphate (MEP) pathways. An alternative MVA pathway was discovered in Archaea with the final two enzymes being phosphomevalonate decarboxylase (MPD) and isopentenyl phosphate kinase (IPK). Even though this alternative pathway is not present in plants, presence of IPK was retained. The overexpression of IPK in planta indicates that IPK plays a significant role in the MVA pathway by synthesizing IPP/DMAPP from an IP/DMAP pool for terpenoid biosynthesis. It has been suggested
0070]Experiments were conducted using various ratios of acetone and toluene, resulting in the discovery that a 50/50 wt % mixture is an acceptable vehicle for dissolving both the inorganic salt and the organic resin in a common solvent system. A set of component samples (Sample Component E, F), were made using an in-house lab-scale pultrusion process that was used to combine 25 g of EPON 862 resin and 0.14 g of cross-linking agent (Lindax 1) with 10 g of a 50/50 mixture of toluene and acetone into which 0.05 gms of PdCl2 was dissolved. A length of about 2 meters of resistive carbon fiber (Hexcel/MTLS produced) was segment-wise immersed into the resin solvent system mixture, resulting in impregnation of the fiber with the liquid solution containing polymer, crosslinking agent and catalyst, then tensioned and pulled into and through a prototype forming die consisting of the tapered section from a conventional, laboratory-grade, glass pipette. The die served to condense and compress the wetted ...
For S. aureus, secreted proteins such as Eap have pleiotropic effects, mediating adherence to host extracellular matrix components (e.g., fibronectin, fibrinogen, vitronectin, collagens, and elastin) and thereby potentially contributing to bacterial colonization of host tissues (10) and, in part, to invasion of nonprofessional phagocytes (19-21). In addition, Eap has anti-inflammatory and immunomodulatory functions in the host, indicating that Eap constitutes a potent virulence factor in the course of staphylococcal infections (1, 8, 15, 18, 25). Thus, Eap acts as a secretable expanded repertoire adhesive molecule (9).. In this study, the Eap-encoding gene (eap) was studied as a diagnostic target for specific identification of S. aureus by PCR amplification. Whereas sequencing results suggest that the eap sequences are in general highly conserved, size differences are known for the encoding gene, as a result of different numbers of repeats, as well as for the translated product, as additionally ...

The importance of the wash step in Gram stain is, that to remove the dye from the walls of bacteria so coloring is not mixed with final results, as well the dye for gram positive stays one while from gram negative lightens. The order of dyes are important because, Gram Positive bacteria have thick peptidoglycon walls that retain the dark color, so violet is first used, then after the wash for gram negative the color washes off a bit, and then a lighter color is added so the Gram Positive will show purple and negative will show light red/pink ...
In this paper, we continue exploration of the dynamical and parameter planes of one-parameter families of Schwarz reflections that was initiated in \cite{LLMM1,LLMM2}. Namely, we consider a family of quadrature domains obtained by restricting the Chebyshev cubic polynomial to various univalent discs. Then we perform a quasiconformal surgery that turns these reflections to parabolic rational maps (which is the crucial technical ingredient of our theory). It induces a straightening map between the parameter plane of Schwarz reflections and the parabolic Tricorn. We describe various properties of this straightening highlighting the issues related to its anti-holomorphic nature. We complete the discussion by comparing our family with the classical Bullett-Penrose family of matings between groups and rational maps induced by holomorphic correspondences. More precisely, we show that the Schwarz reflections give rise to anti-holomorphic correspondences that are matings of parabolic anti-rational maps ...
