The isolated material contained 0.7-0.8 mole of formaldehyde/mole of tetrahydrofolate rather than 1 required for the 5,10-methylene structure. This finding was verified by chemical determination of CH2O on the isolated material. Comparable experiments with CH2O derivatives of tetrahydroaminopterin yielded 0.7 mole of CH2O/mole of tetrahydropteridine. I am aware of some unpublished data that has shown that the levels of 5-MTHF are quite low in the plasma of CFS/ME patients, typically something like 60 to 70% lower than in normal controls. Other folate derivatives are also low, but not as low relative to normals as 5-MTHF. The only interpretation of this that I am aware of is that peroxynitrite, presumably elevated as a consequence of the NO/ONOO- cycle, is leading to the oxidation of 5-MTHF and subsequently some loss of total folate as well.
