Taxus » Musei Villa Baciocchi
Taxus (Taxus baccata L., 1753) is a genus of small coniferous trees in the yew family Taxaceae, it is widely used in landscaping and ornamental horticulture or it is used for formal hedges and topiary. Taxus is also known as the tree of death.. It is a small to medium-sized evergreen tree, the bark is thin, scaly brown, coming off in small flakes aligned with the stem. They are relatively slow-growing and can be very long-lived, and reach heights of 10-20 meters, with irregular globose top and low-positioned branches. They have dark-green leaves, 3 cm long, arranged spirally on the stem, but with the leaf bases twisted to align the leaves in two flat rows either side of the stem. The leaves are poisonous. Chiral Enzymatic Activity in Cell Cultures of Taxus Species
The results showed an oscillatory behavior between er and reaction time, resulting in enantioselectivity changes according to pH conditions within 1 week. In the course of 1 day, we observed an increase of er in all pH treatments, but by the second day the opposite was observed with the exception of the treatment at pH 6.5 and 8.0. By the third day, a decrease in er was observed at pH 6.5, whereas a continuous increase was observed at pH 8.0. These results exhibited no periodic dependence between er and reaction time. Both enantiomers were identified as the main product in the enantiomeric mixture. At pH values of 4.0 and 6.0, the oscillatory behavior was also present, but the opposite enantioselectivity was observed during the reaction course because the (R)-enantiomer was mainly obtained. Enantioselective changes were previously reported when racemic benzoin was deracemized by Rhizopus oryzae at pH 7.5-8.0 and pH 4.0-5.0 [29].. The present results suggest that Taxus globosa Schltdl. cells are ...

SYNTAX Study: TAXUS Drug-Eluting Stent Versus Coronary Artery Bypass Surgery for the Treatment of Narrowed Arteries - Tabular...
Due to the introduction of drug-eluting stents (DESs) and to improvements in therapy for both percutaneous coronary intervention (PCI) and coronary artery bypass surgery (CABG) patients, PCI is challenging CABG as the gold standard for treatment of three vessel (3VD) and left main (LM) coronary disease.. SYNTAX is a novel, randomized trial with nested registries comparing PCI with paclitaxel-eluting TAXUS stents to CABG for 3VD and LM patients to evaluate the best treatment for these patients with complex coronary disease.. Patients at participating centers will be evaluated by both a cardiothoracic surgeon and by an interventional cardiologist.. Those patients who are determined to be eligible for treatment by both PCI and CABG will be randomized to receive either PCI with a polymer-based paclitaxel-eluting TAXUS stent or CABG.. Patients who are determined to be unsuitable for treatment by PCI will be treated by CABG and will be entered into a CABG registry to help define the patient population ...
Paclitaxel total synthesis - Wikipedia
Paclitaxel total synthesis in organic chemistry is a major ongoing research effort in the total synthesis of paclitaxel (Taxol). This diterpenoid is an important drug in the treatment of cancer but, also expensive because the compound is harvested from a scarce resource, namely the Pacific yew (Taxus brevifolia). Not only is the synthetic reproduction of the compound itself of great commercial and scientific importance, but it also opens the way to paclitaxel derivatives not found in nature but with greater potential. The paclitaxel molecule consists of a tetracyclic core called baccatin III and an amide tail. The core rings are conveniently called (from left to right) ring A (a cyclohexene), ring B (a cyclooctane), ring C (a cyclohexane) and ring D (an oxetane). The paclitaxel drug development process took over 40 years. The anti-tumor activity of a bark extract of the Pacific yew tree was discovered in 1963 as a follow up of a US government plant screening program already in existence 20 years ...

Adam Boxer, University of California, San Francisco, presented his centers Phase 1 trial of TP1 287. Also known as abeotaxane, this small-molecule taxol derivative stabilizes microtubules. TPI 287 accumulates in the brain, and has been tested primarily to treat central nervous system tumors. Its application to tauopathies grew out of work showing beneficial effects of the microtubule stabilizer epothilone D in tau transgenic mice (Zhang et al., 2012). Testing of epothilone D in AD patients started in 2012 but was discontinued for lack of efficacy.. Boxers group examined the safety and tolerability of TP1 287 in 44 people with the primary four-repeat tauopathies cortical basal degeneration (CBD) or progressive supranuclear palsy (PSP), and in 33 people with AD. Participants received abeotaxane by intravenous infusion once every three weeks for nine weeks, with an option for open-label extension up to three months.. Why do cats eat plants, and should I be worried? | Dr. Justine Lee, DVM, DACVECC, DABT, Board-Certified Veterinary Specailist -...
Other dangerous types of plants? Cardiac glycosides like foxglove, lily of the valley, kalanchoe, Japanese yew, etc. Most of these grow outside, but when in doubt, Id keep these out of reach. This type does not cause kidney failure, but can cause life-threatening heart arrhythmias and death when ingested by dogs or cats.. Another common houseplant that cats seen to chew on are Dieffenbachia or philodendron. These plants contain insoluble calcium oxalate (not soluble calcium oxalate, like so many websites erroneously mention!), which cause oral pain when chewed on. This isnt life-threatening however, and typically just results in foaming and frothing at the mouth. My advice? Keep these plants elevated out of reach, and if your cat does get into it, just offer something tasty to flush out the mouth - a small amount of chicken broth, canned tuna water (not oil!), or even chicken noodle soup. If your cat continues to vomit, a veterinary visit is a must.. My hint? I keep one spider plant in my ...

Arsenic and Old Leaves: The Art of Poisoning Your Fantasy Characters - Part 2 « Fantasy-Faction
Oooh, yummy red berries… and look, the birds are eating them! How tasty, Ill have another one… *yawn*… and maybe just lay down here for a quick nap…. …and then never wake up? Ah, yes. The allure of the English or European yew tree-lovely to look at, and tempting to take a bite from, and even better if you never want to wake up again.. In England, the yew is commonly known as the graveyard tree, though actually for more of an historical reason than anything ominous. Roman invaders to England used to hold church services under yew trees, and thus ancient yews are still occasionally found next to churches in England-just like graveyards.. Nearly every part of the yew tree is poisonous, with one small exception: the flesh of its bright, red berries. However, inside the aril is a poisonous seed, which can easily be ingested by accident. The color and shape of the berries is highly tempting for children, animals, and hungry passers-by, but eating even just a few seeds or leaves can be ...

Development of an automated, multiwell plate based screening system for suspension cell culture | BMC Proceedings | Full Text
An automated media blend screening was carried out in a second experiment highlighting another main area of application. Two seed trains of a CHO cell line, media blends of two growth media and two feeding strategies were screened in 120 wells on 6 well plates. A success rate of 100 % enabled the evaluation of all wells in terms of cell growth and productivity. An increase in viable cell density and product titer of about 20% in comparison to the reference process was achieved.. 2 L bioreactor runs were performed to confirm these optimized parameters. A total of 6 bioreactor runs using the identified best combination of media blends, seed train and feeding strategy verified the predicted results from the multiwell plates and showed an increase in productivity of about 15 %.. ...
DMOZ - Health: Pharmacy: Drugs and Medications: P: Paclitaxel
Paclitaxel is a chemotherapeutic drug originally developed from the bark of the Pacific Yew tree. Given the relative scarcity of Pacific Yew trees and environmental concerns over their loss, scientists sought to synthesize the product but settled instead on semisynthesizing it from the needle of the English Yew, a procedure now used in the creation of Bristal-Myers Squibbs Taxol. Paclitaxel was approved by the FDA in 1992 for use in the treatment of ovarian cancer and in 1994 for use in the treatment of breast cancer. It is not a cure for cancer but it may stop the abnormal cancer cells from dividing and growing so they eventually die.

Cancer Drug Stimulates Tripolar Mode of Mitosis - Science and Technology Research News
Taxane-based radiochemotherapy* is widely used in the treatment of various locally advanced cancers - including non-small-cell lung carcinoma (NSCLC). Taxanes have two effects: they inhibit cell division, and they also sensitize tumor cells for radiation therapy. Researchers at the Department of Radiation Therapy & Radiooncology at Munich University Hospital and the Radiation Cytogenetics Research Unit at Helmholtz Zentrum München have now demonstrated in detail how this works.. Specifically, the scientists investigated the effect of Paclitaxel, a member of the taxane family isolated from the bark of the Pacific yew (Taxus brevifolia). By using cell biological and biochemical approaches, we were able to show in the present study that clinically relevant doses of Paclitaxel induce tripolar instead of normal bipolar cell division, said first author Dr. Michael Orth of Munich University Hospital. Put in simple terms, instead of dividing into two cells, the cell divides into three. Tumor cells ...

We believe, unfortunately, that Hsien Loong is driven by a desire for power and personal popularity. His popularity is inextricably linked to Lee Kuan Yews legacy.. His political power is drawn from his being Lee Kuan Yews son. We have observed that Hsien Loong and Ho Ching want to milk Lee Kuan Yews legacy for their own political purposes.. We also believe, based on our interactions, that they harbour political ambitions for their son, Hongyi.. They reiterated their fathers wish to demolish his house at 38, Oxley Road (photo, above) upon his passing, and accused Hsien Loong and Ho Ching of wanting to preserve the house to enhance his political capital.. The values of Lee Kuan Yew are being eroded by his own son. Our father placed our country and his people first, not his personal popularity or private agendas.. We are very sad that we have been pushed to this. We feel hugely uncomfortable and closely monitored in our own country. We do not trust Hsien Loong as a brother or as a ...

Future Plants by Randy Stewart: Cephalotaxus - Plum Yew
Where during the winter season many typical Taxus - Yews are getting eaten to sticks in deer country; the similar looking but less well known Plum Yew is deer resistant making this evergreen very useful in many places the regular Yews are not. There are few plants other than Rhododendrons that are as good for creating a permanent green undergrowth in a hardwood forest. It also does well in sun or shade, and is tolerant of heat, drought and clay but not salt. These hardy Asian natives deserve to be much more widely used in the landscape ...
Plant cell culture
To: plant-biology at net.bio.net , From: fwl at nova.np.ac.sg (Forday Wayne Lee) , Subject: Plant cell culture , Date: 3 Sep 93 05:03:13 GMT , I know that plant cell and tissue culture has been around for long time. , , Does anyone know if any commercially viable products (except for more , plants) has come out of this technology ? , , From what I gather, plants dont excrete very well and they grow slowly. , , , How is it that high value products such as vanilla, taxol, quinine etc , are not widely produced by plant cell culture ? Certain plant cell lines can be genetically unstable. Thus they would be quite prone to loosing the valuable gene that they would contain. An example of such unstable types of cells is Alfalfa suspension cells, which frequently loose chromosomes and double others, if they are kept growing in culture for a long time. , , Again sorry if my questions are a bit simple, Im not a plantologist by , training. Dont be sorry asking questions is the only way youll learn. Oh, ...

Mosses are traditionally used as ornamental plants, especially in Japan, USA, England (moss gardens) and Germany, French (green roofs). In shadow areas (where the members of Poaceae usually cannot grow well), mosses are potentially use as grass. The stocks of moss colonies maintain optimal microclimate and decrease desiccation of soils (l...ike mulch). Additionally, mosses are evergreen, attractive all year, during winter. In our study, 18 moss species were propagated by fragments (as mixture, with the use of 16 species) and transplantation of carpets (with Brachythecium rivulare and Calliergonella cuspidata) in Szentendre. The aim was to find the most durable species and the best way of propagation. In an outdoor, irrigated garden, propagation by fragments was effective (with 63% coveration) and higher values (93% and 76%) were obtained in the cases of non-irrigated stocks of Amblystegium serpens (in trays) and moss carpets. In vertical structures (moss picture-frames with the use of ...

The Story of Taxol: Nature and Politics in the Pursuit of an Anti-Cancer Drug - Jordan Goodman, Vivien Walsh, Professor in...
Taxol is arguably the most celebrated, talked about, and controversial natural product in recent years. Celebrated because of its efficacy as an anticancer drug and because its discovery has provided powerful support for policies concerned with biodiversity. Talked about because in the early 1990s the American public was bombarded with news reports about the molecule and its host, the slow-growing Pacific yew tree. Controversial because the drug and the yew tree became embroiled in several sensitive political issues with broad public policy implications. Taxol has revolutionized the treatment options for patients with advanced forms of breast and ovarian cancers and some types of leukemia; it shows promise for treating AIDS-related Kaposis sarcoma. It is the best-selling anticancer drug ever, with world sales of $1.2 billion in 1998 and expected to grow. Goodman and Walshs careful study of how taxol was discovered, researched, and brought to market documents the complexities and conflicting interests

Photo: http://photos.prnewswire.com/prnh/20120516/LA07787) (Logo: http://photos.prnewswire.com/prnh/20051018/CARDIUMLOGO) The new research findings demonstrate that cardiac ischemia plays an important role in adenovector gene transfection (delivery) in mammalian hearts. Based on this understanding, using a standard balloon angioplasty catheter, researchers have developed and tested a new method to induce transient ischemia during a non-surgical interventional cardiac procedure, which when coupled with the infusion of nitroglycerin, boosts the delivery (cell transfection) of an adenovector gene construct into heart cells. The increase in adenovector-based gene transfection with the new technique is over two orders of magnitude (,100 fold).. Cardiums new method of adenovector delivery takes advantage of the findings that transient ischemia appears to alter the permeability barrier of the vascular endothelium and may expose the blood to the coxsackie-adenovirus receptor mediating adenovector ...

Transient Transformation of Maize BMS Suspension Cells via ...
Maize is one of the most important crop species and serves also as a model plant for grass research. A major bottleneck in maize research is stable transformation, which is both time and cost consuming, but also a technical challenge for most labs due to limited access to sufficient and optimal plant growth facilities. However, many studies in maize cell biology, physiology and biochemistry dont require stable transformed plants and can be accomplished by using transiently transformed suspension cultures. Here, we report a detailed protocol to establish Black Mexican Sweet (BMS) maize suspension cell cultures and transiently transform it via particle bombardment. We demonstrate how reliable subcellular protein localization data can be obtained within a very short time period and analyzed using open access software.

Numerous compounds are known which are useful in the prevention and treatment of various types of cancer. In order to effectively deliver these compounds by intravenous administration, it is generally preferred that the compounds be in solution to avoid or reduce the risk of blood clotting or other adverse effects that could result if the compounds were delivered in particulate form. Unfortunately, many of these compounds have poor solubility in water, the preferred solvent, and must be delivered using solvents which can cause adverse patient reactions that must in turn be prevented or controlled through the administration of other compounds. For example, paclitaxel is a known inhibitor of cell division or mitosis and is widely used in the treatment of ovarian, breast, lung, esophageal, bladder, head and neck cancers. Paclitaxel is a natural product originally purified from the bark of yew trees, but now obtained by semisynthesis from 10-desacetylbaccatin, a precursor purified from yew leaves. ...

cells with GpIba receptors in their membranes and platelets formed long beads-on-a-string formations on the endothelial culture cells when exposed to ultra large von Willebrand factor (ULVWF) multimers under high shear stress conditions.. When blood from a patient with TTP is tested using the Impact-R, surface coverage and aggregate size are expected to be large because of the presence of ULVWF in the patients plasma. When the same blood is exposed to plasma from a normal patient (which contains ADAMTS-13), and then tested by the Impact-R, theoretically, there will be a lessening of the surface coverage and aggregate size. This is the subject of a study by Shenkman in which he proposed that Impact-R could be used as a rapid screening test for TTP.. ...

Revenues in the first half of 2009 will be below expectations. Customers have found it difficult to gain credit to pay for orders and foreign exchange rate fluctuations haven t helped. Sales of paclitaxel, an anti-cancer ingredient made from yew trees, and leukaemia treatment ingredient Homoharringtonine have both suffered. Taihua says that it has been refusing orders from what it describes as unacceptably high risk customers. Sales have started to rise again, partly due to indications that the company s credit risk policy may be relaxed. Getting the balance right between a sensible credit risk policy and one that increases sales but leads to bad debts is going to be difficult. Shares in Taihua fell 1.5p to 7.75p each, which values the company at 6.33m. Taihua reported a 2008 profit of RMB16.4m on revenues of RMB42.3m, although that was well down on the profit of RMB39.8m and revenues of RMB71.8m in 2007. Nearly all of last year s profit and more than two-thirds of revenues were in the first ...

Temperature sensitivity of SOM decomposition governed by aggregate protection and microbial communities | Science Advances
Our results demonstrate that SOM decomposition in the topsoil was more sensitive to warming than in the subsoil. This significant difference could be due to the following two reasons. First, the stronger SOM protection by aggregates in the subsoil could attenuate Q10. Aggregate protection exerts important control over the stabilization of SOM (27), and it has been reported that small sizes of aggregates need more energy to be disrupted (11). That is to say, the degree of protection increases with the decline in aggregate sizes and would be stronger in microaggregates (27). In our study, a higher proportion of C stored in microaggregates in the subsoil compared with the topsoil indicated that aggregate protection was stronger in the subsoil, which further resulted in a lower Q10 value through the following two aspects. On the one hand, SOM is occluded in the interior of microaggregates, inducing spatial disconnection between substrates and enzymes, and thus protecting SOM from microbial ...
