Taxus cuspidata, commonly known as Japanese yew, is a broad-columnar needled evergreen tree or multistemmed shrub that is native to Korea, China, Russia and Japan. In its native habitat, it will grow to as much as 30-50 tall. In cultivation it will grow much smaller, particularly if regularly pruned. It features linear, spiny-tipped, dark green needles (to 1 long). Leaves are often tinged yellow beneath. Foliage may turn reddish-brown or yellow in winter. Scaly, reddish brown bark. Although classified as a conifer, female yews (plants are dioecious) do not produce cones, but instead produce red, ornamentally-attractive, berry-like fruits, each having a single seed almost completely surrounded by a fleshy red aril. Birds will feed on the berry-like fruits and help spread plants to other locations ...
Gymnosperms represent five of the six lineages of seed plants. However, most sequenced plant mitochondrial genomes (mitogenomes) have been generated for angiosperms, whereas mitogenomic sequences have been generated for only six gymnosperms. In particular, complete mitogenomes are available for all major seed plant lineages except Conifer II (non-Pinaceae conifers or Cupressophyta), an important lineage including six families, which impedes a comprehensive understanding of the mitogenomic diversity and evolution in gymnosperms. Here, we report the complete mitogenome of Taxus cuspidata in Conifer II. In comparison with previously released gymnosperm mitogenomes, we found that the mitogenomes of Taxus and Welwitschia have lost many genes individually, whereas all genes were identified in the mitogenomes of Cycas, Ginkgo and Pinaceae. Multiple tRNA genes and introns also have been lost in some lineages of gymnosperms, similar to the pattern observed in angiosperms. In general, gene clusters could be less
Taxine is a toxic alkaloid compound present in the Yew tree, Taxus baccata. It is composed of more than seven alkaloids, the main constituents being taxine A and taxine B; the other components are their derivatives, such as 2-deacetyltaxine A, isotaxine B and 1-deoxytaxine B. This substance is characterized by high melting and boiling point. Taxine is soluble in chloroform and alcohol, and insoluble in water or oil. Taxine can be found in Taxus species: Taxus cuspidata, T. baccata (English yew), Taxus x media, Taxus canadensis, Taxus floridana and Taxus brevifolia (Pacific or western yew). All of these species contain taxine in every part of the plant except in the aril, the fleshy covering of the seeds (berries). Each species contains a different amount of taxine concentration, which leads to varying toxicities within the genus. This is the case of the Taxus brevifolia or Pacific yew and the Taxus baccata (English yew); T. baccata contains high taxine concentration, which leads to a high ...
Taxus x media is a hybrid designation for a large number of shrubby, often wide-spreading crosses of English yew (Taxus baccata) and Japanese yew (Taxus cuspidata). These hybrids are noted for combining the ornamental excellence of English yew with the winter hardiness of Japanese yew. Although primarily resembling T. cuspitata in appearance, the various hybrid cultivars can vary considerably in size and character. Height ranges from 2-20 tall depending on the cultivar. Two ranked, pointed, oblong to needle-like, olive to dark green leaves are attractive year round. Bark is scaly brown. Plants are dioecious (separate male and female plants). Female plants produce red, berry-like fruits instead of cones. Each fruit has a single seed almost completely surrounded by a fleshy red aril. All parts of this plant are poisonous if ingested ...
Worldwide, Japanese yew (Taxus cuspidata Sieb. & Zucc.) is a popular garden tree, with large trees also being used for timber. In July 2012, leaf blight was observed on 10% of Japanese yew seedling leaves planted in a 500-m2 field in Andong, Gyeongsangbuk-do Province, South Korea. Typical symptoms included small, brown lesions that were first visible on the leaf margin, which enlarged and coalesced into the leaf becoming brown and blighted. To isolate potential pathogens from infected leaves, small sections of leaf tissue (5 to 10 mm2) were excised from lesion margins. Eight fungi were isolated from eight symptomatic trees, respectively. These fungi were hyphal tipped twice and transferred to potato dextrose agar (PDA) plates for incubation at 25°C. After 7 days, the fungi produced circular mats of white aerial mycelia. After 12 days, black acervuli containing slimy spore masses formed over the mycelial mats. Two representative isolates were further characterized. Their conidia were straight or ...

N2 - English yew (Taxus baccata L., Taxaceae), a Tertiary relict, provides a seminal example of a widespread albeit locally endangered (often close to extinction) tree species. In order to gain detailed insights into the evolutionary dynamics of the species on a broad geographical scale, over 1000 trees from 91 populations of English yew in the western Mediterranean were analyzed using seven nuclear microsatellite markers. Our results revealed contrasting patterns of genetic structure at different spatial scales: genetic variation was highly structured at the local scale, while only a low proportion of the observed variation was attributed to regional differences. Irans Hyrcanian forests cover a relatively narrow strip in the northeastern part of the country, and are among the most important and valuable ecosystems inscribed in United Nations Educational, Scientific and Cultural Organization (UNESCO) World Heritage List. European yew Taxus baccata L. is a Tertiary relict in the region and a long-lived dioecious tree with high ecological and economic importance in the Hyrcanian forests. To study the structure and analysing the survivorship of yew stands, we selected two forest reserves (Gazou and Afratakhteh) with European yew. In the two study areas, we established 165 0.1 ha circular sample plots (75 of the sample plots were in Gazou and 90 sample plots were in Afratakhteh) and measured three characteristics of each tree. The structure of the stands was quantified by means of 1) size distributions by diameter at breast height (DBH), 2) height classes, 3) and stand basal area. Comparison of the diameter distribution by DBH classes in two the forest ...

Taxus chinensis has undergone a large population reduction (more than 50%) since the 1990s as a result of the discovery of the importance of this species for treating cancer. There are attempts now to propagate this species to meet the demand, however, exploitation of wild plants is likely to continue but probably at a lower rate than before. Exploitation has reduced the population in many areas of China although the species is still wide-spread. Logging and deforestation in Viet Nam may have had a bigger impact as populations there are small; it may have reduced the species to some extent to the less accessible places. The plant is classified as Endangered in the IUCN Red List of Threatened Species(2013 ...

Elsbeth Walkers main area of research focuses on understanding the fundamental mechanisms underlying iron homeostasis in plants, which can set the foundation for increasing available iron in food crops. Her work on this subject began with the identification of Yellow Stripe1 (YS1), which encodes the iron-phytosiderophore transporter that is responsible for primary iron uptake in grass species. Subsequently, her group investigated the related family of proteins, YELLOW STRIPE1-LIKEs (YSLs), and demonstrated their involvement in the long distance movement of metals in plants. She is keenly interested in uncovering additional features of both the iron uptake, and long distance iron movement pathways. Elsbeth has more recently begun to investigate the genetic basis of taxol biosynthesis using cultured Taxus cells, using transcriptome analyses. She hopes that this effort will enable cheaper and more efficient production of this important anti-cancer drug.. Elsbeth is an enthusiastic teacher, and ...

Senescence is a key developmental process occurring during the life cycle of plants that can be induced also by environmental conditions, such as starvation and/or darkness. During senescence, strict control of genes regulates ordered degradation and dismantling events, the most remarkable of which are genetically programmed cell death (PCD) and, in most cases, an upregulation of flavonoid biosynthesis in the presence of light. Flavonoids are secondary metabolites that play multiple essential roles in development, reproduction and defence of plants, partly due to their well-known antioxidant properties, which could affect also the same cell death machinery. To understand further the effect of endogenously-produced flavonoids and their interplay with different environment (light or dark) conditions, two portions (red and green) of a senescing grapevine callus were used to obtain suspension cell cultures. Red Suspension cell Cultures (RSC) and Green Suspension cell Cultures (GSC) were finally grown under

The enzyme taxoid 14β-hydroxylase (14OH) directs a side-route of taxol pathway to 14β-hydroxy taxoids. Suppression of this side-route could increase the production of taxol. To suppress taxoid 14β- hydroxylase gene (14OH) expression in theTaxus × media TM3 cell line, antisense RNA inhibition approach was used in this study. Following the construction of an antisense RNA expression vector of 14OH from Taxus chinensis, the antisense 14OH cDNA (as14OH) was introduced into TM3 cells by Agrobacterium tumefaciens-mediated transformation. Southern blot analysis of hygromycin phosphotransferase gene (HYG) revealed that this selection gene was integrated successfully into the genome of Taxus × media cells. Reverse transcription-polymerase chain reaction (RT-PCR) analysis showed that the 14OH mRNA level in transgenic cells dropped dramatically, suggesting that the expression of endogenous14OH gene was significantly suppressed by the exogenous as14OH gene. Correspondingly, the total yield of three major C-14

Abstract: This study was conducted to find out the cause of root rot disease of blue pine (Pinus wallichiana) seedlings which is an important limiting factor in its successful production right from the nursery raising. The pathogens associated with the disease were isolated, morphologically characterised and identified as Fusarium oxysporum f.sp. pini (Schlecht.) Synd. and Hans., Rhizoctonia solani Kuhn and Macrophomina phaseolina (Tassi) Goid. F. oxysporum was the most abundant pathogenic fungus in diseased roots of blue pine seedlings with isolation frequency of 38.6%, whereas R. solani and M. phaseolina showed isolation frequencies of 11.0 and 3.3%, respectively. All the three fungi proved pathogenic and caused characteristic root rot symptoms when inoculated separately. Root rot disease of Pinus wallichiana seedlings was prevalent in all the three districts of Kashmir valley surveyed, during 2008 and 2009. ...

Taxus wallichiana Zucc. (Himalayan Yew) is often used in northern areas of Pakistan for the treatment of pyrexia, acute pains and epilepsy. We have investigated certain pharmacological activities of the methanol leaf extract against convulsion, nociception and pyrexia induced in rodents. The aim was to justify and explore its folk uses in these pathological conditions, on scientific basis. The studies were carried out using acetic acid-induced nociception and pentylenetetrazole-induced convulsions in mice, while formalin test and yeast-induced pyrexia in rats. Significant analgesic (67.77 and 74.29%) effect was found in acetic acid-induced model at doses of 100 and 200 mg/kg, i.p. respectively. Crude extract exhibited significant (P | 0.05) inhibition of the formalin noxious stimulation on both early and late phases of pain by the extracts (100 and 200 mg/kg doses). In case of yeast-induced pyrexia model, 200 mg/kg dose showed very significant (P | 0.01) inhibition while 50 and 100 mg/kg dose caused a
Sales of Boston Scientific Corp.s Taxus drug-eluting stent line fell 25 percent during the three months ended June 30, according to a detailed earnings report filed with the Securities and Exchange Commission. Total second-quarter sales for the coronary stent system were $269 million, compared to $358 million during the same period last year. Thats a slight improvement from the first quarter, which saw Taxus sales slide 28 percent compared with the first quarter of 2008.. The modest rebound jibes with a report by Angiotech, the Vancouver-based pharmaceutical company that makes the drug used in the stents coating. Angiotech said the royalties it derives from Taxus sales were showing small signs of improvement after several down quarters. But the details suggest the rebound for BSCs overall drug eluting stent business wasnt as robust as initially reported. In July, BoSci said second-quarter drug-eluting stent sales increased by $63 million in the U.S., or 36 percent. Included in that ...

In general, randomized trials are more rigorous than observational studies, yet they are often limited in power to address many individual clinical questions, due to the sample size of individual populations. Therefore, an acceptable compromise of a prospectively planned analysis compared with a performance goal was derived from historical data. This acceptability of a performance goal study on the basis of historical data was used for U.S. approval of the first-generation Taxus Liberté PES (Boston Scientific, Natick, Massachusetts) (31). The toxicity of yew (Taxus spp) is well known from ancient times and is mainly due to taxins acting as inhibitors of calcium and sodium transport across the cell membrane of cardiac myocytes. The confirmation of yew taxins in body fluids can be carried out by liquid chromatography-tandem mass spectrometry (LC-MS/MS). However, before selecting this precise but expensive technique, an orientation test should be done to ascertain yew presence as toxic agent in the organism. As the 3,5-dimethoxyphenol (3,5-DMP), myrtenol and 1-octen-3-ol appear as glycosidically bound volatile compounds and are very yew specific, the detection of 3,5-DMP and the measurement of 1-octen-3-ol / myrtenol concentration ratio constitute reliable indicators of yew presence in forensic cases. The detection of these compounds is easily performed by gas chromatography-mass spectrometry (GC-MS) (SIM) after an enzymatic hydrolysis (β-glucosidase) allowing the release of volatile compounds from yew glycosides. SID-530 is an intravenous formulation containing docetaxel, a semi-synthetic, second-generation taxane derived from a compound found in the European yew tree, Taxus baccata, with potential antineoplastic activity. Taxol analogue SID 530 binds to and stabilizes tubulin, inhibiting microtubule disassembly, which results in cell-cycle arrest at the G2/M phase and cell death. Check for active clinical trials or closed clinical trials using this agent. (NCI Thesaurus) (last updated: 6/23/2015)
A semi-synthetic, second-generation taxane derived from a compound found in the European yew tree, Taxus baccata. Docetaxel displays potent and broad antineoplastic properties; it binds to and stabilizes tubulin, thereby inhibiting microtubule disassembly which results in cell- cycle arrest at the G2/M phase and cell death. This agent also inhibits pro-angiogenic factors such as vascular endothelial growth factor (VEGF) and displays immunomodulatory and pro-inflammatory properties by inducing various mediators of the inflammatory response. Docetaxel has been studied for use as a radiation-sensitizing agent. (NCI04)

So how did the Yew go from deadly to medicinal? In 1962 the US National Cancer Institute collected plant specimens all over the country to test for any useful cancer treatment properties. The bark of T. brevifolia was found to have taxanes, a diterpene, which contains paclitaxel. This discovery was significant because paclitaxel has chemical properties that, when in the body of an animal, disrupt mitosis and acts as an antitumor treatment (Abal, 2003). Once it was found that Yew bark was a source of this new anticancer drug there was a rush to collect as much as possible putting the trees in danger of extinction. Push-back from environmental groups who feared losing these trees helped drive the pharmaceutical industry to create a synthetic version of the phytochemical (biologically active chemicals in plants). With the discovery of endophytic fungi growing with Yew trees and producing paclitaxel some relief may come to the trees, though this is still a relatively new discovery (Somjaipeng, ...

The results showed an oscillatory behavior between er and reaction time, resulting in enantioselectivity changes according to pH conditions within 1 week. In the course of 1 day, we observed an increase of er in all pH treatments, but by the second day the opposite was observed with the exception of the treatment at pH 6.5 and 8.0. By the third day, a decrease in er was observed at pH 6.5, whereas a continuous increase was observed at pH 8.0. These results exhibited no periodic dependence between er and reaction time. Both enantiomers were identified as the main product in the enantiomeric mixture. At pH values of 4.0 and 6.0, the oscillatory behavior was also present, but the opposite enantioselectivity was observed during the reaction course because the (R)-enantiomer was mainly obtained. Enantioselective changes were previously reported when racemic benzoin was deracemized by Rhizopus oryzae at pH 7.5-8.0 and pH 4.0-5.0 [29].. The present results suggest that Taxus globosa Schltdl. cells are ...

Patients at participating centers will be evaluated by both a cardiothoracic surgeon and by an interventional cardiologist.. Those patients who are determined to be eligible for treatment by both PCI and CABG will be randomized to receive either PCI with a polymer-based paclitaxel-eluting TAXUS stent or CABG.. Patients who are determined to be unsuitable for treatment by PCI will be treated by CABG and will be entered into a CABG registry to help define the patient population ...
Paclitaxel total synthesis in organic chemistry is a major ongoing research effort in the total synthesis of paclitaxel (Taxol). This diterpenoid is an important drug in the treatment of cancer but, also expensive because the compound is harvested from a scarce resource, namely the Pacific yew (Taxus brevifolia). Not only is the synthetic reproduction of the compound itself of great commercial and scientific importance, but it also opens the way to paclitaxel derivatives not found in nature but with greater potential. The paclitaxel molecule consists of a tetracyclic core called baccatin III and an amide tail. The core rings are conveniently called (from left to right) ring A (a cyclohexene), ring B (a cyclooctane), ring C (a cyclohexane) and ring D (an oxetane). The paclitaxel drug development process took over 40 years. The anti-tumor activity of a bark extract of the Pacific yew tree was discovered in 1963 as a follow up of a US government plant screening program already in existence 20 years ...

Adam Boxer, University of California, San Francisco, presented his centers Phase 1 trial of TP1 287. Also known as abeotaxane, this small-molecule taxol derivative stabilizes microtubules. TPI 287 accumulates in the brain, and has been tested primarily to treat central nervous system tumors. Its application to tauopathies grew out of work showing beneficial effects of the microtubule stabilizer epothilone D in tau transgenic mice (Zhang et al., 2012). Testing of epothilone D in AD patients started in 2012 but was discontinued for lack of efficacy.. Boxers group examined the safety and tolerability of TP1 287 in 44 people with the primary four-repeat tauopathies cortical basal degeneration (CBD) or progressive supranuclear palsy (PSP), and in 33 people with AD. Participants received abeotaxane by intravenous infusion once every three weeks for nine weeks, with an option for open-label extension up to three months.. In recruiting for the CBD cohort, Boxer screened with amyloid PET to exclude ...
Today, Biolyse Pharma is the industry leader in the research and development of injectable Paclitaxel products for use in cancer treatments.. Since Biolyse researchers first discovered that Paclitaxel could be produced from Taxus canadensis the company has gained fourteen years of valuable experience with the product. In September of 1994, Biolyse obtained authorization from the Federal Health Protection Branch (HPB) to administer its Paclitaxel for Injection, to patients under a clinical trial program.. Paclitaxel is a chemotherapeutic agent that is currently prescribed worldwide to treat the most aggressive forms of ovarian, lung and breast cancer, as well as AIDS related Kaposis sarcoma. Paclitaxel belongs to the chemical group known as taxanes, a family of molecules found in various natural sources. Paclitaxel is considered by the scientific and medical community as a first line of defense in the treatment of cancer. In 1992, both the American and Canadian National Cancer Institutes ...

Other dangerous types of plants? Cardiac glycosides like foxglove, lily of the valley, kalanchoe, Japanese yew, etc. Most of these grow outside, but when in doubt, Id keep these out of reach. This type does not cause kidney failure, but can cause life-threatening heart arrhythmias and death when ingested by dogs or cats.. Another common houseplant that cats seen to chew on are Dieffenbachia or philodendron. These plants contain insoluble calcium oxalate (not soluble calcium oxalate, like so many websites erroneously mention!), which cause oral pain when chewed on. This isnt life-threatening however, and typically just results in foaming and frothing at the mouth. My advice? Keep these plants elevated out of reach, and if your cat does get into it, just offer something tasty to flush out the mouth - a small amount of chicken broth, canned tuna water (not oil!), or even chicken noodle soup. If your cat continues to vomit, a veterinary visit is a must.. My hint? I keep one spider plant in my ...
Numerous studies have documented the negative effects of ozone (O3) on tree species in growing season, however, little is done in non-growing season. Three evergreen tree species, Phoebe bournei (Hemsl.) Yang (P. bournei), Machilus pauhoi Kanehira (M. pauhoi) and Taxus chinensis (Pilger) Rehd (T. chinensis), were exposed to non-filtered air, 100 nmol mol-1 O3 air (E1) and 150 nmol mol-1 O3 air (E2) in open-top chambers in subtropical China. In the entire period of experiment, O3 fumigation decreased net photosynthesis rate (Pn) through stomatal limitation during the transition period from growing to non-growing season (TGN), and through non-stomatal limitation during the period of non-growing season (NGS) in all species tested ...
Oooh, yummy red berries… and look, the birds are eating them! How tasty, Ill have another one… *yawn*… and maybe just lay down here for a quick nap…. …and then never wake up? Ah, yes. The allure of the English or European yew tree-lovely to look at, and tempting to take a bite from, and even better if you never want to wake up again.. In England, the yew is commonly known as the graveyard tree, though actually for more of an historical reason than anything ominous. Roman invaders to England used to hold church services under yew trees, and thus ancient yews are still occasionally found next to churches in England-just like graveyards.. Nearly every part of the yew tree is poisonous, with one small exception: the flesh of its bright, red berries. However, inside the aril is a poisonous seed, which can easily be ingested by accident. The color and shape of the berries is highly tempting for children, animals, and hungry passers-by, but eating even just a few seeds or leaves can be ...

An automated media blend screening was carried out in a second experiment highlighting another main area of application. Two seed trains of a CHO cell line, media blends of two growth media and two feeding strategies were screened in 120 wells on 6 well plates. A success rate of 100 % enabled the evaluation of all wells in terms of cell growth and productivity. An increase in viable cell density and product titer of about 20% in comparison to the reference process was achieved.. 2 L bioreactor runs were performed to confirm these optimized parameters. A total of 6 bioreactor runs using the identified best combination of media blends, seed train and feeding strategy verified the predicted results from the multiwell plates and showed an increase in productivity of about 15 %.. ...
Paclitaxel is a chemotherapeutic drug originally developed from the bark of the Pacific Yew tree. Given the relative scarcity of Pacific Yew trees and environmental concerns over their loss, scientists sought to synthesize the product but settled instead on semisynthesizing it from the needle of the English Yew, a procedure now used in the creation of Bristal-Myers Squibbs Taxol. Paclitaxel was approved by the FDA in 1992 for use in the treatment of ovarian cancer and in 1994 for use in the treatment of breast cancer. It is not a cure for cancer but it may stop the abnormal cancer cells from dividing and growing so they eventually die.

Taxane-based radiochemotherapy* is widely used in the treatment of various locally advanced cancers - including non-small-cell lung carcinoma (NSCLC). Taxanes have two effects: they inhibit cell division, and they also sensitize tumor cells for radiation therapy. Researchers at the Department of Radiation Therapy & Radiooncology at Munich University Hospital and the Radiation Cytogenetics Research Unit at Helmholtz Zentrum München have now demonstrated in detail how this works.. Specifically, the scientists investigated the effect of Paclitaxel, a member of the taxane family isolated from the bark of the Pacific yew (Taxus brevifolia). By using cell biological and biochemical approaches, we were able to show in the present study that clinically relevant doses of Paclitaxel induce tripolar instead of normal bipolar cell division, said first author Dr. Michael Orth of Munich University Hospital. Put in simple terms, instead of dividing into two cells, the cell divides into three. Tumor cells ...

We believe, unfortunately, that Hsien Loong is driven by a desire for power and personal popularity. His popularity is inextricably linked to Lee Kuan Yews legacy.. His political power is drawn from his being Lee Kuan Yews son. We have observed that Hsien Loong and Ho Ching want to milk Lee Kuan Yews legacy for their own political purposes.. We also believe, based on our interactions, that they harbour political ambitions for their son, Hongyi.. They reiterated their fathers wish to demolish his house at 38, Oxley Road (photo, above) upon his passing, and accused Hsien Loong and Ho Ching of wanting to preserve the house to enhance his political capital.. The values of Lee Kuan Yew are being eroded by his own son. Our father placed our country and his people first, not his personal popularity or private agendas.. We are very sad that we have been pushed to this. We feel hugely uncomfortable and closely monitored in our own country. We do not trust Hsien Loong as a brother or as a ...
