TY - JOUR. T1 - Low-dose aspirin does not attenuate platelet aggregation or atherosclerosis in miniature swine but decreases production of aortic wall prostacyclin. AU - Smith, M. J.. AU - Allen, K. G.D.. AU - Norman, J. F.. AU - Harris, M. A.. AU - Miller, C. W.. PY - 1995/11. Y1 - 1995/11. N2 - The objectives of this study were to determine if, and at what dose, aspirin could attenuate atherosclerosis in hypercholesterolemic Yucatan miniature swine, and to determine the influence of aspirin on aortic wall prostacyclin production and platelet aggregation. 30 Yucatan miniature swine (age 3 months) were fed either regular diet (RD), atherogenic diet (AD), or AD plus one of four aspirin dosages (2,4,8, or 16 mg/kg/d) for 6 months. The extent of atherosclerotic lesions in the abdominal aorta and coronary arteries was evaluated by sudanophilic staining and histological grading using Starys classification, respectively. Aortic wall production of prostacyclin (PGI2) and platelet aggregation were ...
ffERGs were used to assess changes in retinal function resulting from expression of the P23H RHO transgene and to characterize the progression of the RP phenotype in all six founders. ffERGs were recorded at 3 months of age and at regular intervals thereafter for 2 years. WT littermates served as controls, and the amplitudes of their b-wave were stable from 3 months of age and upward. In Figures 4A and 4B, the average scotopic (rod) and photopic (cone) b-wave amplitudes are plotted for two groups of founders. In two founders, 63-1 and 51-1, both scotopic and photopic ffERGs were initially normal, and the onset of photoreceptor dysfunction was delayed until 7 to 9 months of age. In the other four founders (53-1, 62-1, 63-2, and 64-1) both scotopic and photopic ffERGs were significantly reduced as early as 3 months and declined further with age, especially in the scotopic ERG. The retinal morphology of three of the six founders (51-1 at 18 months, 63-1 at 22 months, and 63-2 at 12 months) was ...
This study involved female Ossabaw miniature pigs. Unlike their larger cousins, Ossabaw miniature pigs have an average length of 14 - 20 inches and weigh 25 - 90 pounds. They were chosen over mice or rats because female Ossabaw mini-pigs could pose as metabolic animals model that could be understood if bacterial vaginosis (BV) were present.. Bacterial vagonis (BV) in human women is associated with an imbalance in their gut microbiome, which may lead to infertility and help determine the balance of good and bad bacteria within the intestines, which also lead to inflammatory conditions.. Using miniature pigs as feed models for female human obesity and reproductive issues has been done before because they more closely represent those female human characteristics than mice or rats. Analyzing markers resulting from different dietary sources while attempting to measure microbial gut flora content and balance with these pigs was a "thrifty" (their word) metabolic phenotype [2] model for these ...

The very first thing you need to do is check for a fever. Pigs core body temperatures shouldnt be more than 100 degrees in my experience; although each pigs normal temperature can be slightly different. Mini pigs have a lower core temperature than farm pigs and likely slightly lower than piglets whose tend to be a bit higher. Cathy Zolicani, DVM, was kind enough to write up an article about dealing with a vomiting pig and you can check that article out in our first aid section or by clicking here. Know that just because your pig doesnt have a temperature right now, doesnt mean your pig will not develop one in a couple of hours. Things can go from slightly worried to full panic mode super quick. A normal temp for most mini pigs older than 3 months is somewhere between 98-100. If my pig has a fever of 101? I personally monitor and take it again every 4 hours or so until its normal. At 102, I treat the fever. At 103 or above?, I call my vet. Having a low grade temperature can be helpful because ...

Homozygous Genotype This model features targeted disruption of the endogenous LDLR (low-density lipoprotein receptor) gene in Yucatan miniature swine genetics. Background In humans, loss-of-function mutations in LDLR lead to familial hypercholesterolemia, characterized by elevated plasma concentrations of cholesterol. Heterozygotes exhibit significantly higher cholesterol
The effect of ethanol feeding on the essential fatty acid content of tissues has been contradictory. To define the effect, we analyzed fatty acid profiles in various tissues from five miniature pigs fed daily 105 kJ basal diet/kg body wt and 146 kJ e
Treatments for bone nonunions (fractures that fail to heal) include surgery and bone grafting. As an alternative to viral gene delivery, Bez et al. developed a two-step therapy. First, endogenous mesenchymal stem/progenitor cells were recruited to the bone nonunion by implanting a collagen sponge in the defect site. Two weeks later, bone morphogenetic protein-6 (BMP-6) plasmid DNA and microbubbles were injected into nonunions, and ultrasound was applied to oscillate the microbubbles, which helped the recruited progenitors take up the BMP-6. This therapy led to transient BMP-6 secretion, bone regeneration, and fracture healing over 6 weeks in critical-sized tibial nonunions in mini-pigs. ...
We present experimental evidence that, in Ossabaw miniature swine, selective surgical excision of adipose tissue in direct contiguity with one of the epicardial coronary arteries attenuated the progression of atherosclerosis, thus supporting the hypothesis that cEAT could contribute to underlying coronary atherogenesis [13]. The findings are applicable to the early stages of CAD because of the relatively young age of the animals, the short duration of atherogenic diet feeding, and the lack of observed flow-limiting coronary stenosis typical of advanced clinical disease. We acknowledge the substantially high LDL cholesterol levels (,500 m/dL) in the obese group. We have conducted other studies in which the LDL was ~250 mg/dL (e.g. [10, 12]) for a longer duration, which yielded substantial coronary atherosclerosis. We predict a similar result of adipectomy. This very high LDL cholesterol level for a short duration is similar to LDLR−/− humans, who have substantial atherosclerosis. Horton and ...
Molecular characterization of the SLA (Swine Leukocyte Antigen) genes is important for understanding the immune responses between swine-donor and human-recipient in course of xenotransplantation. Explanation of association between alleles of SLA class I genes, type of pigs genetic modification, the PERV viral titer, and its subtypes may shed light on the nature of xenograft acceptance or rejection and the safety of xenotransplantation. No significant differences in PERV gag RNA level between transgenic and non-transgenic pigs were noted, likewise the type of applied transgene had no impact on PERV viremia. Type of SLA-1 gene profile may correspond with PERVs level in blood and thereby influence on their infectiveness. Screening tests of pigs should also enable selection of animals with low expression of PERV and exclusion specimens with PERV-C in the genome due to possible recombination between A and C subtypes, which may lead to autoinfection. 31.25% of study specimens shows the presence of ...
2018 Wolters Kluwer Health, Inc. All rights reserved. Background Over 130 000 patients in the United States alone need a lifesaving organ transplant. Genetically modified porcine organs could resolve the donor organ shortage, but human xenoreactive antibodies destroy pig cells and are the major barrier to clinical application of xenotransplantation. The objective of this study was to determine whether waitlisted patients possess preformed antibodies to swine leukocyte antigen (SLA) class II, homologs of the class II HLA. Methods Sera from people currently awaiting solid organ transplant were tested for IgG binding to class II SLA proteins when expressed on mammalian cells. Pig fibroblasts were made positive by transfection with the class II transactivator. As a second expression system, transgenes encoding the alpha and beta chains of class II SLA were transfected into human embryonic kidney cells. Results Human sera containing IgG specific for class II HLA molecules exhibited greater binding to ...

OBJECTIVE--To prove the safety and effectiveness of high frequency rotational ablation of coronary artery stenoses and occlusion in humans. SUBJECTS--106 patients with symptoms (91 men, 15 women) who had 67 significant stenoses, mainly types B and C, and 46-chronic occlusions. MAIN OUTCOME MEASURES--Mean change in diameter stenosis after rotational angioplasty alone and in combination with percutaneous transluminal coronary angioplasty immediately after treatment and 24 hours and six months later; restenosis rates at six months; complication of treatment. RESULTS--Rotational ablation could not be used in five stenoses and 16 chronic occlusions because of inability to reach or cross the lesion with the Rotablator guide wire. In four cases rotational ablation failed. Initial angiographic and clinical success by rotational ablation was achieved in 40 of the 67 stenoses (60%) and in 18 of the 46 chronic occlusions (39%). Additional balloon angioplasty was performed in 45 patients, increasing the ...
Dr. Sachs major areas of research are the following:. A. Induction of transplantation tolerance:. A major thrust of transplantation research in this laboratory has been directed toward induction of tolerance as a means of permitting transplantation of organs to be achieved without a requirement for immunosuppressive drugs. Two major models for tolerance induction are studied:. 1) Regulatory tolerance: Using miniature swine with defined MHC haplotypes (see below), Dr. Sachs and colleagues demonstrated that a short course of immunosuppression with a high-dose of calcineurin inhibitors is capable of inducing long-term tolerance in juvenile animals. This phenomenon has become and remains an important basic model for the study of tolerance induction across MHC barriers in a large animal model. Among active studies being carried out in this model are: a) the role of the thymus in permitting regulatory tolerance to develop; b) the dependence of tolerance on T regulatory cells; c) the adoptive transfer ...

Introduction: Notch signaling is a highly conserved pathway that promotes various types of proliferation including vascular and myocardial growth and development. Ongoing research suggests the VEGF and Notch pathways are tightly linked. We assessed the hypothesis that exogenous VEGF administration to ischemic myocardium would enhance the neovascular response and upregulate Notch signaling. Methods: Fourteen male Yorkshire swine underwent placement of an ameroid constrictor on the left circumflex artery to induce chronic myocardial ischemia. After 3 weeks, a perivascular pump delivering VEGF to the ischemic area was placed in 7 animals. All animals were examined 7 weeks after ameroid placement. The normal ventricle of the non-pump animals served as the control (NV, n=7), while the two experimental groups were comprised of the area at risk of the non-pump animals (AAR, n=7) and the area at risk of animals treated with perivascular VEGF (VEGF, n=7). Tissues were assessed by Western blot for protein ...

Site specific targeting remains an elusive goal for stem cell and gene therapies in the cardiovascular field. One promising option involves use of modified 3D stent designs that deliver larger cell/gene payloads to specific disease sites using the versatility of percutaneous vascular access. Here, we engineered smooth muscle cells (SMCs) placed in a 3D metallic stent scaffold to deliver high local concentrations of an angiogenic gene (VEGF165) in a porcine model of chronic total occlusion involving ameroid placement on the proximal left circumflex (LCx) artery(n≥8 per group). Engineered SMCs were retained within the mesh stent structure (479 ± 47.4 SMC/mm2) and were competent for VEGF production in vitro (8.8 ± 1.8 ng/ml in media). Following implantation, micro CT analyses revealed that stents loaded with VEGF producing SMCs significantly enhanced vasa vasora microvessel density (5.43 ± 0.46 vs 2.14 ± 0.19 vessels/mm2, p,0.001) with a concomitant increase in tissue VEGF protein levels ...
Purpose: : To evaluate ocular pharmacokinetics of TG100801 (VEGFR2 inhibitor) and its metabolite (TG100572, a potent multi-targeted Src/VEGFR2/PDGFR kinase inhibitor) following topical instillation in multiple preclinical species Methods: : TG100801 or TG100572 was administered as eye drops (10 - 50 µL/eye) to mice, rabbit, dog or mini-pig. Animals were sacrificed at various time-points and ocular tissues and plasma samples were collected. For rabbit study, both the left and right dissected ocular tissues from each animal were combined for analysis. In dogs and mini-pigs, dissected tissues from each eye were analyzed individually. In mice, ocular tissues from 2-4 eyes were combined and analyzed. Tissues were homogenized with RIPA buffer and analytes were extracted in acetonitrile. TG100801 and TG100572 were quantitated using an LC/MS/MS assay and concentrations were normalized using tissue weight. Data was analyzed using WINNONLIN. Results: : TG100801 is primarily converted to TG100572 by ...

In 1793 Young began to study medicine at St. Bartholomews Hospital in London although he was to leave in 1794 to continue his medical studies in Edinburgh. It was during this time that Young began to distance himself from his Quaker roots. After completing his studies in Edinburgh in 1795, Young travelled to Gottingen in Germany where he was awarded a Doctor of Physics degree in 1796.After obtaining his doctorate, Young moved to Emmanuel College, Cambridge where he taught and also began to research scientific matters outside the scope of medicine.. Having received a substantial inheritance on the death of his uncle, Young moved to London in 1799 and established himself as a physician at 48 Welbeck Street. Although his medical practice was never particularly successful, Young maintained that medicine was his primary occupation and so often published his academic articles anonymously to avoid damaging his reputation as a physician.. 37 Rather than work in isolation, several students would be doing experimental work at any one time, sharing facilities and assisting each other in various ways. They saw themselves as working in the samefield, they shared certain theoretical interests, and their various experimental projects frequently had a bearing on each other. In other words, these students, together with Wundt himself, formed a viable community of experimenters. 38 In Germany an independent second laboratory emerged at the University of Gottingen, and in 1890 a second journal, the Zeitschrift fur Psychologie und Physiologie der Sinnesorgane, made its appearance. 6 As long as that distinction was not made, psychology and philosophy remained indissolubly fused and the question of a specifically psychological methodology could not arise. Empiricist philosophers could go on appealing to their readers everyday self-awareness as they appealed to other aspects of their experience, without any feeling that this required any ...
A feral breed found on Ossabaw Island off the coast of Georgia near Savannah, Ossabaw hogs are said to have been brought to America by the Spanish during early American settlement. They are typically black, although some are black with white spots, white with black spots, or rarely red. Ossabaw are unusual and important for three reasons. First, its history as an isolated island population has meant that the Ossabaw is the closest genetic representative of historic Spanish stock. Second, the presence of pigs on Ossabaw Island provides scientists with an exceptional opportunity to study a long-term feral population that is well-documented. Third, the Ossabaw breed is biologically unique, having been shaped by natural selection in a challenging environment known for heat, humidity, and seasonal scarcity of food. They are able to store astounding amounts of body fat in order to survive. This biochemical adaptation is similar to non-insulin-dependent diabetes in humans, making the pigs a natural ...
