The present study examined the follicular populations prior to and during superovulation and investigated their relationship with superovulatory response in crossbred cattle. Eleven animals were administered i.m. 8 doses of Folltropin of 2.5 ml each (1.75 mg/ml) spread over 4 days beginning on Day 10 of oestrous cycle, and 30 and 20 mg Lutalyse was given along with the 5th and 6th injections of Folltropin respectively, to induce luteolysis. The animals were artificially inseminated 48, 60 and 72 h after the first Lutalyse injection. The number of corpora lutea (CL) was recorded by palpation per rectum and embryos were recovered non-surgically on Day 6 (Day 0 = day of superoestrus). The ovarian follicular population was examined by transrectal ultrasonography 15 h prior to and 52 h after the first FSH injection, and then on the day of superoestrus and the day of flushing. The follicles were classified on the basis of diameter as small (3-5 mm), medium (6-9 mm) and large (> 10 mm). The total ...

GnRHa is less potent at inducing ovarian vascular endothelial growth factor (VEGF), but may also affect endometrial angiogenesis and early placental development. In this study, we explore the effect of superovulation on endometrial angiogenesis during critical periods of gestation in a mouse model. We assigned female mice to three groups: natural mating or mating following injection with equine chorionic gonadotropin and trigger with GnRHa or hCG trigger. Females were killed prior to implantation (E3.5), post-implantation (E7.5), and at midgestation (E10.5), and maternal serum, uterus, and ovaries were collected. During peri
The effect of different eCG concentrations on the efficiency of superovulation in Formosan sambar dose. Hsin-Hung Lin, Chih-Hua Wang, Shann-Ren Kang, Wen-Lin Song, Chin-Hui Tseng, Mu-Jung Cheng, Shyh-Shyan Liu, and Perng-Chih Shen. The aim of this study was to examine the effect of different eCG concentrations on the efficacy of superovulation in Formosan Sambar does. All the does were implanted with CIDR for 12 days and superovulation was induced by intramuscularly injection with different concentrations of eCG at day 10. Embryos were collected by flushing the oviduct through midventral laparotomy at day 4 after mating. Results showed that the corpus luteum (CL) numbers of dose superovulated with 1500 (7.3), 2000 (10.6), 3000 (8.5) IU of eCG were significantly higher than that superovulated with 1000 IU of eCG (3.5). However, the embryo recovery rates of dose superovulated with 1000 (85.7%) and 1500 (62.0%) IU of eCG were significantly higher than those superovulated with 2000 (25.5%) and 3000 ...

Embryos from ageing (20 to 148 weeks of age) and young (20 to 30 weeks of age) donors were transferred to ageing (52 to 221 weeks of age) and young (18 to 30 weeks of age) recipients to partition the effects of ageing oocytes and uterine environment on embryo mortality. More than 3300 two- to eight-cell embryos collected following superovulation at six 6-month intervals were transferred. The average number of ovulation points per ageing donor doe superovulated at the six intervals declined with age and repeated superovulation. The average number of ovulations for the young donors at the six intervals also differed slightly, the low number (forty-one) for the last group possibly being due to the crossbred strain used. Both embryo recovery and cleavage rates usually exceeded 80% and did not differ between young and ageing donors. The percentage of viable young developing from embryos transferred from ageing and young donors showed that the potential for embryo development had not been impaired ...
The composition of follicular fluid (FF) varies with the cyclical hormonal changes and developmental stage of follicle. During superovulation programme, the large number of unovulated follicles is a major constraint and affects adversely the embryo recovery. An attempt was made to study the hormonal and biochemical profiles of unovulated follicles in goat superovulated with either Synthetic FSH (133 mg Folltropin) or 1000 IU PMSG (Folligon). The study revealed that Progesterone and testosterone concentrations were significantly (P. Key words: Goat, Superovulation, Unovulated Follicles, Follicular Fluid Composition ...

Superovulation of Bos taurus beef cattle with two injections of Folltropin. John F. Hasler. Bioniche Animal Health, Inc.. [email protected]. Due to the rapid absorption and short half-life of FSH, superovulation of cattle has traditionally been done with 7 or 8 bi-daily injections of FSH over a four day period. Restraining beef cattle for multiple injections involves time and expense. In addition, depending on breed and handling facilities, there are various degrees of stress to the cattle and increased risk of injury to both cattle and humans.. Trials conducted during the recent past involving Angus and Simmental cattle in Argentina demonstrated that when Folltropin was diluted in the glycosaminoglycan hyaluronan, two injections of Folltropin 48 h apart resulted in superovulation and embryo production comparable to the traditional 8 injection protocol (Tribulo, et al., 2011, 2012). The diluent containing hyaluronan is now characterized as a slow release diluent (SRF). These studies were ...
Abstract: Animal surgery procedures are being progressively restricted worldwide in the context of embryo transfer. In small ruminants, the needs for development of alternative and efficient non-surgical techniques for embryo transfer have been emphasized. Non-surgical embryo recovery is well consolidated in Brazil in cattle and goats, while in sheep it remains a challenge. The aim of this study was to check the efficiency of different ways of estradiol benzoate administration on cervix dilation and embryo recovery in synchronized Santa Inês ewes. Considering that Santa Inês sheep have up to 1.3 lambs we can conclude that it is possible to perform efficient non-surgical embryo recovery in non-superovulated synchronized Santa Inês ewes, regardless the way ...

Surrogate children - what kind of children are they? No similar children No similar questions are given rise to in respect of children who were conceived naturally. All seems to be clear here. However, a special interest is drawn to children who were conceived in-vitro and stayed in an artificial medium during the very first days of their development.. The beginning. In the process of hormonal stimulation of superovulation, female ovaries generate a number of oocytes having different sizes. Following their puncture (retrieval), genetics test each follicle to choose the top-quality follicles for their further fertilization.. As the man donates his semen, it goes through a special cleaning process. Using high-power laboratory microscopes, genetics examine seminal fluid to choose best-quality spermatozoa for fertilization purposes.. After genetic matter of both partners is ready, the fertilization procedure is run. Using an ultra-fine needle, a spermatozoid is introduced in each selected follicle. ...

Thirty-six domestic cats received 100 iu hCG (i.m.) on day 1, 2 or 3 of a natural, behavioural oestrus. Twenty-two anoestrous cats were injected with 150 iu pregnant mares serum gonadotrophin (PMSG; i.m.) followed 84 h later by 100 iu hCG. Twenty-four to 26 h after hCG, all cats were examined laparoscopically to determine the number of ovarian follicles and to recover follicular eggs. Mature eggs were cultured with conspecific spermatozoa and examined 30 h later for cleavage. Within the natural oestrus group, cats on day 1 produced fewer (P | 0.05) follicles and total eggs than females on day 2 or 3, and 88.9% of the day 1 eggs were degenerate or immature and unsuitable for in vitro fertilization (IVF). Although only 54.5% of the cats in the PMSG/hCG group exhibited overt oestrus, mean (± SEM) numbers of follicles (9.7 ± 0.8) and oocytes recovered (8.7 ± 0.8) were at least twofold greater (P | 0.001) than those measured in the natural oestrus group (3.7 ± 0.6; 3.4 ± 0.6, respectively) or subgroups
The hypothesis that pregnancy success could be improved in early postpartum ewes by prolonging the lifespan of the corpus luteum via active immunization against prostaglandin F2α (PGF2α) was tested. Further experiments in ewes immunized against PGF2α investigated the effects of exogenous PGF2α on the preovulatory follicle and the effects of PGF2α and oestradiol benzoate on corpus luteum function. Four weeks prepartum, 39 ewes bred to lamb during seasonal anoestrus received either 5 mg PGF2α-ovalbumin conjugate (n = 20; immunized) or ovalbumin (n = 19; control) in Freunds complete adjuvant. Treatments were repeated on day 5 post partum with reagents emulsified in Freunds incomplete adjuvant. On day 17 post partum, ewes received 500 iu pregnant mares serum gonadotrophin (PMSG) and 48 h later 50 μg gonadotrophin-releasing hormone (GnRH). Laparoscopy was performed 36 h after GnRH to assess ovarian activity and ewes with recent ovulations were inseminated into the uterus. No immunized ewes ...
The purpose of this paper was to assess the effect of supplementation of royal jelly on the morphometry of the genital tract, superovulatory response and embryonic quality in adult rabbits. 36 rabbits were distributed in four groups (G), as follows: G1 (n=9): no supplementation with royal jelly, G2, 3 and 4 (n=9): supplementation with 10, 20 and 40mg/day of royal jelly. The superovulatory protocol consisted in the application of a dose of 40UI of Equine Chorionic Gonadotrophin, followed by a dose of UI of Human Chorionic Gonadotrophin, intramuscularly, 48 hours later, and submitted to natural cover. The animals were euthanasiated and the embryos collected 72 hours after copulation. There was no statistical difference for the analyzed variables (P,0.05). The average weight for the female genital tract was 10.89±0.38g; the average weight of right and left ovaries was 0.28±0.02g and the average gonadosomatic index rate was 0.02±0.0g. The number of total recovered structures was 9.2±1.4. The ...
In vitro fertilization (IVF) is the most common technique in assisted reproductive technology and in most cases the last resort for infertility treatment. It has four basic stages: superovulation, egg retrieval, fertilization, and embryo transfer. Superovulation is a drug-induced method to enable multiple ovula- tion per menstrual cycle and key component towards a successful IVF cycle. Although there are the general guidelines for dosage, the dose is not optimized for each patient, and complications, such as overstimulation, can occur. To overcome the shortcomings of this general system, a mathematical procedure is developed based theory of particulate processes like crystallization. The procedure can provide a customized model of this stage regarding the size distribution of eggs (follicles/ oocytes) obtained per cycle as a function of the chemical interactions of the drugs used and the conditions imposed on the patient during the cycle, which provide a basis for predicting the possible ...
Reproductive biotechnologies continue to be developed for genetic improvement of both river and swamp buffalo. Although artificial insemination using frozen semen emerged some decades back, there are still considerable limitations. The major problem appears to be the lack of efficient methods for estrus detection and timely insemination. Controlled breeding experiments in the buffalo had been limited and similar to those applied in cattle. Studies on multiple ovulation and embryo transfer are essentially a replica of those in cattle, however with inherent problems such as lower number of primordial follicles on the buffalo ovary, poor fertility and seasonality of reproduction, lower population of antral follicles at all stages of the estrous cycle, poor endocrine status and a high incidence of deep atresia in ovarian follicles, the response in terms of transferable embryo recovery has remained low with 0.51 to 3.0 per donor and pregnancy rates between 15 to 30%. In vitro production of buffalo ...

Embryo Cryopreservation: We will cryopreserve 2-cell embryos for heterozygous males mated to wild type females (for those strains amenable to this procedure) or 2-cell embryos for homozygous males mated to wild type or homozygous females. A project will be considered complete when 200 recoverable embryos are frozen for heterozygous x wild type lines and 100 recoverable embryos are frozen for homozygous lines. [A straw of 8-10 embryos is thawed for each freezing day and the % recovery is used to determine the total number of remaining recoverable embryos for that day]. We generally average 15 good morulae from a C57BL/6 or hybrid, non-transgenic, superovulated female that is mated to a fertile male. Due to increased mouse costs, investigators will be expected to purchase females for projects where the background strain is not maintained in-house ...
Embryo transfer in pigs has been performed surgically for a long time. However, a less invasive, non-surgical, procedure of embryo transfer could be a valuable tool for research (to study embryo survival and embryo-uterus interactions) and practical applications (export, prevention of disease transmission and prevention of long distance transport of animals). Therefore the aim of this study was to develop non-surgical procedures for the collection and transfer of porcine embryos which can be used for both practical and research purposes.. The development of these procedures was expected to give insight in factors which are important for embryonic survival. Non-surgical (trans-cervical) embryo collection appeared to be possible from sows with surgically resectioned uterine horns. On average 8 embryos were collected from these sows. After superovulation induction the average but variable yield was increased to 18 embryos. Non-surgical (trans-cervical) transfer of embryos to non-sedated recipients ...

The Garole is a prolific breed of microsheep that possesses the FecB gene, which increases ovulation rate. The purpose of this study was to compare embryo production by multiple ovulation in seven Garole ewes with that in seven normal size, non-prolific Malpura ewes, and assess the influence of the large body size of Awassi crossbred recipient ewes on the birth-weight of Garole lambs. Oestrus was synchronised with two intramuscular injections of 7.5 mg prostaglandin F(2alpha) administered 10 days apart. The donor ewes were superovulated by the use of pregnant mare serum gonadotrophin and follicle-stimulating hormone. The onset and duration of oestrus were similar in both breeds. The Garole donors had higher total mean (se) ovarian responses (15.6 [3.6] v 9.1 [2.3]), ovulation rate (13.6 [3.1] v 8.4 [2.2]) and produced more transferable embryos (6.0 [3.5] v 4.0 [0.9]) than the Malpura donors, but the differences were not statistically significant. Methods Rat antiserum, raised against a 60-70 kDa band of partially purified GnSAF, was immobilised on anti-rat IgG Dynabeads. In addition, a human antibody, derived from a phage display antibody library and expressed by CHO-K1 cells, was immobilised on protein L agarose beads. Both antibodies were then used for 15 consecutive immunopurification steps with excess human granulosa-luteal cell-conditioned medium (GCM). In both cases 2 M NaI was used to elute the bound GCM proteins. The eluted fractions were bioassayed for GnSAF and inhibin bioactivities and the proteins separated by 1-D and 2-D gel electrophoresis. Spots and bands of interest were excised from the gels and identified by mass spectroscopic peptide mass mapping ...
Donor Mare - Embryo transfer begins with breeding the donor mare to semen from the chosen stud. Bred as usual, the donor is monitored daily using ultrasound to establish the exact date of ovulation.. Recipient Mare - Recipient mares (the ones receiving the embryo) are also monitored with ultrasound, and synchronized to prepare them to receive the donated embryo around day 7 of pregnancy.. The Flush - Seven days after the ovulation date, the donor mares uterus is flushed. This timing maximizes the chance of recovering an embryo at the ideal age. The fluid from the uterus is run through a special filter designed to catch the embryo.. Embryo Recovery - The flushed embryo is identified under a microscope. A transferable embryo is between 6 and 8 days old. At 5 days, the embryo is not yet in the mares uterus and cannot be flushed. At 9 days, the embryo becomes too large and fragile to handle. The prime 7-day embryo is approximately ¼ to ¾ millimeters in diameter. It is visible to the ...
Abstract:. The Criolla breed is a local genetic resource, distributed in Patagonia, Argentina, whose primary production is meat. In the Criolla goat, efforts were made to locate productively superior males and to conserve their genetic material. Studies were carried on the feasibility of obtaining superior offspring through the implementation of embryo transfer programs. We assessed the fertilization rate and embryo production following AI with frozen semen or natural mating in Criolla goats subjected to a superovulation program. During the breeding season (May, 41° S), 26 Criolla goats were treated for oestrus with sponges (60 mg of medroxyprogesterone acetate, Progespon®, Syntex, Buenos Aires, Argentina) placed for 13 days. Goat donors were superovulated with a total of 80 mg of porcine (p)FSH (Folltropin V®, Bioniche, Ontario, Canada) every 12 h in 6 decreasing doses (18, 18, 14, 14, 8, and 8 mg) during the last 3 days of progestagen treatment. A dose of 125 μg of cloprostenol ...
All ovulation induction drugs work by either increasing the patients own FSH (oral meds including Clomid or Letrozole) or by providing exogenous (already made from an outside source) FSH (injectables like Gonal F, Puregon, Menopure/HMG). It is the injectable drugs that achieve superovulation. Injectables can also be used for a small percent of patients who are resistant to oral medications in order to achieve single ovulation (monofollicular development).. Careful monitoring and dose adjustment are mandatory for safe ovulation induction, especially when injectables are used. Proper timing of the ovulation trigger is mandatory for proper egg maturation, to deliver the sperm at the ideal time, or to time when to collect the eggs in cases of IVF treatment.. Proper monitoring and frequent dose adjustment with proper, timely communication are mandatory for safe and successful treatment.. Ovulation induction treatment is usually continued for 3-4 cycles. The success rate per cycle is in the range of ...
Most mammalian embryos implant on the uterine endometrium after hatching fromzona pellucida of the expanded blastocyst and pregnancy takes place. The blastocysts produceand control a variety of prostaglandins which activate a few proteolytic enzymes thatdissolve the zona pellucida of the embryos. In the present study, the goal was to investigatethe indomethacin and aspirin(inhibitors of cyclooxygenase pathway which regulates thepathway from arachidonic acid to prostaglandins), which can affect the hatching and implantationof the mouse embryos by the treatment of the different dose level of indomethacinand aspirin in the culture of mouse embryos. The female and male ICR mice, 6~8 weeks and were used for superovulation andmating and M16 was used as a basic culture medium. The above results can be summarized as following:1. Indomethacin seemed to inhibit the development of the mouse embryos and hatchingprocess because of inhibiting or blocking the activation of hatching related enzymes andhigh dose ...
For women to undergo a superovulation and to store their young eggs is, first of all, a kind of insurance. Women may still decide to conceive through sexual intercourse. There are, however, other advantages to storing and preserving eggs at an earlier age. It is now possible, for instance, to carry out genetic analyses on a single cell of an embryo that is fertilized outside the uterus before it is implanted in it. Such genetic analysis enables women to screen for certain birth defects and avoid using a defective embryo. For many fertile women who plan on having no more than one or two children, and are prepared to pay for such information, this would be a major incentive to pursue the IVF route to conception rather than ordinary coitus.. In my view, during the next few decades the number of women using this type of simplified IVF who do not have fertility problems will overtake those with impaired fertility. This is a crucial point that has been virtually ignored by critics of the wider use of ...

Generation and culture of PSCs. We induced superovulation in 6- to 8-week-old (C57BL/6J × DBA/2J)F1 (B6D2F1) mice by i.p. injection of 7.5 U pregnant mare serum gonadotropin (PMSG) and, 48 hours later, 7.5 U human choriogonadotropin (hCG). We harvested oviducts and collected cumulus oocyte complexes (COCs) 16 hours after hCG injection. Cumulus cells were removed with hyaluronidase (1 mg/ml for 10 minutes at room temperature). We subsequently activated oocytes with strontium chloride (10 mmol/l in Ca2+-free CZB medium in mmol/l: NaCl 82.7, KCl 4.68, KH2PO4 1.17, MgSO4 1.18, D-glucose 5.6, Na-lactate 30.1, EDTA2Na 0.1, NaHCO3 25, Na-pyruvate 0.62, glutamine 1; supplemented with 5 mg/ml BSA, and 100 U/ml penicillin and 100 μg/ml streptomycin [100 P/S]) for 6 hours and prevented second polar body extrusion with cytochalasin B (5 μg/ml). Blastocysts formed during culture in mineral oil-covered Ca2+-containing (1.71 mmol/l) CZB medium for 6 days. We transferred blastocysts onto mitomycin C (10 ...
Genetic improvement of farm animals is a prime concern over the years for researchers. Several reproductive technologies have been employed to achieve this. Assisted reproductive technologies like Artificial insemination, Superovulation, In vitro Fertilization, Embryo Transfer have been introduced to overcome reproductive problems, to increase the offspring from selected females and to reduce the generation intervals in farm animals. The progress achieved during the last few years in the assisted reproductive technologies field has been phenomenal. Artificial Insemination (AI) is the most effective method being used for the genetic improvement of animals. Reproductive capacity and efficiency has been improved tremendously since the introduction of artificial insemination. The development of cloning using various cells from the animal body has created opening of a fascinating scientific arena. These technologies have been propounded as saviors of indigenous livestock breeds. These alternative ...
Research on in vitro embryo production (IVP) in the equine is impeded by the limited availability of mature oocytes as the mare is mono ovulating and superovulation is still difficult (Dippert and Squires, 1994; Bezard et al., 1995; Alvarenga et al., 2001b). Despite recent improvement in IVM of equine oocytes, success rates of IVM in that species remain low in all culture media tested compared to other species (Goudet et al., 2000b). However, most studies have focused on the percentage of oocytes reaching the metaphase II stage (nuclear maturation) but few concentrated on the final oocyte competence as measured by its ability to develop into a blastocyst and further establish a pregnancy. Blastocyst production rate is influenced not only by culture environment but also by oocyte maturation conditions. Under in vitro culture conditions, oxidative modifications of cell components via increased ROS represent a major culture induced stress (Johnson and Nasr-Esfahani, 1994). Anti-oxidant systems can ...

A case-control study was conducted in 2013 to investigate the use of pituitary-derived hormones from sheep as a potential risk factor for the presence of atypical scrapie in Great Britain sheep holdings. One hundred and sixty-five holdings were identified as cases. Two equal sets of controls were selected: no case of scrapie and cases of classical scrapie. A total of 495 holdings were selected for the questionnaire survey, 201 responses were received and 190 (38.3 per cent) were suitable for analysis. The variables use-of-heat-synchronisation/superovulation and flock size were significantly associated with the occurrence of atypical scrapie. Farms with atypical cases were less likely (OR 0.25, 95 per cent CI 0.07 to 0.89) to implement heat synchronisation/superovulation in the flock than the control group. Atypical cases were 3.3 times (95 per cent CI 1.38 to 8.13) more likely to occur in large holdings (,879 sheep) than in small flocks (,164 sheep). If the ...

In humans, multi-fetal pregnancy occurs when a mother carries more than one fetus during the pregnancy. The most common multi-fetal pregnancy is twins, but mothers have given birth to up to eight children (octuplets) from a single pregnancy. Multiple fetusus can result from the release of multiple eggs or multiple ovulations, the splitting of a single fertilized egg, and fertility treatments such as in vitro fertilization (IVF) which involves the insertion of many fertilized eggs into the mothers uterus..
