E. coli and most other pathogens affecting humans (which are the most heavily studied microbes) tend to have evolved to grow best at the temperature of the human body, 37C. Other organisms, such as yeast, grow best at 32C (or even lower). Trying to grow a thermophylic archaea at 37C would kill it - they require temperatures often near boiling. Most moulds grow quite comfortable at 25C. So, it is important to remember the natural habitat of a microbe before deciding on a temperature at which to try and culture it. ...

Thermophilic, acidophilic chemolithoautotrophs are thought to be important microorganisms in the evolutionary history of Earth (37). Their ability to thrive in high-temperature environments with minimal requirements other than carbon dioxide and inorganic constituents suggests that these organisms are important primary producers in extreme environments. Acid-tolerant thermophiles have been found in a variety of geothermal springs, mud pots, and deep-sea vents (10, 25, 29, 37). Many of these organisms utilize reduced inorganic compounds, such as hydrogen, sulfide, elemental sulfur, thiosulfate, methane, or ferrous iron, as energy sources for metabolism (2, 7). The remarkable diversity of the morphologies of viruses found in terrestrial hydrothermal environments with temperatures ,80°C is unprecedented for aquatic ecosystems. The best-studied viruses from these habitats have been assigned to novel viral families: Fuselloviridae, Lipothrixviridae and Rudiviridae. However, so far no transport system for d-xylose could be identified, and whether pentose assimilation strictly relies on both the aldolase-dependent and the aldolase-independent pathways is not yet fully understood. In this study, we compared growth on complex, proteinaceous media in the absence or presence of d-xylose. RNA sequencing of S. acidocaldarius MW001 revealed only five highly upregulated transcripts in response to d-xylose. Obsidian Hydration Dating: Obsidian hydration dating is based on the fact that a fresh surface is created on a piece of obsidian in the tool manufacturing, or flintknapping, process. Obsidian contains about 0.2 percent water. When a piece of obsidian is fractured, atmospheric water is attracted to the surface and begins to diffuse into the glass. This results in the formation of a water rich hydration rind that increases in depth with time. The hydration process continues until the fresh obsidian surface contains about 3.5 percent water. This is the saturation point. The thickness of the hydration rind can be identified in petrographic thin sections cut normal to the surface and observed under a microscope. A distinct diffusion front can be recognized by an abrupt change in refractive index at the inner edge of the hydration rind. The Global Ocean Sampling database contains half as many 4-hydroxybutyryl-CoA dehydratase sequences as compared with those found for another key photosynthetic CO2-fixing enzyme, ribulose-1,5-bisphosphate carboxylase-oxygenase, indicating the importance of this enzyme in global carbon cycling. The assimilation of carbon dioxide (CO2) into organic material is quantitatively the most important biosynthetic process. We discovered that an autotrophic member of the archaeal order Sulfolobales, Metallosphaera sedula, fixed CO2 with acetyl-coenzyme A (acetyl-CoA)/propionyl-CoA carboxylase as the key carboxylating enzyme. In this system, one acetyl-CoA and two bicarbonate molecules were reductively converted via 3-hydroxypropionate to succinyl-CoA. This intermediate was reduced to 4-hydroxybutyrate and converted into two acetyl-CoA molecules via 4-hydroxybutyryl-CoA dehydratase. The key genes of this pathway were found not only in Metallosphaera but also in Sulfolobus, Archaeoglobus, and Cenarchaeum species.
Several archaeal species from the order Sulfolobales are interesting from the biotechnological point of view due to their biomining capacities. Within this group, the genus Acidianus contains four biomining species (from ten known Acidianus species), but none of these have their genome sequenced. To get insights into the genetic potential and metabolic pathways involved in the biomining activity of this group, we sequenced the genome of Acidianus copahuensis ALE1 strain, a novel thermoacidophilic crenarchaeon (optimum growth: 75 °C, pH 3) isolated from the volcanic geothermal area of Copahue at Neuquén province in Argentina. Previous experimental characterization of A. copahuensis revealed a high biomining potential, exhibited as high oxidation activity of sulfur and sulfur compounds, ferrous iron and sulfide minerals (e.g.: pyrite).
