The Global Ocean Sampling database contains half as many 4-hydroxybutyryl-CoA dehydratase sequences as compared with those found for another key photosynthetic CO2-fixing enzyme, ribulose-1,5-bisphosphate carboxylase-oxygenase, indicating the importance of this enzyme in global carbon cycling. The assimilation of carbon dioxide (CO2) into organic material is quantitatively the most important biosynthetic process. We discovered that an autotrophic member of the archaeal order Sulfolobales, Metallosphaera sedula, fixed CO2 with acetyl-coenzyme A (acetyl-CoA)/propionyl-CoA carboxylase as the key carboxylating enzyme. In this system, one acetyl-CoA and two bicarbonate molecules were reductively converted via 3-hydroxypropionate to succinyl-CoA. This intermediate was reduced to 4-hydroxybutyrate and converted into two acetyl-CoA molecules via 4-hydroxybutyryl-CoA dehydratase. The key genes of this pathway were found not only in Metallosphaera but also in Sulfolobus, Archaeoglobus, and Cenarchaeum species.

Acidianus two-tailed virus (ATV) infects crenarchaea of the genus Acidianus living in terrestrial thermal springs at extremely high temperatures and low pH. ATV is a member of the Bicaudaviridae virus family and undergoes extra-cellular development of two tails, a process that is unique in the viral world. To understand this intriguing phenomenon, we have undertaken structural studies of ATV virion proteins and here we present the crystal structure of one of these proteins, ATV. ATV forms tetramers in solution and a molecular envelope is provided for the tetramer, computed from small-angle X-ray scattering (SAXS) data. The crystal structure has properties typical of hyperthermostable proteins, including a relatively high number of salt bridges. However, the protein also exhibits flexible loops and surface pockets. Remarkably, ATV displays a new protein fold, consistent with the absence of homologues of this protein in public sequence databases.

This study was undertaken to test whether 100-200 mM Na-chloride enhances the chemical and bacterial leaching of chalcopyrite in shake flasks and stirred tank bioreactor conditions at mesophilic temperatures. Acidithiobacillus ferrooxidans, Acidithiobacillus thiooxidans and abiotic controls were employed for the leaching experiments. The most common problem occurring in sewerage is leaking, whose position and failure level is typically identified through closed circuit television (CCTV) inspection in order to facilitate rehabilitation process. This paper proposes a novel method of computer vision, morphological segmentation based on edge detection (MSED), to assist inspectors in detecting pipeline defects in CCTV inspection images. In addition to MSED, other mathematical morphology-based image segmentation methods, including opening top-hat operation (OTHO) and closing bottom-hat operation (CBHO), were also applied to the defect detection in vitrified clay sewer pipelines. The CCTV inspection images of the sewer system in the 9th district, Taichung City, Taiwan were selected as the experimental materials. The swamp eel, Monopterus albus, is an obligatory air-breathing teleost which can survive long period of emersion, has high environmental and tissue ammonia tolerance, and acclimate from fresh to brackish water. This study was undertaken to clone and sequence gdh expressed in the liver, intestine and brain of M. albus, to verify whether more than one form of gdh were expressed, and to examine the gdh mRNA expressions in these three organs in fish exposed to various adverse conditions using quantitative real-time PCR. Only one gdh gene sequence, consisted of a 133 bp 5 UTR, a CDS region spanning 1629 bp and a 3 UTR of approximately 717 bp, was obtained from the liver, intestine and brain of M. albus. The translated Gdh amino acid sequence from the liver of M. albus had 542 residues and was confirmed to be Gdh1a. It had sequence identity of |90% with Oncorhynchus mykiss Gdh1a, Salmo salar Gdh1a1, Bostrychus sinensis Gdh1a and Tribolodon hakonensis Gdh1a, and formed a monophyletic clade with B.

The extraction of metals from ores causes various environmental pollutions. Since Iran is located on the so-called copper belt and holds a significant share of the worlds copper mines and resources, reduction of pollution from these mines can have an important effect on the overall reduction of pollution. Copper processing methods include pyrometallurgy and hydrometallurgy. Pyrometallurgy is mainly used in high grade mines whereas hydrometallurgy process is used in lower grade mines. In low grade copper sulfide mines, hydrometallurgy processes are used which use a lot of energy to covert mineral deposits into oxide forms which are then leached using sulfuric acid, or are extracted using bioleaching process. In acidic leaching, a lot of environmental pollution is created. Bioleaching process is an environmentally-friendly method which is mainly used in mines where the common physicochemical methods are not profitable.
