Arachnomelia is a monogenic recessive defect of skeletal development in cattle. The causative mutation was previously mapped to a similar to 7 Mb interval on chromosome 5. Here we show that array-based sequence capture and massively parallel sequencing technology, combined with the typical family structure in livestock populations, facilitates the identification of the causative mutation. We re-sequenced the entire critical interval in a healthy partially inbred cow carrying one copy of the critical chromosome segment in its ancestral state and one copy of the same segment with the arachnomelia mutation, and we detected a single heterozygous position. The genetic makeup of several partially inbred cattle provides extremely strong support for the causality of this mutation. The mutation represents a single base insertion leading to a premature stop codon in the coding sequence of the SUOX gene and is perfectly associated with the arachnomelia phenotype. Our findings suggest an important role for ...

Recently, Guenter Schwarz and colleagues published an elegant study in the Biochemical Journal (2019) 476, 1805-1815 which combines kinetic and spectroscopic studies with protein engineering to provide a mechanism for sulfite oxidase (SO)-catalyzed nitrite reduction that yields nitric oxide (NO). This work is noteworthy as it demonstrates that (i) for NO generation, both sulfite and nitrite must bind to the same molybdenum (Mo) center; (ii) upon sulfite reduction, Mo is reduced from +6 (MoVI) to +4 (MoIV) and MoIV reduces nitrite to NO yielding MoV; (iii) the heme moiety, linked to the Mo-center by an 11 amino acid residue tether, gets reduced by intramolecular electron transfer (IET) resulting in MoV being oxidized to MoVI; (iv) the reduced heme transfers its electron to a second nitrite molecule converting it to NO; (v) the authors demonstrate steady-state NO production in the presence of the natural electron acceptor cytochrome c; (vi) Finally, the authors use protein engineering to shorten ...
An essential trace element. It helps regulate iron stores in the body and is a key component of at least three enzymes: xanthine oxidase, aldehyde oxidase and sulfite oxidase. These enzymes are involved with carbohydrate metabolism, fat oxidation and urine metabolism. The average adult has about 9mg of molybdenum concentrated mostly in the liver, kidney, adrenal glands, bones and skin. Molybdenum deficiencies are associated with esophageal cancer, sexual impotency and tooth decay ...

Product name: p-AcPhe(Acp). Synonym: (S)-3-(4-acetylphenyl)-2-aminopropanoic acid,4-Acetylphenylalanine,p-Acetylphenylalanine,4AF, L-4-acetylphe,. Catalog #: 8215112. Purity: 95%. CAS No.: 122555-04-8. Molecular Formula: C11H13NO3. Molecular Weight: 207.23. Compound ID: 6420123. Description: The incorporated UAA p-acetylphenylalanine (p-AcPhe) acts as a reactive chemical handle enabling site-specific direct covalent coupling to the Fab. The ketone reacts with hydrazines or aminooxy groups to produce a hydrazone or oxime adduct, respectively. Site-directed spin labeling of the unnatural amino acid p-acetylphenylalanine (p-AcPhe) using oxime based coupling chemistry is successfully applied to investigate human sulfite oxidase (hSO), a protein containing an essential cysteine residue, which impedes the use of thiol based coupling chemistry. Antibody Fab fragments with an unnatural p-acetylphenylalanine residue could be used to create immunoconjugates and multimers to improve upon existing mAb ...

SHANGHAI will list two to three state-owned enterprises this year to boost SOE mixed-ownership reform, and will provide 400 billion yuan (US$58 billion) to help SOEs develop advanced technologies, said
The encounter of myth and reality concerning the nature of Chinese SOEs, globally and within Canada, is the source of public unease, aggravated by its newness and cross-cultural factors. Yet caution was registered on overgeneralizing on both the character and the behaviour of Chinese SOEs, particularly assumptions of irretrievable incompatibilities of mission, governance and management style. Perceptions risk distorting public policy. To be certain, Chinese SOEs are different, for instance, in their executive recruitment and training practices. Governance practices remain opaque. Yet at least in their manner of doing business abroad, SOEs have trended to a more truly commercial orientation and to management evaluation that accents operating performance over political considerations. Efforts are evident to accommodate of Canadian business practices and public expectations and to promote community integration. These efforts must be reciprocated by the Canadian side in investment policy ...

In this Letter, we propose an intracorporeal illumination system for providing uniform and wide-field illumination during minimally invasive surgery. The illumination system is comprised of an Alexis wound retractor, a set of LEDs, and secondary optical elements (SOEs). The SOE was composed of a Fresnel lens and a total internal reflection lens, which was designed to improve the optical performance of the LED. The results of simulation demonstrate that the optical efficiency of each LED with an SOE could be increased from 33.6% to 82.9%. To avoid damage to human tissue by thermal effect, the number of LEDs with SOEs was optimized. The results indicate that our design to be applicable for practical surgery.. © 2014 Optical Society of America. Full Article , PDF Article ...

Реферат: Electron spin−echo envelope modulation (ESEEM) spectroscopy of phospholipids spin-labeled systematically down the sn-2 chain was used to detect the penetration of water (D2O) into bilayer membranes of dipalmitoyl phosphatidylcholine with and without 50 mol % cholesterol. Three-pulse stimulated echoes allow the resolution of two superimposed 2H-ESEEM spectral components of different widths, for spin labels located in the upper part of the lipid chains. Quantum chemical calculations (DFT) and ESEEM simulations assign the broad spectral component to one or two D2O molecules that are directly hydrogen bonded to the N−O group of the spin label. Classical ESEEM simulations establish that the narrow spectral component arises from nonbonded water (D2O) molecules that are free in the hydrocarbon chain region of the bilayer membrane. The amplitudes of the broad 2H-ESEEM spectral component correlate directly with those of the narrow component for spin labels at different positions down ...
Nuclear quadrupole resonance spectroscopy or NQR is a chemical analysis technique related to nuclear magnetic resonance (NMR). Unlike NMR, NQR transitions of nuclei can be detected in the absence of a magnetic field, and for this reason NQR spectroscopy is referred to as zero Field NMR. The NQR resonance is mediated by the interaction of the electric field gradient (EFG) with the quadrupole moment of the nuclear charge distribution. Because the EFG at the location of a nucleus in a given substance is determined primarily by the valence electrons involved in the particular bond with other nearby nuclei, the NQR frequency at which transitions occur is unique for a given substance. A particular NQR frequency in a compound or crystal is proportional to the product of the nuclear quadrupole moment, a property of the nucleus, and the EFG in the neighborhood of the nucleus. It is this product which is termed the nuclear quadrupole coupling constant for a given isotope in a material and can be found ...
Second-order scalar-relativistic Douglas-Kroll-Hess density functional calculations of the electric field gradient, including an analytic correction of the picture change error, were performed for 34 tin compounds of which molecular structures and Sn-119 Mossbauer spectroscopy parameters are experimentally known. The components of the diagonalized electric field gradient tensor, V-xx,V-yy, V-zz, were used to determine the quantity V, which is proportional to the nuclear quadrupole splitting parameter Delta E. The slope of the linear correlation plot of the experimentally determined Delta E parameter versus the corresponding calculated V data allowed us to obtain an absolute value of the nuclear quadrupole moment Q of Sn-119 equal to Q = 13.2 +/- 0.1 fm(2). This is about 11% larger than the picture-change-error-affected value and in good agreement with previous estimates of the picture change error in compounds of similar atomic charge. Moreover, despite the variety of the tin compounds ...
1. The activity of rat liver microsomal sulphite oxidase (EC 1.8.3.1) was increased several-fold on aging of microsomes, on delipidation by extraction with acetone or on solubilization with deoxycholate, suggesting its existence in a cryptic state. 2. In rat liver most of the sulphite oxidase was present in the nuclear fraction and only a small portion in the microsomes. 3. Microsomal sulphite oxidase activity was low in the developing embryo and increased rapidly after birth.. ...
A system for detecting a target substance within a class of explosives and narcotics containing quadrupolar nuclei through the use of nuclear quadrupole resonance (NQR). The system applies an RF signal to a coil to excite the substance under test. If the target material is present, an NQR signal will be picked up by the same coil. That signal is compared with known NQR signals in frequency and amplitude. A signal is displayed in an appropriate way if a threshold value of the NQR signal is equalled or exceeded. The coil is tuned by means of an auto-tune feature. Effective RFI shielding is provided to prevent external contaminating signals from being detected by the coil and to prevent RF signals from escaping from the scanner. The invention also includes the method for performing tests with the system.

The conventional understanding of pi stacking involves quadrupole interactions between delocalized electrons in p-orbitals. In other words, aromaticity should be required for this interaction to occur. However, several groups have provided contrary evidence, calling into question whether pi stacking is a unique phenomenon or whether it extends to other neutral, closed-shell molecules. In an experiment not dissimilar from others mentioned above, Paliwal and coauthors constructed a molecular torsion balance from an aryl ester with two conformational states.[15] The folded state had a well-defined pi stacking interaction with a T-shaped geometry, whereas the unfolded state had no aryl-aryl interactions. The NMR chemical shifts of the two conformations were distinct and could be used to determine the ratio of the two states, which was interpreted as a measure of intramolecular forces. The authors report that a preference for the folded state is not unique to aryl esters. For example, the cyclohexyl ...
An ssu2 mutation in Sacccharomyces cermisiae, previously shown to cause sulfite sensitivity, was found to be allelic to GRR1, a gene previously implicated in glucose repression. The suppressor rgt1, which suppresses the growth defects of grr1 strains on glucose, did not fully suppress the sensitivity on glucose or nonglucose carbon sources, indicating that it is not strictly linked to a defect in glucose metabolism. Because the Cln1 protein was previously shown to be elevated in grr1 mutants, the effect of CLN1 overexpression on sulfite sensitivity was investigated. Overexpression in GRR1 cells resulted in sulfite sensitivity, suggesting a connection between CLN1 and sulfite metabolism. Multicopy FZF1, a putative transcription factor, was found to suppress the sulfite sensitive phenotype of grr1 strains, but not the glucose derepression or aberrant cell morphology. Multicopy FZF1 was also found to suppress the sensitivity of a number of other unrelated sulfite-sensitive mutants, but not that of ...

Amongst the world-wide developed pulping technologies the kraft process is by far the most dominant process for the production of bleachable pulp grades. Because of the high strength properties of the produced pulps and the ability to pulp basically all hard- and softwoods the kraft process has outperformed the former leading acid sulfite process. However, pulping processes based on sulfite have also distinct advantages. Compared to the strongly alkaline kraft process, sulfite pulping is highly adjustable since it can be applied over a wide pH range and allows the production of various pulp grades. For example, the ASAM process developed by the work group of Professor Patt at the University of Hamburg received considerable attention in the 1980s - 90s due to the outstanding properties of the resultant pulps. Additionally the AS/AQ (alkaline sulfite anthraquinone) process was considered by some authors as a promising alternative to kraft pulping. But for several reasons, which shall not be discussed here
An estimated 22% of the worlds largest firms are now effectively under state control, this is the highest percentage in decades. These firms are likely to remain a prominent feature of the global marketplace in the near future. The upsurge of state-owned enterprises (SOEs) as global competitors has given rise to concerns related to a level playing field. Some business competitors and observers claim that preferential treatment granted by governments to SOEs in return for public policy obligations carried out at home can give SOEs a competitive edge in their foreign expansion. The OECD has taken a multidisciplinary approach, looking at the issue from the competition, investment, corporate governance and trade policy perspectives. The report aims to sort fact from fiction, and develop a stronger understanding, based on empirical evidence, on how to address growing policy concerns with regard to SOE internationalisation. The report concludes that although there is no clear evidence of systematic ...
