We have selected E. coli strains for sulfathiazole resistance and have shown that each contains a resistant DHPS. Two independently isolated mutant folP alleles each contained the same Pro64→Ser amino acid substitution. The site of the substitution lay within a highly conserved sequence block, -GGESTRPG-, which is identical among E. coli, Bacillus subtilis, Neisseria meningitidis, andStreptococcus pneumoniae (9, 26, 33). The Pro64→Ser mutation is distinct from two other mutations reported inE. coli folP, each of which was selected for sulfathiazole resistance and simultaneous temperature sensitivity. Both of thets mutations lie at amino acid position Phe28 but have different substitutions, Leu (9) and Ile (10). While the determination of differences in the kinetic constants of the Pro64→Ser enzyme is in process, we have not noted a significant difference in the specific activity of the DHPS present in crude extracts prepared from wild-type or mutant strains (data not shown).. Pro64 is in a ...

A short-acting sulfonamide with similar properties to sulfamethoxazole. Rarely administered systemically due to its toxicity (Toxnet 2009). Used topically along with other medications for the treatment of vaginal, dermatological, ocular, otic and gastrointestinal infections. Since the last update we have not found published data on its excretion in breastmilk. Sulfonamides are excreted in small amounts in breastmilk: 6 to 94 micrograms/ml, 1.6% of the total dose (Briggs 2017). Reports of adverse effects are rare. An old publication reports cases of diarrhea and rash in children breastfed by mothers who were being treated with sulfathiazole. (Von Freissen 1951). Sulfathiazole is absorbed in very small amounts through the dermis (Waud 1944) and the intestine (Lasca 2018). The small dose and poor plasma absorption of most topical ophthalmological, otic and dermatological preparations make it unlikely that significant amounts will transfer into breastmilk. Do not apply to the chest in
The effect of milling on the solid-state transitions of sulfathiazole polymorphs in the absence and presence of solvent and excipients was monitored by X-ray powder diffraction (XRPD), attenuated total reflectance infrared (ATR-IR) and near-infrared (NIR) spectroscopy. Sulfathiazole forms FII-FV undergo a transformation toward the metastable FI, which involves an intermediate amorphous stage upon milling at ambient temperature. Milling FIII with catalytic amounts of solvent converts FIII to FIV or to mixtures of FI and FIV depending on the solvent used. Pure FIV can be easily prepared from FIII by this method. The physical stability of pure sulfathiazole forms in the presence of different levels of relative humidity (RH) was also investigated. At low RH all sulfathiazole forms are stable but at RH levels above 70% FII, FIII and FIV remain stable while FI and FV transform to mixtures of FII and FIV without any apparent change in the external form of the crystals. Co-milling FIII with... ...
A. Generally. By this regulation, the Virginia Racing Commission prohibits the feeding or administration to a horse on race day of any bicarbonate-containing substance or other alkalinizing substance that effectively alters the serum or plasma pH or concentration of bicarbonates or carbon dioxide in the horse. B. Test values. A serum or plasma total carbon dioxide level exceeding 37.0 millimoles per liter constitutes a positive test. C. Testing procedure. The stewards or commission veterinarian may, at their discretion and at any time, order the collection of test samples from any horses present within the enclosure for determination of serum or plasma pH or concentration of bicarbonate, carbon dioxide, or electrolytes. Preracing testing or post-race testing may be done at a time and manner directed by the commission veterinarian. Whether prerace or post-race, the sample shall consist of at least two blood tubes taken from the horse to determine the serum total carbon dioxide concentration. If ...

Using a structure-based approach, several small molecule inhibitors of POT1 have been designed to affect telomere integrity by disrupting the binding interaction of human POT1 with its target DNA sequence thereby driving cancer cells into senescence/apoptosis. Using a range of computational tools, a suitable drug binding pocket in POT1 has been identified and the de novo design of a specific class of POT1 inhibitor was completed. Using this novel scaffold, a small focussed library of hit-like compounds were synthesised and screened in a new POT1 fluorescence polarisation displacement assay developed by scientists at the University of Nottingham. In total, over 90 small molecule inhibitors based on two different scaffolds: pyrido[1,2-a]pyrimidines and sulfathiazoles have been synthesized with some inhibitors effectively decreasing POT1-DNA binding between 10-54% at 100μM ligand concentration. The biological results have established that electron-withdrawing substituents on the pendent phenyl ...

Summary Atabrine cured a case of acute amebic hepatitis which received the drug orally, and a case of advanced amebic abscess of the liver to which, besides oral therapy, intrahepatic instillations of the drug were given. It failed to cure two other cases of advanced amebic liver abscess which received the drug by the oral route only. Inadequate dosage in one of the two latter cases, and secondary infection in the other probably contributed to the failure. In vitro studies on Endamoeba histolytica showed that atabrine is about 10 times more effective than chloroquine and about 40 per cent as effective as emetine. A dosage schedule for atabrine in hepatic amebiasis has been set up on a purely tentative basis.
Milk-containing hydrogels from 2-hydroxyethylmethacrylate (HEMA) and acrylamide (AAm) as p(HEMA)/whole milk, p(HEMA)/semi-skimmed milk, p(HEMA)/skimmed milk, p(AAm)/whole milk, p(AAm)/semi-skimmed milk, p(AAm)/skimmed milk were synthesized by conventional redox polymerization technique using corresponding monomers and milk as solvents. The aim of this study was to investigate the use of milk-containing hydrogels as a nutritional medium, e.g., up 124 days for enhanced or longer cryostorage media for various microorganisms such as Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, and Bacillus subtilis, and fungi, Aspergillus niger, Aspergillus fumigates, and a yeast Candida albicans. It was shown that milk hydrogels can be used as nutritional media for the growth of these microorganisms, whereas the control hydrogels cannot be used for the same purpose. The survival rates of p(HEMA)/whole milk and p(AAm)/whole milk hydrogels for E. coli after 124 days were found to exceed those of ...
The nucleotide sequence of a small plasmid, designated pRF-1, isolated from Salmonella enterica serovar Choleraesuis, was determined. We identified seven open reading frames (ORFs) encoded by 6066 nucleotides with a total G + C content of 53.6%. Analysis of the complete nucleotide sequence revealed a replicon of pRF-1 to have high similarity to the p15A origin of replication, with a possible cer-like region. ORF1, which is composed of 816 nucleotides, shows a high degree of similarity to dihydropteroate synthetase encoded by the sulII gene from plasmids in several enteropathogenic bacteria, which functions as the sulfonamide resistance determinant. In fact, Salmonella and Escherichia coli strains carrying pRF-1 were found to show strong resistance to sulfathiazole, suggesting that orf1 is a functional gene. Four of seven ORFs were found to encode putative proteins of unknown function ...
1. The literature on conditions affecting the activity of proteolytic enzymes has been reviewed.. 2. Experimental data on the control of the activity of trypsin, leucoprotease, papain, serum antiprotease, leucopeptidase, and pancreatic peptidase have been presented. These data indicate that:. (a) The polymorphonuclearleucocytes of the cat contain abundant proteinase and peptidase active at neutral pH; those of the rabbit lack proteinase active at neutral pH.. (b) Reducing agents, including several biologically important thiol-sulfhydryl compounds and ascorbic acid, inhibit the activity of leucoprotease and trypsin. For each reductant the degree of inhibition is proportional to the reducing capacity of the medium.. (c) p-Aminobenzoic acid, sulfonamides (especially sulfathiazole), and many diphenyl sulfones inhibit the activity of leucoprotease.. (d) Serum, plasma, several heavy metals, ammonium salts, asparagine, thiourea, heparin, glutamic acid, tyrothricin, calcium chloride, and bile salts and ...

Snowflakey background details: This weekend I was at a Korean spa, and after a thorough scrubbing I was given a milk-containing rinse to splash on my face a few times. This resulted in a spectacular display of red splotches, hot to the touch, all over my face and chest (where the milk hit me.) Even one of my eyes became pink/bloodshot. After I saw the splotches develop I used soap and water to wash my face off, and after that and some time in the dark dry sauna (where I was hiding, because I looked scary) I recovered. It subsided in about 25-40 minutes, and I was in the clear by ~2 hours later ...

SUMMARY. An 18-year-old white man with congenital agammaglobulinemia that was probably sex-linked developed clinically and pathologically typical regional enteritis. The association of these diseases provides evidence that antibody is not directly involved in the pathogenesis of regional enteritis and that plasma cells, a usual histologic feature of regional enteritis, are not essential. Antibody lack probably does not predispose to regional enteritis, and cellular immunity may be important in its pathogenesis. ...
A pressurized liquid extraction (PLE) method was developed for the quantification of five sulfonamide antibiotics (sulfadiazine, sulfadimethoxine, sulfamethazine, sulfamethoxazole, and sulfathiazole) in aged soil samples. To account for sequestration effects the extraction was optimized using a composite grassland soil sample collected 11 days after the application of manure containing these substances. The optimized method uses a mixture of buffered water (pH 8.8) and acetonitrile (85:15) as solvent for the extraction at 200 degrees C and 100 bar during five min. The most important parameter for the extraction efficiency was temperature whereas the pH of the extraction solvent did hardly influence extraction efficiency between pH 4.1 and 8.8. A temperature increase from 100 to 200 degrees C improved the extraction efficiency up to a factor of six for aged residues in soils. In contrast, no temperature dependence was observed during short-term spike experiment. After 90 min exposure in these ...
To investigate the role of cross-infection in nonepidemic nosocomial bacteriuria in a large, university-affiliated hospital, we identified in adult patients admitted over an 11-week period all cases caused by organisms of the same genus, species, and antimicrobial susceptibility and clustered by date of onset and hospital ward. Further laboratory studies were conducted to verify clustering. Among the 3452 patients studied, 194 cases of nosocomial bacteriuria were identified; 49 appeared clustered by epidemiologic evidence. Additional laboratory tests verified clustering in 30 cases (15.5%). We found that 90% of clustered and 76% of nonclustered cases had had previous urinary catheterization; Pseudomonas aeruginosa, Serratia marcescens, and Citrobacter freundii often caused clustered infection while Escherichia coli predominated in nonclustered cases; and resistance to gentamicin, sulfathiazole, and carbenicillin was significantly greater for pathogens from clustered cases than for nonclustered ...

A dihydropteroate synthase inhibitor is a drug that inhibits the action of dihydropteroate synthase. Most are sulfonamides.. In bacteria, antibacterial sulfonamides act as competitive inhibitors of the enzyme dihydropteroate synthase, DHPS. DHPS catalyses the conversion of PABA (para-aminobenzoate) to dihydropteroate, a key step in folate synthesis.[1] Folate is necessary for the cell to synthesize nucleic acids (nucleic acids are essential building blocks of DNA and RNA), and in its absence cells will be unable to divide. Hence the sulfonamide antibacterials exhibit a bacteriostatic rather than bactericidal effect.[2]. ...

Myeloid angiogenic cells (MACs) play a key role in endothelial repairing processes and functionality but their activity may be impaired by the lipotoxic effects of some molecules like stearic acid (SA). Among the dietary components potentially able to modulate endothelial function in vivo, (poly)phenolic compounds represent serious candidates. Here, we apply a comprehensive multidisciplinary approach to shed light on the prospects of Bergamot (Citrus bergamia), a citrus fruit rich in flavanones and other phenolic compounds, in the framework of lipotoxicity-induced MACs impairment. The flavanone profile of bergamot juice was characterized and 16 compounds were identified, with a new 3-hydroxy-3-methylglutaryl (HMG) flavanone, isosakuranetin-7-O-neohesperidoside-6″-O-HMG, described for the first time. Then, a pilot bioavailability study was conducted in healthy volunteers to assess the circulating flavanone metabolites in plasma and urine after consumption of bergamot juice. Up to 12 flavanone phase II
The white tiger or bleached tiger is a pigmentation variant of the Bengal tiger, which is reported in the wild from time to time in the Indian states of Assam, West Bengal and Bihar in the Sunderbans region and especially in the former State of Rewa. Such a tiger has the black stripes typical of the Bengal tiger, but carries a white or near-white coat. The white Bengal tigers are distinctive due to the color of their fur. The white fur caused by a lack of the pigment pheomelanin, which is found in Bengal tigers with orange color fur. When compared to Bengal tigers, the white Bengal tigers tend to grow faster and heavier than the orange Bengal tiger. They also tend to be somewhat bigger at birth, and as fully grown adults. White Bengal tigers are fully grown when they are 2-3 years of age. White male tigers reach weights of 200 to 230 kilograms and can grow up to 3 meters in length. As with all tigers, the white Bengal tigers stripes are like fingerprints, with no two tigers having the same ...
The influence of a variety of metabolic factors on DNA excision repair was examined in studies involving measurements of damaged base removal, repair replication, and size of the resulting resynthesized regions or repair patches.;First, ultraviolet light (UVL)- and dimethylsulfate (DMS)-induced excision repair was examined in quiescent and lectin-stimulated bovine lymphocytes. Upon mitogenic stimulation, UVL-induced repair increased by a factor of 2 to 3, and reached this maximum 2 days before the onset of DNA replication. However, DMS-induced repair increased sevenfold in parallel with DNA replication. Repair patch sizes were smaller for DMS-induced damage reflecting patches of 7 nucleotides in quiescent lymphocytes compared to 20 nucleotides induced by UVL. The patch size increased during lymphocyte stimulation until one day prior to the peak of DNA replication when patch sizes of 45 and 35 nucleotides were produced in response to UVL- and DMS-induced damage, respectively. At the peak of DNA
Purified penicillinase, in gram quantities, has been prepared from Escherichia coli strain W3310 by using methods developed to handle large amounts of material. The final product had a specific enzyme activity of 3.08 units/μg of protein, which was over twice as high as that reported previously (Datta & Richmond, 1966). The purified enzyme was similar to that from E. coli strain TEM, but different in molecular weight and some other respects. The differences observed may be a result of the greater purity obtained.. ...

The main reason for the relatively low specific value of carbon dioxide emissions in Stuttgart can be found in the conversion sector. Due to the very high percentage of nuclear energy (currently about 90 %), power generation contributes with only about 11 % to the total carbon dioxide emissions, even if power generation has a share of about 25 % in the total final energy consumption in Stuttgart ...

If topical or IL therapy fails, hydroxychloroquine (HCQ), or another antimalarial should be tried. The usual dose for HCQ is 200-400 mg/day. If this fails, atabrine (quinacrine) 100 mg/day may be added. Chloroquine is also efficacious and may be tried. This approach is effective for the majority of cases. Smoking cuts in half the response to antimalarials [JAAD 2015;72;634]. For patients seemingly resistant to HCQ, the blood levels should be checked. Increasing the dose to get blood levels above 750 ng/ml significantly improved results in 81% of patients analyzed [JAAD 2016;74;693]. For patients with side effects to one antimalarial, switching to another is usually well tolerated [JAAD 2018;78;107-114.e1].. ...

The Common Garter Snake has been said to be the best-studied snake in the world. Among other studies, its interaction with the Rough-skinned Newt is perhaps the most interesting. These newts are extremely poisonous, the tetrodotoxin in their body enough to kill anything that tries to eat them (including even large mammals). Some populations of garter snakes have evolved physiological defenses against this toxin and can eat newts freely. Other populations cannot. This is said to be an arms race between predator and prey, the prey evolving defenses and the predator evolving mechanisms of countering them ...
