The single dose oral toxicity of succinic anhydride in Sprague-Dawley rats was reported as 1794.9 mg/kg bw for males and females. The acute dermal median lethal dose for succinic anhydride was found to exceed the limit dose level in rats, ,2000 mg/kg bw. Clinical signs included reduced appetite and activity, increasing weakness, collapse and death inalbino rabbits following acute dermal administration of maleic anhydride. The LD50 is between 1600 and 2600 mg/kg/day. Macroscopic evaluation showed enlarged gall bladder, discoloration of spleen and kidneys, and gastrointestinal inflammation. Similar results would be expected with succinic anhydride Acute inhalation testing is not required. The results of the granulometry indicate the L10 value for particle size was 377 µm and the L50 was greater than 1100 µm. On the basis of these results only a very small part of the particles in the batch of succinic anhydride may be smaller than 100 µm and may be respirable. The lager part of the particles ...
Succinic anhydride can be expected to have a low potential for bioaccumulation (due to low log Pow). Information regarding the toxicokinetics and percutaneous absorption of cyclic anhydrides is available in the published literature. Toxicokinetics data on hexahydrophthalic anhydride and phthalic anhydride indicate that cyclic anhydrides are readily hydrolysed to the corresponding dicarboxylic acid, which is mainly excreted in the urine and to a lesser extent in expired air after exposure via the inhalation route. Cyclic anhydrides share structural and physicochemical properties such that following inhalation exposure. The hydrolysis product of succinic anhydride is succinic acid, an endogenous substance and food ingredient that is expected to be effectively excreted in urine. A separate toxicokinetics study on succinic anhydride is not proposed and cannot be justified based upon animal welfare considerations. Percutaneous absorption of cyclic anhydrides was investigated using hexahydrophthalic ...

The development and the utilization of starch as a packaging material which is eco-friendly, renewable and biodegradable have increased in recent years. In this study, cassava starch was modified into composites by adding glycerol as plasticizer and oil palm empty fruit bunches (OPEFB) microcrystalline cellulose (MCC) as fillers. The MCC was prepared via the pulping process of OPEFB and acid hydrolysis. The composites were cast into films by varying glycerol and MCC compositions. The effect of glycerol and OPEFBMCC loading on tensile properties and water absorption of composite was investigated. The results showed that MCC content at 10% created denser composite surface and enhanced the interfacial adhesion which led the tensile strength to increase but the elongation at break decreased. The filler loading also reduced the diffusion of water molecules and water absorption. However, the addition of glycerol in cassava starch composite to any MCC composition lowered the tensile strength but ...
Influence of Acid Hydrolysis Reaction Time on the Isolation of Cellulose Nanowhiskers from Oil Palm Empty Fruit Bunch Microcrystalline Cellulose
Non-covalent binding of avidin to biotinylated erythrocytes results in complement-dependent haemolysis. Biotinylated erythrocytes, as well as native cells, are not lysed by complement. Complement activation requires a tight contact between avidin and the erythrocyte membrane, since avidin does not in itself activate complement and does not inhibit lysis of sensitized sheep erythrocytes. The efficiency of haemolysis depends on avidins surface density. When the avidin concentration in the reaction mixture is less than 15 micrograms/ml, erythrocyte lysis is not induced. However, the attachment of biotinylated antibodies to avidin-carrying erythrocytes decreases dramatically. Acylation of avidin with succinic anhydride strongly decreases its ability to induce complement-dependent haemolysis. However, the ability of avidin to cross-link the biotin-containing structures decreases after acylation. A 50% modification of avidin by succinic anhydride (pI about 7.0) allows preparation of ...

Brevetoxin-1 (BTX-1), a marine toxin mostly produced by the dinoflagellatae Karenia brevis, has caused the death of marine organisms and has had numerous toxicological effects on human health. Hence, it is very necessary to develop a rapid, economical, and reliable immunoassay method for BTX-1 detection. In this study, two kinds of complete antigen were synthesized using the succinic anhydride and isobutyl chloroformate two-step methods. Conjugate BTX-1-OVA was used as an antigen for mice immunization, and BTX-1-BSA for measuring the titer of the produced antibodies. A hybridoma cell line 6C6 stably secreting monoclonal antibody (mAb) against BTX-1 was obtained by fusing SP2/0 myeloma cells with the spleen cells from the immunized mouse. The hybridoma 6C6 was injected into the abdomen of BALB/c mice to obtain ascites, and the anti-BTX-1 mAb was harvested from ascites by precipitation with caprylic acid/ammonium sulfate (CA-AS). The anti-BTX-1 mAb was identified as an IgG1 subtype, and the cross

Fibres from different parts of empty fruit bunch, which is a major solid waste from oil palm processing, were subjected to different pretreatments and characterised for variability in length and diameter, mechanical performance, and proximate and trace element composition. Morphology and surface composition of the fibres were determined using scanning electron microscopy with energy dispersive X-ray. The fibres were further treated with KOH-boric acid and characterized by Fourier transform infrared spectroscopy, X-ray diffraction, and scanning electron microscopy. Fibre yield was higher for spikelet than stalk. Fibres from stalk were generally larger in diameter and showed significant differences in potassium and galacturonic acid content, strength, and rigidity. Scanning electron microscopy confirmed the widespread occurrence of silica bodies as well as significant differences in the microstructure of stalk and spikelet fibres. Stalk fibres showed a greater level of porosity than spikelet fibres in the
The pretreatment of empty fruit bunch (EFB) was conducted using an integrated system of IL and cellulases (IL-E), with simultaneous fermentation in one vessel. The cellulase mixture (PKC-Cel) was derived from Trichoderma reesei by solid-state fermentation. Choline acetate ChoOAc was utilized for the pretreatment due to its biocompatibility and biodegradability. The treated EFB and its hydrolysate were characterized by the Fourier transform infrared spectroscopy, scanning electron microscopy, and chemical analysis. The results showed that there were significant structural changes in EFB after the treatment in IL-E system. The sugar yield after enzymatic hydrolysis by the PKC-Cel was increased from 0.058 g/g of EFB in the crude sample (untreated) to 0.283 and 0.62 ± 06 g/g in IL-E system after 24 and 48 h of treatment, respectively. The objective of this study was to identify the performance of empty fruit bunch (EFB) to assess its use as potential adsorbent in treat ammonia nitrogen in natural rubber wastewater. The adsorption method was employed to investigate the effect of a number of factors, namely, adsorbent dosage, pH, shaking speed and contact time. The results revealed that the removal of ammonia nitrogen up to 79.5% from natural rubber wastewater. The adsorption efficiency of EFB fibre was maximum at 3.5 g dosage, pH 7, 150 rpm shaking speed and 120 min contact time. The results proved that EFB fiber are good alternative low cost adsorbent for the removal of ammonia nitrogen from natural rubber wastewater. The Langmuir isotherm (R2=0.9923) described ammonia nitrogen adsorption slightly better than the Freundlich isotherm (R2 = 0.9450), suggesting a monolayer adsorption behaviour of the adsorption processes ...

In this study, polysaccharides of marine origin (agar, alginate and κ-carrageenan) were used to embed nutrients to fabricate biocomposites to be employed in animal feeding. The consistency of biocomposites in water has been evaluated up to 14 days, by several methods: swelling, nutrient release and granulometric analysis. Biocomposites were produced with varying percentages of nutrients (5%-25%) and polysaccharides (1%-2%-3%). All possible biopolymer combinations were tested in order to select those with the best network strength. The best performing biocomposites were those manufactured with agar 2% and nutrients 10%, showing the lowest percentage of water absorption and nutrient release. Biocomposites made of agar 2% and nutrients 10% were the most stable in water and were therefore used to analyze their behavior in water with respect to the release of quercetin, a phenolic compound with demonstrated high antibacterial and antioxidant activities. The leaching of such molecules in water was therefore

The invention is directed to enhanced methods for detecting an analyte of interest in situ, by immunoassay, or by hybridization comprising binding an enzyme-labeled conjugate molecule to an analyte of interest in the presence of a redox-inactive reductive species and a soluble metal ion. The enzyme catalyzes the conversion of the inactive reductive species to an active reducing agent, which in turn reduces the metal ion to a metal atom thereby providing an enhanced means of detecting the analyte via metal deposition.
Laser direct metal deposition has wide applications in the areas of rapid manufacturing, surface coating and component repair. Defects of interlayer and intralayer porosity are often observed in laser deposited structures and repaired components. Interlayer porosity can be controlled to some extent by adjusting processing parameters, but there is still disagreement as to the source of intralayer porosity and whether processing conditions, parameters or initial powder materials are the dominant causal factors. In this work, two samples of Ti-6Al-4V powder, prepared using gas-atomization and plasma-atomization, were analyzed using laser diffraction and micro computed tomography with the Xradia Micro XCT machine for any initial porosity content. A 1.5 kW diode laser with a coaxial deposition head was then used to deposit a number of thin-wall structures with the different powders at different processing parameters. The deposited structures were characterized using scanning electron microscopy (SEM) ...

As an expert in the closing of nutrient cycles, Lindeboom focusses on the empty fruit bunches that are the main source of organic waste during palm oil production. These fruit bunches are roughly the size of a banana bunch, but as the fruits are much smaller, half of that is organic waste. Part of it is usually burnt to make steam for the sterilisation of the fruits, but still some 80% remains. These empty fruit bunches could be an excellent source of biogas. It would need some kind of pre-treatment to break down the lignocellulose before the biogas producing microorganisms could do their work, Lindeboom explains. Al-Muraisy is looking at natural resources such as concentrated solar power (CSP) to break down the chemical bonds for that. Or you could use fungi that produce enzymes such as laccase and peroxidase, depending on local circumstances. Concentrated Solar Power, which is abundant in Malaysia and is high in total solar irradiance, is a novel technology in the palm oil industry. The ...
The automotive sector has taken a keen interest in lightweighting as new required performance standards for fuel economy come into place. This strategy includes parts consolidation, design optimization, and material substitution, with sustainable polymers playing a major role in reducing a vehicles weight. Sustainable polymers are largely biodegradable, biocompatible, and sourced from renewable plant and agricultural stocks. A facile way to enhance their properties, so they can indeed replace the ones made from fossil fuels, is by reinforcing them with fibers to make composites. Natural fibers are gaining more acceptance in the industry due to their renewable nature, low cost, low density, low energy consumption, high specific strength and stiffness, CO2 sequestration potential, biodegradability, and less wear imposed on machinery. Biocomposites then become a very feasible way to help address the fuel consumption challenge ahead of us. This book, entitled Biocomposites in Automotive ...
Cellular metabolites, such as acyl-CoA, can modify proteins, leading to protein posttranslational modifications (PTMs). One such PTM is lysine succinylation, which is regulated by sirtuin 5 (SIRT5). Although numerous proteins are modified by lysine succinylation, the physiological significance of lysine succinylation and SIRT5 remains elusive. Here, by profiling acyl-CoA molecules in various mouse tissues, we have discovered that different tissues have different acyl-CoA profiles and that succinyl-CoA is the most abundant acyl-CoA molecule in the heart. This interesting observation has prompted us to examine protein lysine succinylation in different mouse tissues in the presence and absence of SIRT5. Protein lysine succinylation predominantly accumulates in the heart whenSirt5is deleted. Using proteomic studies, we have identified many cardiac proteins regulated by SIRT5. Our data suggest that ECHA, a protein involved in fatty acid oxidation, is a major enzyme that is regulated by SIRT5 and ...

Post-Translational Modification (PTM) is a biological reaction which contributes to diversify the proteome. Despite many modifications with important roles in cellular activity, lysine succinylation has recently emerged as an important PTM mark. It alters the chemical structure of lysines, leading to remarkable changes in the structure and function of proteins. In contrast to the huge amount of proteins being sequenced in the post-genome era, the experimental detection of succinylated residues remains expensive, inefficient and time-consuming. Therefore, the development of computational tools for accurately predicting succinylated lysines is an urgent necessity. To date, several approaches have been proposed but their sensitivity has been reportedly poor. In this paper, we propose an approach that utilizes structural features of amino acids to improve lysine succinylation prediction. Succinylated and non-succinylated lysines were first retrieved from 670 proteins and characteristics such as ...

Additive manufacturing (AM) techniques such as Selective Laser Melting (SLM) or Direct Metal Deposition (DMD) have been identified as an attractive option for the manufacture of novel functional components in comparison with conventional manufacturing techniques. They allow building components with complex 3D geometries layer by layer. The very rapid consolidation of the base material in a small material volume and the achieved high solidification rates allow for the manufacture of components containing meta-stable materials which tend to decompose in conventional casting or sintering processes. At the same time, these conditions may lead to complex out-of-equilibrium microstructures, pronounced element segregation and crack formation in the bulk alloy. Despite AM of metallic materials is a commonly established processing technology, the materials science in AM is only at its infancy. We therefore try to obtain fundamental knowledge of reactions and phase transformations in rapidly solidified ...

N2 - In the present work, two types of treatment namely alkaline hydrolysis and glycerolysis have been conducted for lignin extraction from oil palm empty fruit bunch (EFB) fiber. Lignin has been retrieved from two sequential methods, which was the klason lignin from residue and lignin from precipitation of the filtrate. Alkaline hydrolysis was performed using 10% NaOH solution at room condition. This has extracted 13.0 % lignin. On the other hand, glycerolysis was carried out using 70% glycerol catalyzed with 5% of 1 M NaOH at 60-70 °C. This has successfully extracted 16.0% lignin. The SEM micrographs exhibited some physical changes on the surface where the impurities and waxes have been removed, exposing the, lumen. Besides that, FTIR analysis was conducted on untreated EFB, treated EFB and extracted lignin. Delignification ...
Titanium dioxide (TiO2) is added in sunscreens due to its ability to absorb ultraviolet (UV) light. However, upon irradiation of UV light, reactive oxygen species particularly hydroxyl radical which can damage human skin will be generated. In this study, lignin/TiO2 composites were employed to quench the hydroxyl radicals generated by the TiO2. The lignin was extracted from oil palm empty fruit bunch (OPEFB) via kraft and soda pulping processes. The kraft lignin composite was labelled as KL/TiO2 whereas the soda lignin composite was labelled as SL/TiO2. The lignins and the composites were characterized by FTIR, UV spectroscopy, 13C NMR, SEM, EDX, and XRD. The relative hydroxyl radical production of composites and TiO2 were compared through photo-oxidation of coumarin to 7-hydroxycoumarin as a test medium. The effect of types and amounts of lignin used were studied. The KL/TiO2 composite showed the least radical production due to higher phenolic hydroxyl content of kraft lignin. The activity of the
The skin sensitising potential of butyl acrylate was investigated in a Local Lymph Node Assay (LLNA) conducted according to OECD guideline 429 and GLP regulations (BAMM 2006). The application of the test substance at concentrations of 1, 2.5, 5, 10, or 25 % w/v in acetone : olive oil (4:1) resulted in an increase in isotope incorporation which was greater than 3-fold at the 25 % w/v concentration. Consequently, the test substance was shown to be a potential skin sensitiser. The concentration giving rise to a 3 fold increase in lymphocyte proliferation (EC3) was calculated to be 11.2 % w/v (2800 μg/cm2), indicative of a sensitiser of weak potency. Butyl acrylate was tested in guinea pigs and mice using a variety of methods and gave a positive response in most of the tests. Butyl acrylate (purity , 99%) was positive in the Guinea Pig Maximization test. Positive results were noted in 7 out of 10 animals at challenge and re-challenge (Van der Walle, 1982). Butyl acrylate was tested by Parker and ...

The thermal and structural properties were attributed to the nonstructural and structural constituents of the fibers, respectively.. ...
The quality and efficiency of laser-aided direct metal deposition largely depends on the powder stream structure below the nozzle. Numerical modeling of the powder concentration distribution is complex due to the complex phenomena involved in the two-phase turbulence flow. In this paper, the gravity-driven powder flow is studied along with powder properties, nozzle geometries, and shielding gas settings. A 3-D numerical model is introduced to quantitatively predict the powder stream concentration variation in order to facilitate coaxial nozzle design optimizations. Effects of outer shielding gas directions, inner/outer shielding gas flow rate, powder passage directions, and opening width on the structure of the powder stream are systematically studied. An experimental setup is designed to quantitatively measure the particle concentration directly for this process. The numerical simulation results are compared with the experimental data using prototyped coaxial nozzles. The results are found to ...

The structural and mechanical characteristics of polymeric biocomposites based on calcium lactate (CL) and either partially (88 mol%) or fully (98 mol%) hydrolyzed poly(vinyl alcohol) (PVA) were studied using optical microscopy, Fourier transform infrared spectroscopy equipped with attenuated total reflectance accessory, water content determination, and differential scanning calorimetry. In addition, the moisture absorption effect on the mechanical properties of the biocomposites was tested in this work. The results reveal that CL is a suitable modifier for both types of PVA. However, a more efficient enhancement of uniform shape and size distribution of CL particles within the PVA matrix was noticed in fully hydrolyzed matrix at low water contents. It is in agreement with glass transition temperature?s observations. The samples conditioned at 50% of relative humidity (RH) showed decrease in E modulus and tensile strength in comparison with the material stored below 20% RH. Nevertheless, the ...

Post-translational modifications (PTMs) play an important role in modulating diverse cellular processes, which are dynamic and reversible modifications of proteins during or after protein biosynthesis [1]. PTMs can change protein functions by introducing new functional groups such as acetyl, phospho, ubiquityl, succinyl and methyl groups. Among these changes, acetylation of lysine has been extensively studied in both eukaryotes and prokaryotes [2, 3]. Similar to lysine acetylation, lysine succinylation is one of the newly discovered PTMs that has been found in both eukaryotes and prokaryotes [1]. As one important PTM of proteins, lysine succinylation was defined as the transfer of a succinyl group to a lysine residue of a protein molecule [4].. Lysine succinylation was firstly discovered in histone proteins, and its role has been investigated in the regulation of gene transcription [5]. In addition to histones, other proteins in the nucleus, cytoplasm and mitochondria were also found to be ...

A new method for the determination of amino acids is presented. It combines established methods for the derivatization of primary and secondary amino groups with 9-fluorenylmethoxycarbonyl chloride (Fmoc-Cl) with the subsequent amino acid specific detection of the derivatives by LC-ESI-MS/MS using multiple reaction monitoring (MRM). The derivatization proceeds within 5 min, and the resulting amino acid derivatives can be rapidly purified from matrix by solid-phase extraction (SPE) on HR-X resin and separated by reversed-phase HPLC. The Fmoc derivatives yield several amino acid specific fragment ions which opened the possibility to select amino acid specific MRM transitions. The method was applied to all 20 proteinogenic amino acids, and the quantification was performedusing l-norvaline as standard. A limit of detection as low as 1 fmol/μl with a linear range of up to 125 pmol/μl could be obtained. Intraday and interday precisions were lower than10 % relative standard deviations for most of the ...

The protein human CCL2 has been synthesized using a combination of solid phase peptide synthesis (SPPS) and native chemical ligation (NCL). The thioester-peptide segment was synthesized using the sulfonamide safety-catch linker and 9-fluorenylmethoxycarbonyl (Fmoc) SPPS, and pseudoproline dipeptides were used to facilitate the synthesis of both CCL2 fragments. After assembly of the full-length peptide chain by NCL, a glutathione redox buffer was used to fold and oxidize the CCL2 protein. CCL2 was crystallized and the structure was determined by X-ray diffraction at 1.9-A resolution. This is probably one of the first crystal structures of a protein prepared using the sulfonamide safety-catch linker and NCL. ...
Alternatively, as shown in figure 5, the protective group of compound IX can be removed under conditions that are compatible with the Boc protecting group, e.g. by hydrogenation, when PG is a benzyl or benzyloxycarbonyl, or basic conditions, such as diethylamine, when PG represents fluorenylmethoxycarbonyl, to obtain the compound XVI. Other methods of selective removal of the protective groups can be found in the directory Greene. In this case, the connection of the XVI coincides with compound X, and PG is a Boc. In this case, the removal of the protective group from the XI-XII can be carried out in conditions similar to the conversion of II to XIV and IV V.. The synthesis methods described above in schemes 1-5, also can be performed using racemic Proline derivatives or derivatives of D-Proline instead of L-Proline. So about what atom, it is possible to obtain the compounds of formula I with an alternate stereochemistry.. In the following aspect the present invention relates to pharmaceutical ...

Bagasse has been used as reinforcing filler in different thermoplastic matrices such as poly(eth - ylene-co-vinyl acetate) (or EVA) [152-153], polyolefins [154-155] and starch-based biodegrad-able polyester [140-141, 156-157]. The effect of cultivar type and surface cleaning of the bagasse fiber on the tensile properties of the bagasse-EVA composites have been investigated [152]. The results suggested that blends of bagasse from various cultivars can be used for commercial applications of these composites. Also, the surface cleaning of the bagasse obtained from sugar mill was good enough to use the bagasse without further surface treatment. Another study on the impact behavior of the bagasse-EVA composites showed that the mechanical performance of this type of composites could be tailored by varying the bagasse volume fraction in order to reproduce the behavior of wood-based particleboards [153. ]. Luz et al. [154] explored the efficiency of two different processing methods, injection molding ...

If left unresolved, the following category 1 glitches will be icing on the cake of Lockheeds $400 billion quagmire (partial list via Defense News); When the F-35B vertically lands on very hot days, older engines may be unable to produce the required thrust to keep the jet airborne, resulting in a hard landing.. After doing certain maneuvers, F-35B and F-35C pilots are not always able to completely control the aircrafts pitch, roll and yaw.. Supersonic flight in excess of Mach 1.2 can cause structural damage and blistering to the stealth coating of the F-35B and F-35C.. Cabin pressure spikes in the cockpit of the F-35 have been known to cause barotrauma, the word given to extreme ear and sinus pain.. The spare parts inventory shown by the F-35s logistics system does not always reflect reality, causing occasional mission cancellations.. If the F-35A and F-35B blows a tire upon landing, the impact could also take out both hydraulic lines and pose a loss-of-aircraft risk.. Possible maneuvering ...

Vp/Vs models provide important complementary information to Vp and Vs models, relevant to lithology, rock damage, partial melting, water saturation, etc. However, seismic tomography using body-wave traveltime data from local or regional earth- quakes does not constrain Vp/Vs well due to the different resolution of Vp and Vs models, with the Vp models usually better constrained than Vs. Since surface- wave data are most sensitive to Vs, which leads to complementary strengths of body-wave data, we jointly invert body- and surface-wave data to better resolve the Vp/Vs models. In order to show the robustness of our joint inversion method, we compare the results to other approaches, including dividing Vp by Vs models, and Vp/Vs parameterization with body-wave or both body- and surface-wave data, using synthetic data and real data from the southern California plate boundary region. We confirm that Vp/Vs models from body-wave inversion obtained by divi- sion tend to include artifacts, even when both Vp ...
