Vasoactive intestinal peptide (VIP) immunoreactive secretomotor neurons in the submucous plexus are involved in mediating bacterial toxin-induced hypersecretion leading to diarrhoea. VIP neurons become hyperexcitable after the mucosa is exposed to cholera toxin, which suggests that the manipulation of the excitability of these neurons may be therapeutic. This study used standard intracellular recording methods to systematically characterize slow excitatory postsynaptic potentials (EPSPs) evoked in submucosal VIP neurons by different stimulus regimes (1, 3 and 15 pulse 30 Hz stimulation), together with their associated input resistances and pharmacology. Limited studies have examined Panx2 in the intestine, where it may have important physiological roles. The present study characterized Panx2 expression and localization in the human colon in health and disease states. Methods:|br|Immunofluorescence determined Panx2 localization and co-localization, and quantitative real-time PCR and Western blot determined gene and protein expression in ulcerative colitis (UC), Crohns disease (CD), and control human colon. Key Results: Panx2 was widely expressed in myenteric and submucosal ganglia, particularly in the cytoplasm of neurons. We find extensive colocalisation of GFAP and S-100 in the submucosal plexus. This is illustrated below in fig 1 in a whole mount preparation of the submucosal plexus from the rat colon. This confocal image reveals colocalisation of GFAP and S-100 in enteric glia (17 µm z stack of 1 µm optical sections; scale bar 50 µm) (fig 1). S-100 is also found in the cytoplasm of the glial perikarya; there is virtually no nuclear labelling, which was the most obvious element of the staining demonstrated by von Boyen et al.. In fig 1 of the paper of von Boyen et al, the nature of the GFAP immunoreactivity is not fibrous, but granular, while the predominant labelling of S-100 is nuclear.
